    Meaningful connection in virtual classrooms: graduate students’ perspectives on effective instructor presence in blended courses

    This qualitative study explored 15 graduate students’ perspectives on effective online instructor presence. Analysis of interviews, a survey, and a focus group revealed students value early relationship-building through consistent participation, authentic personality-sharing, and learner-centered course design. Results indicate effective instructor presence fosters trust, satisfaction, engagement, and positive student mindsets while reducing stress and anxiety. Students preferred visible, accessible instructors who connect through prompt communication, constructive feedback, and active listening. Additional findings suggest leveraging synchronous interactions enhances social presence and relationship-building. However, disconnected instructor presence caused frustration and negative emotions. Overall, intentional instructor presence is critical for successful online instruction and profoundly shapes learners’ holistic experiences beyond solely academic goals. While limited to one program, these learner-centered insights provide a starting point for identifying high-impact presence-building strategies tailored to graduate contexts

    Processing bodies oscillate in neuro 2A cells

    Circadian rhythms are biological variables that oscillate with periods close to 24 h that are generated internally by biological clocks. Depending on the tissue/cell type, about 5–20% of genes are expressed rhythmically. Unexpectedly, the correlation between the oscillations of messengers and the proteins they encode is low. We hypothesize that these discrepancies could be because in certain phases of the circadian cycle some messengers could be translationally silenced and stored. Processing bodies (PBs) are membraneless organelles formed by ribonucleoprotein aggregates located in the cytoplasm. They contain silenced messengers and factors involved in mRNA processing. A previous work showed that the number of cells containing these mRNA granules varies when comparing two time-points in U2OS cell cultures and that these differences disappear when an essential clock gene is silenced. Here we evaluate whether PBs oscillate in Neuro2A cells. We analyzed in cell cultures synchronized with dexamethasone the variations in the number, the signal intensity of the markers used (GE-1/HEDLS and DDX6), and the area of PBs between 8 and 68 h post-synchronization. All three parameters oscillated with periods compatible with a circadian regulated process. The most robust rhythm was the number of PBs. These rhythms could be generated by oscillations in proteins that have been involved in the nucleation of these foci such as LSM1, TTP, and BRF1. The described phenomenon would allow to explain the differences observed in the temporal profiles of some messengers and their proteins and to understand how circadian clocks can control post-transcriptionally cellular functions.Fil: Malcolm, Melisa.     Automatic Construction of Evaluation Suites for Natural Language Generation Datasets

    Machine learning approaches applied to NLP are often evaluated by summarizing their performance in a single number, for example accuracy. Since most test sets are constructed as an i.i.d. sample from the overall data, this approach overly simplifies the complexity of language and encourages overfitting to the head of the data distribution. As such, rare language phenomena or text about underrepresented groups are not equally included in the evaluation. To encourage more in-depth model analyses, researchers have proposed the use of multiple test sets, also called challenge sets, that assess specific capabilities of a model. In this paper, we develop a framework based on this idea which is able to generate controlled perturbations and identify subsets in text-to-scalar, text-to-text, or data-to-text settings. By applying this framework to the GEM generation benchmark, we propose an evaluation suite made of 80 challenge sets, demonstrate the kinds of analyses that it enables and shed light onto the limits of current generation models

    Using heart rate profiles during sleep as a biomarker of depression

    Background: Abnormalities in heart rate during sleep linked to impaired neuro-cardiac modulation may provide new information about physiological sleep signatures of depression. This study assessed the validity of an algorithm using patterns of heart rate changes during sleep to discriminate between individuals with depression and healthy controls. Methods: A heart rate profiling algorithm was modeled using machine-learning based on 1203 polysomnograms from individuals with depression referred to a sleep clinic for the assessment of sleep abnormalities, including insomnia, excessive daytime fatigue, and sleep-related breathing disturbances (n = 664) and mentally healthy controls (n = 529). The final algorithm was tested on a distinct sample (n = 174) to categorize each individual as depressed or not depressed. The resulting categorizations were compared to medical record diagnoses. Results: The algorithm had an overall classification accuracy of 79.9% [sensitivity: 82.8, 95% CI (0.73–0.89), specificity: 77.0, 95% CI (0.67–0.85)]. The algorithm remained highly sensitive across subgroups stratified by age, sex, depression severity, comorbid psychiatric illness, cardiovascular disease, and smoking status. Conclusions: Sleep-derived heart rate patterns could act as an objective biomarker of depression, at least when it cooccurs with sleep disturbances, and may serve as a complimentary objective diagnostic tool.     Pro-Inflammatory and Immunological Profile of Dogs with Myxomatous Mitral Valve Disease

