Tracking the invasive hornet Vespa velutina in complex environments by means of a harmonic radar
An innovative scanning harmonic radar has been recently developed for tracking insects in complex landscapes. This movable technology has been tested on an invasive hornet species (Vespa velutina) for detecting the position of their nests in the environment, in the framework of an early detection strategy. The new model of harmonic radar proved to be effective in tracking hornets either in open landscapes, hilly environments and areas characterised by the presence of more obstacles, such as woodlands and urban areas. Hornets were effectively tracked in complex landscapes for a mean tracking length of 96 ± 62 m with maximum values of ~ 300 m. The effectiveness of locating nests was 75% in new invasive outbreaks and 60% in highly density colonised areas. Furthermore, this technology could provide information on several aspects of insect’s ecology and biology. In this case, new insights were obtained about the mean foraging range of V. velutina (395 ± 208 m with a maximum value of 786 m) and flying features (ground speed), which was 6.66 ± 2.31 m s−1 for foraging individuals (hornets that are not carrying prey’s pellet) and 4.06 ± 1.34 m s−1 for homing individuals
Continuous flow processes for photoconjugation of high-molecular chemical entities
The invention relates to a continuous flow process for the photoconjugation of chemical entities selected from the group comprising carbohydrates, including mono-, oligo- and polysaccharides, amino acids, peptides and proteins, unbranched and branched oligomers and polymers, including dendrimers, which process comprises at least the following steps: a) providing a mixture of the reactants dissolved in an aqueous or organic solvent; b) passing said mixture through a continuous flow reactor and concomitantly irradiating the mixture with light in order to initiate and perform the photoreaction. In preferred embodiments, the photoconjugation is a thiol-ene conjugation or a [2+2] cycloaddition. A further aspect of the invention relates to the conjugates obtainable by this continuous flow process, in particular dendronized polymers such as carbohydrate-dendronized polymers
Skeletal muscle overexpression of sAnk1.5 in transgenic mice does not predispose to type 2 diabetes
Genome-wide association studies (GWAS) and cis-expression quantitative trait locus (cis-eQTL) analyses indicated an association of the rs508419 single nucleotide polymorphism (SNP) with type 2 diabetes (T2D). rs508419 is localized in the muscle-specific internal promoter (P2) of the ANK1 gene, which drives the expression of the sAnk1.5 isoform. Functional studies showed that the rs508419 C/C variant results in increased transcriptional activity of the P2 promoter, leading to higher levels of sAnk1.5 mRNA and protein in skeletal muscle biopsies of individuals carrying the C/C genotype. To investigate whether sAnk1.5 overexpression in skeletal muscle might predispose to T2D development, we generated transgenic mice (TgsAnk1.5/+) in which the sAnk1.5 coding sequence was selectively overexpressed in skeletal muscle tissue. TgsAnk1.5/+ mice expressed up to 50% as much sAnk1.5 protein as wild-type (WT) muscles, mirroring the difference reported between individuals with the C/C or T/T genotype at rs508419. However, fasting glucose levels, glucose tolerance, insulin levels and insulin response in TgsAnk1.5/+ mice did not differ from those of age-matched WT mice monitored over a 12-month period. Even when fed a high-fat diet, TgsAnk1.5/+ mice only presented increased caloric intake, but glucose disposal, insulin tolerance and weight gain were comparable to those of WT mice fed a similar diet. Altogether, these data indicate that sAnk1.5 overexpression in skeletal muscle does not predispose mice to T2D susceptibility

Astronomy has entered the big data era and Machine Learning based methods
have found widespread use in a large variety of astronomical applications. This
is demonstrated by the recent huge increase in the number of publications
making use of this new approach. The usage of machine learning methods, however
is still far from trivial and many problems still need to be solved. Using the
evaluation of photometric redshifts as a case study, we outline the main

Sak1 kinase interacts with Pso2 nuclease in response to DNA damage induced by interstrand crosslink-inducing agents in Saccharomyces cerevisiae.
