    Effects of zinc oxide filler on the curing and mechanical response of alkyd coatings

    The mechanical properties of an alkyd resin filled with zinc oxide pigment were studied at different concentrations over a wide range of time scales using dynamic mechanical analysis, quartz crystal rheometry and nanoindentation. The motivation for this work stems from the interest in accessing the long-term properties of paint coatings by studying the mechanical properties of historic paints. In this foundational work, we compare three different modalities of mechanical measurements and systematically determine the effect of pigment filler loading on the measured properties. Quantitative agreement between the methods is obtained when the characteristic time scales of each of the methods is taken into account. While nanoindentation is the technique most readily applied to historic paint samples, the rheometric quartz crystal microbalance (rheo-QCM) is the best suited for obtaining mechanistic information from measurements of paint properties over time, provided that appropriate thin-film samples can be produced. In these studies we find that ZnO increases the rate of oxidation of the alkyd during the initial stages of cure by an amount that depends on the ZnO content

    Personality structure in bottlenose dolphins (Tursiops truncatus).

    Comparative studies can help identify selective pressures that contributed to species differences in the number and composition of personality domains. Despite being adapted to an aquatic lifestyle and last sharing a common ancestor with primates some 95 million years ago, bottlenose dolphins (Tursiops truncatus) resemble nonhuman primate species in several behavioral and cognitive traits. For example, like chimpanzees (Pan troglodytes), dolphins live in fission-fusion societies, use tools, and have relatively large brains. To determine the extent to which these and other factors contribute to the evolution of personality structure, we examined personality structure in 134 bottlenose dolphins. Personality was measured in 49 dolphins using a 42-item questionnaire, and in 85 dolphins using a version of the questionnaire that included 7 additional items. We found four domains. Three—openness, sociability, and disagreeableness—resembled personality domains found in nonhuman primates and other species. The fourth, directedness, was a blend of high conscientiousness and low neuroticism, and was unique to dolphins. Unlike other species, dolphins did not appear to have a strong dominance domain. The overlap in personality structure between dolphins and other species suggests that selective pressures, such as those related to group structure, terrestrial lifestyles, morphology, and social learning or tool use are not necessary for particular domains to evolve within a species

    IDO2 in Immunomodulation and Autoimmune Disease.

    IDO2 is a relative of IDO1 implicated in tryptophan catabolism and immune modulation but its specific contributions to normal physiology and pathophysiology are not known. Evolutionary genetic studies suggest that IDO2 has a unique function ancestral to IDO1. In mice, IDO2 gene deletion does not appreciably affect embryonic development or hematopoiesis, but it leads to defects in allergic or autoimmune responses and in the ability of IDO1 to influence the generation of T regulatory cells. Gene expression studies indicate that IDO2 is a basally and more narrowly expressed gene than IDO1 and that IDO2 is uniquely regulated by AhR, which serves as a physiological receptor for the tryptophan catabolite kynurenine. In the established KRN transgenic mouse model of rheumatoid arthritis, where IDO1 gene deletion has no effect, IDO2 deletion selectively blunts responses to autoantigen but has no effect on responses to neoantigen challenge. In human populations, natural variations in IDO2 gene sequence that attenuate enzymatic activity have been reported to influence brain cancer control and adaptive immune responses to the IDO2 protein itself, consistent with the concept that IDO2 is involved in shaping immune tolerance in human beings. Biochemical and pharmacological studies provide further evidence of differences in IDO2 enzymology and function relative to IDO1. We suggest that IDO2 may act in a distinct manner from IDO1 as a set-point for tolerance to altered-self antigens along the self-non-self continuum where immune challenges from cancer and autoimmunity may arise

    Mesenchymal Cells Affect Salivary Epithelial Cell Morphology on PGS/PLGA Core/Shell Nanofibers

    Engineering salivary glands is of interest due to the damaging effects of radiation therapy and the autoimmune disease Sjögren’s syndrome on salivary gland function. One of the current problems in tissue engineering is that in vitro studies often fail to predict in vivo regeneration due to failure of cells to interact with scaffolds and of the single cell types that are typically used for these studies. Although poly (lactic co glycolic acid) (PLGA) nanofiber scaffolds have been used for in vitro growth of epithelial cells, PLGA has low compliance and cells do not penetrate the scaffolds. Using a core-shell electrospinning technique, we incorporated poly (glycerol sebacate) (PGS) into PLGA scaffolds to increase the compliance and decrease hydrophobicity. PGS/PLGA scaffolds promoted epithelial cell penetration into the scaffold and apical localization of tight junction proteins, which is necessary for epithelial cell function. Additionally, co-culture of the salivary epithelial cells with NIH3T3 mesenchymal cells on PGS/PLGA scaffolds facilitated epithelial tissue reorganization and apical localization of tight junction proteins significantly more than in the absence of the mesenchyme. These data demonstrate the applicability of PGS/PLGA nanofibers for epithelial cell self-organization and facilitation of co-culture cell interactions that promote tissue self-organization in vitro

