    Cisplatin exposure acutely disrupts mitochondrial bioenergetics in the zebrafish lateral-line organ

    Cisplatin is a commonly used chemotherapeutic agent that causes debilitating high-frequency hearing loss. No targeted therapies currently exist to treat cisplatin ototoxicity, partly because the underlying mechanisms of cisplatin-induced hair cell damage are not completely defined. Zebrafish may offer key insights to cisplatin ototoxicity because their lateral-line organ contains hair cells that are remarkably similar to those within the cochlea but are optically accessible, permitting observation of cisplatin injury in live intact hair cells. In this study, we used a combination of genetically encoded biosensors in zebrafish larvae and fluorescent indicators to characterize changes in mitochondrial bioenergetics in response to cisplatin. Following exposure to cisplatin, confocal imaging of live intact neuromasts demonstrated increased mitochondrial activity. Staining with fixable fluorescent dyes that accumulate in active mitochondria similarly showed hyperpolarized mitochondrial membrane potential. Zebrafish expressing a calcium indicator within their hair cells revealed elevated levels of mitochondrial calcium immediately following completion of cisplatin treatment. A fluorescent ROS indicator demonstrated that these changes in mitochondrial function were associated with increased oxidative stress. After a period of recovery, cisplatin-exposed zebrafish demonstrated caspase-3-mediated apoptosis. Altogether, these findings suggest that cisplatin acutely disrupts mitochondrial bioenergetics and may play a key role in initiating cisplatin ototoxicity

    Novel Glycerol-Crosslinked Poly(acrylic acid) Hydrogel for Encapsulation and Release of Benzocaine

    Drug delivery by a controlled-release mechanism offers several advantages over traditional delivery systems such as improved therapeutic activity, reduced number of drug administrations, as well as minimizing the risk of over-dose or under-dose. In this study, attempt has been made to develop a novel controlled release device from glycerol crosslinked poly(acrylic acid) hydrogels. Glycerol crosslinked poly(acrylic acid) was fabricated by polymerizing acrylic acid and crosslinking the polymer with glycerol in the same reaction pot in the presence of benzoyl peroxide and Novozym 435. The fabricated hydrogel swelled more than 100% in neutral, acidic and basic buffer solutions. Swelling was pH and temperature dependent. The hydrogel encapsulated the drug benzocaine; the amount encapsulated was dependent on time, temperature and initial benzocaine concentration. The hydrogel also released the drug at a controlled rate

    Culture and Mobility Determine the Importance of Similarity in Friendship

    We test whether the ability to start friendships—and end them—determines what we look for in friends. We hypothesized that individual differences in beliefs about relationship choice would translate into differences in similarity of relationship partners. To test the constructs across their spectrum, we sampled both American and Korean relationships (N=1,603). Study 1 field-sampled naturally-occurring relationship pairs in the USA and Korea; independent self-construal was positively correlated with relational mobility and similarity within relationship pairs. Using the entire data set, relational mobility was positively correlated with similarity, but the two were negatively correlated within nations. Study 2 identified two components of relational mobility, Entry and Exit. Easy entry to relationships correlated with high importance of similarity; easy exit from relationships correlated with low importance of similarity. An experiment (Study 3) manipulating both ease of entry to and exit from friendship found the belief that people are relatively free to end an unsatisfying friendship reduced the importance of similarity for friendship selection

    Extragalactic Magnetic Field and the Highest Energy Cosmic Rays

    The strength and spectrum of the extragalactic magnetic field are still unknown. Its measurement would help answer the question of whether galactic fields are purely a primordial relic or were dynamically enhanced from a much smaller cosmological seed field. In this letter, we show that the composition, spectrum, and directional distribution of extragalactic ultrahigh energy cosmic rays with energies above \simeq 10^{18}\ev can probe the large scale component of the extragalactic magnetic field below the present observational upper limit of 10910^{-9} Gauss.     Evaluation of cisplatin-induced pathology in the larval zebrafish lateral line

    Cisplatin is an effective anticancer agent, but also causes permanent hearing loss by damaging hair cells-the sensory receptors essential for hearing. There is an urgent clinical need to protect cochlear hair cells in patients undergoing cisplatin chemotherapy. The zebrafish lateral line organ contains hair cells and has been frequently used in studies to screen for otoprotective compounds. However, these studies have employed a wide range of cisplatin dosages and exposure times. We therefore performed a comprehensive evaluation of cisplatin ototoxicity in the zebrafish lateral line with the goal of producing a standardized, clinically relevant protocol for future studies. To define the dose- and time-response patterns of cisplatin-induced hair-cell death, we treated 6-day-old larvae for 2 h in 50 µM-1 mM cisplatin and allowed them to recover. We observed delayed hair cell death, which peaked at 4-8 h post-exposure. Cisplatin also activated a robust inflammatory response, as determined by macrophage recruitment and phagocytosis of hair cells. However, selective depletion of macrophages did not affect hair cell loss. We also examined the effect of cisplatin treatment on fish behavior and found that cisplatin-induced lateral line injury measurably impaired rheotaxis. Finally, we examined the function of remaining hair cells that appeared resistant to cisplatin treatment. We observed significantly reduced uptake of the cationic dye FM1-43 in these cells relative to untreated controls, indicating that surviving hair cells may be functionally impaired. Cumulatively, these results indicate that relatively brief exposures to cisplatin can produce hair cell damage and delayed hair cell death. Our observations provide guidance on standardizing methods for the use of the zebrafish model in studies of cisplatin ototoxicity