By isolating putative binding partners through the two-hybrid system (THS) we further extended the characterization of the specific interstrand cross-link (ICL) repair gene PSO2 of Saccharomyces cerevisiae. Nine fusion protein products were isolated for Pso2p using THS, among them the Sak1 kinase, which interacted with the C-terminal b-CASP domain of Pso2p. Comparison of mutagen-sensitivity phenotypes of pso2D, sak1D and pso2Dsak1D disruptants revealed that SAK1 is necessary for complete WT-like repair. The epistatic interaction of both mutant alleles suggests that Sak1p and Pso2p act in the same pathway of controlling sensitivity to DNA-damaging agents. We also observed that Pso2p is phosphorylated by Sak1 kinase in vitro and co-immunoprecipitates with Sak1p after 8-MOP+UVA treatment. Survival data after treatment of pso2D, yku70D and yku70Dpso2D with nitrogen mustard, PSO2 and SAK1 with YKU70 or DNL4 single-, double- and triple mutants with 8-MOP+UVA indicated that ICL repair is independent of YKu70p and DNL4p in S. cerevisiae. Furthermore, a non-epistatic interaction was observed between MRE11, PSO2 and SAK1 genes after ICL induction, indicating that their encoded proteins act on the same substrate, but in distinct repair pathways. In contrast, an epistatic interaction was observed for PSO2 and RAD52, PSO2 and RAD50, PSO2 and XRS2 genes in 8-MOP+UVA treated exponentially growing cells.DOI 10.1016/j.jphotobiol.2013.11.02
Multiple Routes of Pesticide Exposure for Honey Bees Living Near Agricultural Fields
Populations of honey bees and other pollinators have declined worldwide in recent years. A variety of stressors have been implicated as potential causes, including agricultural pesticides. Neonicotinoid insecticides, which are widely used and highly toxic to honey bees, have been found in previous analyses of honey bee pollen and comb material. However, the routes of exposure have remained largely undefined. We used LC/MS-MS to analyze samples of honey bees, pollen stored in the hive and several potential exposure routes associated with plantings of neonicotinoid treated maize. Our results demonstrate that bees are exposed to these compounds and several other agricultural pesticides in several ways throughout the foraging period. During spring, extremely high levels of clothianidin and thiamethoxam were found in planter exhaust material produced during the planting of treated maize seed. We also found neonicotinoids in the soil of each field we sampled, including unplanted fields. Plants visited by foraging bees (dandelions) growing near these fields were found to contain neonicotinoids as well. This indicates deposition of neonicotinoids on the flowers, uptake by the root system, or both. Dead bees collected near hive entrances during the spring sampling period were found to contain clothianidin as well, although whether exposure was oral (consuming pollen) or by contact (soil/planter dust) is unclear. We also detected the insecticide clothianidin in pollen collected by bees and stored in the hive. When maize plants in our field reached anthesis, maize pollen from treated seed was found to contain clothianidin and other pesticides; and honey bees in our study readily collected maize pollen. These findings clarify some of the mechanisms by which honey bees may be exposed to agricultural pesticides throughout the growing season. These results have implications for a wide range of large-scale annual cropping systems that utilize neonicotinoid seed treatments
Current use of androgens in bone marrow failure disorders: a report from the Severe Aplastic Anemia Working Party of the European Society of Blood and Marrow Transplantation
Androgens have represented the historical therapeutic backbone of bone marrow failure (BMF) syndromes. However, their role has been rarely analyzed in prospective setting and systematic and long-term data are currently unavailable regarding their usage, effectiveness and toxicity in both acquired and inherited BMF.
Here, taking advantage of a unique disease-specific international dataset, we retrospectively analyzed the so far largest cohort of BMF patients who received androgens before or in absence of an allogeneic hematopoietic cell transplantation (HCT), reappraising their current use in these disorders. We identified 274 patients across 82 EBMT affiliated centers, 193 with acquired (median age of 32) and 81 with inherited BMF (median age of 8 years). With a median duration of androgen treatment of 5.6 and 20 months respectively, complete/partial remission rates at 3 months were of 6%/29% in acquired and 8%/29% in inherited disorders. Five-year overall survival and failure free survival (FFS) were respectively 63% and 23% in acquired and 78% and 14% in inherited contexts. Androgen initiation after second line treatments for acquired, and after > 12 months post-diagnosis for inherited group were identified as factors associated with improved FFS in multivariable analysis. Androgen use was associated with a manageable incidence of organ-specific toxicity and low rates of solid and hematological malignancies. Sub-analysis of transplant-related outcomes after exposure to these compounds showed probabilities of survival and complications similar to other transplanted BMF cohorts.
This study delivers a unique opportunity to track androgen use in BMF syndromes and represents the basis for general recommendations on their use on behalf of the SAAWP of the EBMT
