    Investigating visual strategies during the recognition of static and dynamic facial affect in TBI and control cohorts

    bjectives: Traumatic brain injury (TBI) often precipitates socio-emotional problems which impinge on social relationships. Although socio-emotional impairments are debilitating the mechanisms underpinning these are poorly understood. Social interactions are dynamic yet research frequently employs static assessments. We investigated eye scan patterns of TBI and controls viewing static/dynamic facial expressions. Design: Factorial analysis investigated correct response, reaction time, fixation duration/count to areas of interest (eyes, nose and mouth) across six emotions (anger, disgust, fear, happy, sadness and surprise). Methods: 17 TBI participants were recruited from the NHS and age/gender matched controls were recruited using stratified opportunity sampling. Images from the Amsterdam Dynamic Facial Expression Set (ADFES) were presented on a Tobii T120 Eye Tracker screen. Multivariate and correlational methods were used to analyse data. Static Results: Controls displayed greater fixation durations; counts to eyes were more accurate and quicker identifying emotions than TBI participants. The TBI group focused more on the nose compared to controls. Higher scores on the ADFES correlated with quicker responses across all emotions for all participants and positive relationships between empathy, emotion recognition and fixation patterns were revealed. Gaze-patterns for the six emotions differed slightly between the groups although fearful faces induced more/longer fixations and happy faces the least/shortest fixations. Dynamic Results: Controls were more accurate at identifying emotions than the TBI group but no group differences were found for gaze-patterns. Happy faces induced more/longer fixations and sad faces the least/shortest fixations. A positive correlation between correct scores on the ADFES and empathy scores across all emotions for all participants was established. Conclusions: Irregular gaze-patterns could underpin some socio-emotional problems after TBI, highlighting the potential for innovative rehabilitation approaches.     Exploring fixation patterns and social cognition after traumatic brain injury

    Objectives: Social cognition (SC) impairments after traumatic brain injury (TBI) are pervasive. The movie for the assessment of social cognition (MASC) measures different facets of social interactions over the three stages of SC; social perception, social knowledge retrieval and response selection. The mechanisms underpinning SC deficits after TBI are poorly understood but aberrant eye fixation patterns could play a role. The present research explored fixations across social interactions to determine group differences and correlations between eye tracking and behavioural data. Design: Group differences in response selection during the MASC and fixation duration/count to areas of interest (eyes, nose and mouth) were examined. Methods: 18 TBI participants were recruited from the NHS and age/gender matched controls were recruited using stratified opportunity sampling. The MASC allows for quantification of incorrect answers; excessive theory of mind (ToM), reduced ToM and absence of ToM errors. The MASC was presented on a Tobii T120 eye tracker monitor. Results: TBI participants had significantly lower correct scores on the MASC and higher excessive/reduced errors compared to controls. There was no significant interaction between automated optical inspection (AOI) and group. However, significant main effects of group for fixation duration/count indicated that if AOI was ignored, controls displayed longer/more fixations overall suggesting a difference in visual scanning patterns between TBI and control groups. No significant correlations were established. Conclusions: TBI and controls exhibited disparate visual strategies during the MASC and this effect could underpin some SC impairments displayed by TBI participants. TBI participants also displayed insufficient and over-interpretative mental state reasoning compared to controls but it is unclear why. The present research outlines the multifaceted nature of SC impairments after TBI and highlights potential areas for SC intervention post-TB

    A systematic review of digital interventions for improving the diet and physical activity behaviors of adolescents

    Many adolescents have poor diet and physical activity behaviors, which can lead to the development of noncommunicable diseases in later life. Digital platforms offer inexpensive means of delivering health interventions, but little is known about their effectiveness. This systematic review was conducted to synthesize evidence on the effectiveness of digital interventions to improve diet quality and increase physical activity in adolescents, to effective intervention components and to assess the cost-effectiveness of these interventions. Following a systematic search, abstracts were assessed against inclusion criteria, and data extraction and quality assessment were performed for included studies. Data were analyzed to identify key features that are associated with significant improvement in behavior. A total of 27 studies met inclusion criteria. Most (n = 15) were Web site interventions. Other delivery methods were text messages, games, multicomponent interventions, emails, and social media. Significant behavior change was often seen when interventions included education, goal setting, self-monitoring, and parental involvement. None of the publications reported cost-effectiveness. Due to heterogeneity of studies, meta-analysis was not feasible.It is possible to effect significant health behavior change in adolescents through digital interventions that incorporate education, goal setting, self-monitoring, and parental involvement. Most of the evidence relates to Web sites and further research into alternate media is needed, and longer term outcomes should be evaluated. There is a paucity of data on the cost-effectiveness of digital health interventions, and future trials should report these data

    Loss of normal Alzheimer's disease-associated Presenilin 2 function alters antiseizure medicine potency and tolerability in the 6-Hz focal seizure model