    The Effect of Genetic Taste Status on Swallowing: A Literature Review

    Purpose Swallowing and taste share innervation pathways and are crucial to nutritive intake. Individuals vary in their perception of taste due to factors such as genetics; however, it is unclear to what extent genetic taste status influences swallowing physiology and function. The purpose of this review article is to provide background on genetic taste status, review the evidence on the association between genetic taste status and swallowing, and discuss research and clinical implications. Method A comprehensive literature review was conducted using search terms related to swallowing and genetic taste status. Studies were included if they investigated the main effect of genetic taste status on swallowing or the interaction of genetic taste status with other variables. Studies were grouped by participant population (healthy participants or persons with a swallowing disorder), swallowing-related outcome measure, and method of genetic taste status measurement. Results The results were mixed, with five of 10 reviewed studies reporting a statistically significant main or interaction effect on swallowing. Most studies included healthy participants, with only one study investigating participants with dysphagia. Additionally, swallowing-related outcome measures and methods of determining genetic taste status varied greatly between studies conducted on separate cohorts. Conclusion Few studies have incorporated genetic taste status as a variable in swallowing research, and results are mixed. Future research on sensation and swallowing should consider the potential effect of genetic taste status and follow standardized procedures for its determination. Despite the limited evidence, clinicians may consider how individual differences in perception shape swallowing outcomes

    Strain smoothing for compressible and nearly-incompressible finite elasticity

    We present a robust and efficient form of the smoothed finite element method (S-FEM) to simulate hyperelastic bodies with compressible and nearly-incompressible neo-Hookean behaviour. The resulting method is stable, free from volumetric locking and robust on highly distorted meshes. To ensure inf-sup stability of our method we add a cubic bubble function to each element. The weak form for the smoothed hyperelastic problem is derived analogously to that of smoothed linear elastic problem. Smoothed strains and smoothed deformation gradients are evaluated on sub-domains selected by either edge information (edge-based S-FEM, ES-FEM) or nodal information (node-based S-FEM, NS-FEM). Numerical examples are shown that demonstrate the efficiency and reliability of the proposed approach in the nearly-incompressible limit and on highly distorted meshes. We conclude that, strain smoothing is at least as accurate and stable, as the MINI element, for an equivalent problem size

    Measurement of low‐density lipoprotein cholesterol levels in primary and secondary prevention patients: Insights from the PALM registry

    Background The 2013 American College of Cardiology/American Heart Association Guideline on the Treatment of Blood Cholesterol to Reduce Atherosclerotic Cardiovascular Risk in Adults recommended testing low-density lipoprotein cholesterol ( LDL -C) to identify untreated patients with LDL -C ≥190 mg/dL, assess lipid-lowering therapy adherence, and consider nonstatin therapy. We sought to determine whether clinician lipid testing practices were consistent with these guidelines. Methods and Results The PALM (Patient and Provider Assessment of Lipid Management) registry enrolled primary and secondary prevention patients from 140 US cardiology, endocrinology, and primary care offices in 2015 and captured demographic data, lipid treatment history, and the highest LDL -C level in the past 2 years. Core laboratory lipid levels were drawn at enrollment. Among 7627 patients, 2787 (36.5%) had no LDL -C levels measured in the 2 years before enrollment. Patients without chart-documented LDL -C levels were more often women, nonwhite, uninsured, and non-college graduates (all P\u3c0.01). Patients without prior lipid testing were less likely to receive statin treatment (72.6% versus 76.0%; P=0.0034), a high-intensity statin (21.5% versus 24.3%; P=0.016), nonstatin lipid-lowering therapy (24.8% versus 27.3%; P=0.037), and had higher core laboratory LDL -C levels at enrollment (median 97 versus 92 mg/dL; P\u3c0.0001) than patients with prior LDL -C testing. Of 166 individuals with core laboratory LDL -C levels ≥190 mg/dL, 36.1% had no LDL -C measurement in the prior 2 years, and 57.2% were not on a statin at the time of enrollment. Conclusions In routine clinical practice, LDL -C testing is associated with higher-intensity lipid-lowering treatment and lower achieved LDL -C level