    Simple Summary Myxomatous mitral valve disease (MMVD) is the most commonly acquired cardiac disease in dogs and is responsible for congestive heart failure. In this research, some inflammatory, immunological, and echocardiographic parameters were evaluated in dogs affected by MMVD in order to assess the involvement of additional pathophysiological mechanisms during the disease. The main results revealed that inflammation parameters increased according to the severity of the disease and suggested that inflammatory activation may play an important role in cardiac remodeling associated with the progressive volumetric overload in MMVD. Also, a relative increase in Treg cells was detected, suggesting that they could represent a regulatory mechanism for limiting the inflammatory immune response. Myxomatous mitral valve disease (MMVD) is a very frequently acquired cardiac disease in dog breeds and is responsible for congestive heart failure (CHF). The involvement of the immune system and pro-inflammatory cytokines in dogs with CHF due to mitral valve disease has not yet been extensively investigated. Here, we investigate the role of pro-inflammatory cytokines and the dysfunction of the immune system in dogs with different stages of severity through the blood assessment of CD4(+)FoxP3(+)regulatory T cells (Treg) cells, leptin, tumor necrosis factor (TNF)-alpha, interleukin (IL)-1 beta and IL-6 pro-inflammatory cytokines, and immunological and echocardiographic parameters. A total of 36 cardiopathic dogs, 14 females and 22 males, with MMVD were included. Mean age and body weight (BW) at the time of enrollment were 10.7 +/- 2.77 years and 10.9 +/- 6.69 kg, respectively. For the comparison of the pro-inflammatory and immunological parameters, two groups of healthy dogs were also established. Control group 1 consisted of young animals (n. 11; 6 females and 5 males), whose age and mean weight were 4.1 +/- 0.82 years and 13.8 +/- 4.30 kg, respectively. Control group 2 consisted of elderly dogs (n. 12; 6 females and 6 males), whose age and BW were 9.6 +/- 0.98 years and 14.8 +/- 6.15 kg, respectively. Of particular interest, an increase in Treg cells was observed in the cohort of MMVD dogs, as compared to the healthy dogs, as Treg cells are involved in the maintenance of peripheral tolerance, and they are involved in etiopathogenetic and pathophysiological mechanisms in the dog. On the other hand, TNF-alpha, IL-1 beta, and IL-6 significantly increased according to the severity of the disease in MMVD dogs. Furthermore, the positive correlation between IL-6 and the left ventricle diastolic volume suggests that inflammatory activation may be involved in cardiac remodeling associated with the progressive volumetric overload in MMVD

    UNEP-SETAC guideline on global land use impact assessment on biodiversity and ecosystem services in LCA

    Purpose As a consequence of the multi-functionality of land, the impact assessment of land use in Life Cycle Impact Assessment requires the modelling of several impact pathways covering biodiversity and ecosystem services. To provide consistency amongst these separate impact pathways, general principles for their modelling are provided in this paper. These are refinements to the principles that have already been proposed in publications by the UNEP-SETAC Life Cycle Initiative. In particular, this paper addresses the calculation of land use interventions and land use impacts, the issue of impact reversibility, the spatial and temporal distribution of such impacts and the assessment of absolute or relative ecosystem quality changes. Based on this, we propose a guideline to build methods for land use impact assessment in Life Cycle Assessment (LCA). Results Recommendations are given for the development of new characterization models and for which a series of key elements should explicitly be stated, such as the modelled land use impact pathways, the land use/cover typology covered, the level of biogeographical differentiation used for the characterization factors, the reference land use situation used and if relative or absolute quality changes are used to calculate land use impacts. Moreover, for an application of the characterisation factors (CFs) in an LCA study, data collection should be transparent with respect to the data input required from the land use inventory and the regeneration times. Indications on how generic CFs can be used for the background system as well as how spatial-based CFs can be calculated for the foreground system in a specific LCA study and how land use change is to be allocated should be detailed. Finally, it becomes necessary to justify the modelling period for which land use impacts of land transformation and occupation are calculated and how uncertainty is accounted for. Discussion The presented guideline is based on a number of assumptions: Discrete land use types are sufficient for an assessment of land use impacts; ecosystem quality remains constant over time of occupation; time and area of occupation are substitutable; transformation time is Negligible; regeneration is linear and independent from land use history and landscape configuration; biodiversity and multiple ecosystem services are independent; the ecological impact is linearly increasing with the intervention; and there is no interaction between land use and other drivers such as climate change. These assumptions might influence the results of land use Life Cycle Impact Assessment and need to be critically reflected. Conclusions and recommendations In this and the other papers of the special issue, we presented the principles and recommendations for the calculation of land use impacts on biodiversity and ecosystem services on a global scale. In the framework of LCA, they are mainly used for the Assessment of land use impacts in the background system. The main areas for further development are the link to regional ecological models running in the foreground system, relative weighting of the ecosystem services midpoints and indirect land use.Fil: Koellner, Thomas .     Sequential Surgical Procedures in Vascular Surgery Patients Are Associated With Perioperative Adverse Cardiac Events

    Patients at elevated cardiovascular risk are prone to perioperative cardiovascular complications, like myocardial injury after non-cardiac surgery (MINS). We have demonstrated in a mouse model of atherosclerosis that perioperative stress leads to an increase in plaque volume and higher plaque vulnerability. Regulatory T cells (Tregs) play a pivotal role in development and destabilization of atherosclerotic plaques. For this exploratory post-hoc analysis we identified 40 patients recruited into a prospective perioperative biomarker study, who within the inclusion period underwent sequential open vascular surgery. On the basis of protein markers measured in the biomarker study, we evaluated the perioperative inflammatory response in patients' plasma before and after index surgery as well as before and after a second surgical procedure. We also analyzed available immunohistochemistry samples to describe plaque vulnerability in patients who underwent bilateral carotid endarterectomy (CEA) in two subsequent surgical procedures. Finally, we assessed if MINS was associated with sequential surgery. The inflammatory response of both surgeries was characterized by postoperative increases of interleukin-6,−10, Pentraxin 3 and C-reactive protein with no clear-cut difference between the two time points of surgery. Plaques from CEA extracted during the second surgery contained less Tregs, as measured by Foxp3 staining, than plaques from the first intervention. The 2nd surgical procedure was associated with MINS. In conclusion, we provide descriptive evidence that sequential surgical procedures involve repeat inflammation, and we hypothesize that elevated rates of cardiovascular complications after the second procedure could be related to reduced levels of intraplaque Tregs, a finding that deserves confirmatory testing and mechanistic exploration in future populations