    IntroductionPatients with early-onset Alzheimer's disease (EOAD) experience seizures and subclinical epileptiform activity, which may accelerate cognitive and functional decline. Antiseizure medicines (ASMs) may be a tractable disease-modifying strategy; numerous ASMs are marketed with well-established safety. However, little information is available to guide ASM selection as few studies have rigorously quantified ASM potency and tolerability in traditional seizure models in rodents with EOAD-associated risk factors. Presenilin 2 (PSEN2) variants evoke EOAD, and these patients experience seizures. This study thus established the anticonvulsant profile of mechanistically distinct ASMs in the frontline 6-Hz limbic seizure test evoked in PSEN2-knockout (KO) mice to better inform seizure management in EOAD.MethodsThe median effective dose (ED50) of prototype ASMs was quantified in the 6-Hz test in male and female PSEN2-KO and wild-type (WT) C57BL/6J mice (3–4 months old). Minimal motor impairment (MMI) was assessed to estimate a protective index (PI). Immunohistological detection of cFos established the extent to which 6-Hz stimulation activates discrete brain regions in KO vs. WT mice.ResultsThere were significant genotype-related differences in the potency and tolerability of several ASMs. Valproic acid and levetiracetam were significantly more potent in male KO than in WT mice. Additionally, high doses of valproic acid significantly worsened MMI in KO mice. Conversely, carbamazepine was significantly less potent in female KO vs. WT mice. In both male and female KO mice vs. WTs, perampanel and lamotrigine were equally potent. However, there were marked genotype-related shifts in PI of both carbamazepine and perampanel, with KO mice exhibiting less MMI at the highest doses tested. Gabapentin was ineffective against 6-Hz seizures in KO mice vs. WTs without MMI changes. Neuronal activation 90 min following 6-Hz stimulation was significantly increased in the posterior parietal association cortex overlying CA1 and in the piriform cortex of WT mice, while stimulation-induced increases in cFos immunoreactivity were absent in KO mice.DiscussionAcute ASM potency and tolerability in the high-throughput 6-Hz test may be significantly altered with loss of normal PSEN2 function. Seizures in discrete EOAD populations may benefit from precisely selected medicines optimized for primary ASM pharmacological mechanisms

    Dynamic Emotion Recognition and Social Inference Ability in Traumatic Brain Injury: An Eye-Tracking Comparison Study.

    Emotion recognition and social inference impairments are well-documented features of post-traumatic brain injury (TBI), yet the mechanisms underpinning these are not fully understood. We examined dynamic emotion recognition, social inference abilities, and eye fixation patterns between adults with and without TBI. Eighteen individuals with TBI and 18 matched non-TBI participants were recruited and underwent all three components of The Assessment of Social Inference Test (TASIT). The TBI group were less accurate in identifying emotions compared to the non-TBI group. Individuals with TBI also scored lower when distinguishing sincere and sarcastic conversations, but scored similarly to those without TBI during lie vignettes. Finally, those with TBI also had difficulty understanding the actor’s intentions, feelings, and beliefs compared to participants without TBI. No group differences were found for eye fixation patterns, and there were no associations between fixations and behavioural accuracy scores. This conflicts with previous studies, and might be related to an important distinction between static and dynamic stimuli. Visual strategies appeared goal- and stimulus-driven, with attention being distributed to the most diagnostic area of the face for each emotion. These findings suggest that low-level visual deficits may not be modulating emotion recognition and social inference disturbances post-TBI

    Common loss of far-red light photoacclimation in cyanobacteria from hot and cold deserts: a case study in the Chroococcidiopsidales

    Deserts represent an extreme challenge for photosynthetic life. Despite their aridity, they are often inhabited by diverse microscopic communities of cyanobacteria. These organisms are commonly found in lithic habitats, where they are partially sheltered from extremes of temperature and UV radiation. However, living under the rock surface imposes additional constraints, such as limited light availability, and enrichment of longer wavelengths than are typically usable for oxygenic photosynthesis. Some cyanobacteria from the genus Chroococcidiopsis can use this light to photosynthesize, in a process known as far-red light photoacclimation, or FaRLiP. This genus has commonly been reported from both hot and cold deserts. However, not all Chroococcidiopsis strains carry FaRLiP genes, thus motivating our study into the interplay between FaRLiP and extreme lithic environments. The abundance of sequence data and strains provided the necessary material for an in-depth phylogenetic study, involving spectroscopy, microscopy, and determination of pigment composition, as well as gene and genome analyses. Pigment analyses revealed the presence of red-shifted chlorophylls d and f in all FaRLiP strains tested. In addition, eight genus-level taxa were defined within the encompassing Chroococcidiopsidales, clarifying the phylogeny of this long-standing polyphyletic order. FaRLiP is near universally present in a generalist genus identified in a wide variety of environments, Chroococcidiopsis sensu stricto, while it is rare or absent in closely related, extremophile taxa, including those preferentially inhabiting deserts. This likely reflects the evolutionary process of gene loss in specialist lineages

    Kailo: a systemic approach to addressing the social determinants of young people’s mental health and wellbeing at the local level [version 1; peer review: awaiting peer review]

    The mental health and wellbeing of children and young people is deteriorating. It is increasingly recognised that mental health is a systemic issue, with a wide range of contributing and interacting factors. However, the vast majority of attention and resources are focused on the identification and treatment of mental health disorders, with relatively scant attention on the social determinants of mental health and wellbeing and investment in preventative approaches. Furthermore, there is little attention on how the social determinants manifest or may be influenced at the local level, impeding the design of contextually nuanced preventative approaches. This paper describes a major research and design initiative called Kailo that aims to support the design and implementation of local and contextually nuanced preventative strategies to improve children's and young people’s mental health and wellbeing. The Kailo Framework involves structured engagement with a wide range of local partners and stakeholders - including young people, community partners, practitioners and local system leaders - to better understand local systemic influences and support programmes of youth-centred and evidence-informed co-design, prototyping and testing. It is hypothesised that integrating different sources of knowledge, experience, insight and evidence will result in better embedded, more sustainable and more impactful strategies that address the social determinants of young people’s mental health and wellbeing at the local level