    Climate-modulated range expansion of reef-building coral communities off southeast Florida during the late Holocene

    The Holocene reefs off southeast Florida provide unique insights into the biogeographical and ecological response of western Atlantic coral reefs to past climate change that can be used to evaluate future climate impacts. However, previous studies have focused on millennial-scale change during the stable mid-Holocene, making it difficult to make inferences about the impact of shorter-term variability that is relevant to modern climate warming. Using uranium-series dating of newly discovered subfossil coral rubble deposits, we establish a new high-resolution record of coral community development off southeast Florida during a period of variable climate in the late Holocene. Our results indicate that coral communities dominated by reef-building Acropora palmata and Orbicella spp. persisted in the nearshore environments off southeast Florida ~75 km north of their primary historical ranges between ~3500 and 1800 years before present. This timing coincides with regional warming at the northern extent of the Atlantic Warm Pool, suggesting a likely link between regional oceanographic climate and the expansion of cold-sensitive reef-building coral communities to the high-latitude reefs off southeast Florida. These findings not only extend the record of coral-reef development in southeast Florida into the late Holocene, but they also have important implications for future range expansions of reef-building coral communities in response to modern climate change

    LifeGene : a large prospective population-based study of global relevance

    Studying gene-environment interactions requires that the amount and quality of the lifestyle data is comparable to what is available for the corresponding genomic data. Sweden has several crucial prerequisites for comprehensive longitudinal biomedical research, such as the personal identity number, the universally available national health care system, continuously updated population and health registries and a scientifically motivated population. LifeGene builds on these strengths to bridge the gap between basic research and clinical applications with particular attention to populations, through a unique design in a research-friendly setting. LifeGene is designed both as a prospective cohort study and an infrastructure with repeated contacts of study participants approximately every 5 years. Index persons aged 18-45 years old will be recruited and invited to include their household members (partner and any children). A comprehensive questionnaire addressing cutting-edge research questions will be administered through the web with short follow-ups annually. Biosamples and physical measurements will also be collected at baseline, and re-administered every 5 years thereafter. Event-based sampling will be a key feature of LifeGene. The household-based design will give the opportunity to involve young couples prior to and during pregnancy, allowing for the first study of children born into cohort with complete pre-and perinatal data from both the mother and father. Questions and sampling schemes will be tailored to the participants' age and life events.     Melanoma staging: Evidence‐based changes in the American Joint Committee on Cancer eighth edition cancer staging manual

    Answer questions and earn CME/CNETo update the melanoma staging system of the American Joint Committee on Cancer (AJCC) a large database was assembled comprising >46,000 patients from 10 centers worldwide with stages I, II, and III melanoma diagnosed since 1998. Based on analyses of this new database, the existing seventh edition AJCC stage IV database, and contemporary clinical trial data, the AJCC Melanoma Expert Panel introduced several important changes to the Tumor, Nodes, Metastasis (TNM) classification and stage grouping criteria. Key changes in the eighth edition AJCC Cancer Staging Manual include: 1) tumor thickness measurements to be recorded to the nearest 0.1 mm, not 0.01 mm; 2) definitions of T1a and T1b are revised (T1a, <0.8 mm without ulceration; T1b, 0.8‐1.0 mm with or without ulceration or <0.8 mm with ulceration), with mitotic rate no longer a T category criterion; 3) pathological (but not clinical) stage IA is revised to include T1b N0 M0 (formerly pathologic stage IB); 4) the N category descriptors “microscopic” and “macroscopic” for regional node metastasis are redefined as “clinically occult” and “clinically apparent”; 5) prognostic stage III groupings are based on N category criteria and T category criteria (ie, primary tumor thickness and ulceration) and increased from 3 to 4 subgroups (stages IIIA‐IIID); 6) definitions of N subcategories are revised, with the presence of microsatellites, satellites, or in‐transit metastases now categorized as N1c, N2c, or N3c based on the number of tumor‐involved regional lymph nodes, if any; 7) descriptors are added to each M1 subcategory designation for lactate dehydrogenase (LDH) level (LDH elevation no longer upstages to M1c); and 8) a new M1d designation is added for central nervous system metastases. This evidence‐based revision of the AJCC melanoma staging system will guide patient treatment, provide better prognostic estimates, and refine stratification of patients entering clinical trials.
