    Experimental Study on the Flexural Behavior of Reinforced Polystyrene Blocks in Concrete Beams

    A new type of lightweight beam system was recently proposed by embedding polystyrene in beams to improve structural efficiency. This removes the non-performing concrete in the neutral axis and tension region to provide a comparable strength as a solid beam. There are, however, limited studies conducted to investigate the structural behavior of such beams. Therefore, this research presents an experimental investigation to assess the effect of polystyrene shapes in the beams. This involved testing a solid beam and five lightweight beams under flexural load using a four-point load test. The inclusion of polystyrene was estimated to have reduced the self-weight of beams by 8.6% to 11.8% when compared with the solid beam. The results also showed the ellipse polystyrene with a width of 70 mm and height of 50 mm produced the highest effective strength to weight ratio (sw) of 1.12 and performed 12% better than the solid beam. Moreover, the lightweight beams have more weight reduced than the strength, and those with ellipse polystyrene were found to have performed better than circular ones based on first crack load, ultimate load, and effective strength to weight ratio (sw). The beams with ellipse polystyrene allowed better stress distribution and this gave them a higher strength than sphere shape. For industry application, the polystyrene content is recommended to be greater than 10% while the effective strength to weight ratio (sw) of the beam is greater than 1. The successful reduction of the weight without affecting the structural performance has the ability to help in reducing construction costs

Object-based learning approach has been widely adopted in e-learning.The principle concern in the conventional e-learning models is the inflexibility of the e-learning content to assist the use and management of the learning sources which are highly distributed.Rigidity of e-learning content limits its reusability and shareability.Possessing the ability to deliver strong computing power and secure data storage as services, Cloud computing is a promising computing model to promote innovative changes and add notable values to e-learning landscape.The flexibility and on-demand access to a centralized shared pool of computing resources provided by Cloud computing enables high re-usability and share-ability, hence overcomes the principle concern in e-learning due to the rigidity of e-learning content.This paper describes the design of an object-based e-learning framework embracing Cloud computing.The proposed object-based e learning framework can be used to form a new education domain that shares the Cloud characteristics of elasticity, flexibility, efficiency and reliability.Principal to the framework design is the development of Cloud-based e-learning objects where learners have the flexibility to access, personalize and deploy them in e-learning environment.Cloud-based e-learning objects are highly adaptable, reusable and easy changeable, hence allowing them to be used dynamically with greater customizability and flexibility in e-learning.Adapting Cloud computing into object-based e-learning is believed to be able to create a breakthrough in future dynamic -e-learning development

    Toxoplasma gondii infects all warm-blooded animals, including humans, causing serious public health problems and great economic loss for the food industry. Commonly used serological tests require costly and hazardous preparation of whole Toxoplasma lysate antigens from tachyzoites. Here, we have evaluated an alternative method for antigen production, which involved a prokaryotic expression system. Specifically, we expressed T. gondii dense granular protein-5 (GRA5) in Escherichia coli and isolated it by affinity purification. The serodiagnostic potential of the purified recombinant GRA5 (rGRA5) was tested through Western blot analysis against 212 human patient serum samples. We found that rGRA5 protein was 100% specific for analysis of toxoplasmosis-negative human sera. Also, rGRA5 was able to detect acute and chronic T. gondii infections (sensitivities of 46.8% and 61.2%, resp.)

    Genetic assemblage of Sarcocystis spp. in Malaysian snakes

    Abstract Background: Sarcocystis species are protozoan parasites with a wide host range including snakes. Although there were several reports of Sarcocytis species in snakes, their distribution and prevalence are still not fully explored. Methods: In this study, fecal specimens of several snake species in Malaysia were examined for the presence of Sarcocystis by PCR of 18S rDNA sequence. Microscopy examination of the fecal specimens for sporocysts was not carried as it was difficult to determine the species of the infecting Sarcocystis. Results: Of the 28 snake fecal specimens, 7 were positive by PCR. BLASTn and phylogenetic analyses of the amplified 18S rDNA sequences revealed the snakes were infected with either S. nesbitti, S. singaporensis, S. zuoi or undefined Sarcocystis species. Conclusion: This study is the first to report Sarcocystis infection in a cobra, and S. nesbitti in a reticulated python

    Population genetic analysis of Plasmodium knowlesi reveals differential selection and exchange events between Borneo and Peninsular sub-populations

MR/M01360X/1, MR/N010469/1, MR/R025576/1, MR/R020973/1, and MR/X005895/1).The zoonotic Plasmodium knowlesi parasite is a growing public health concern in Southeast Asia, especially in Malaysia, where elimination of P. falciparum and P. vivax malaria has been the focus of control efforts. Understanding of the genetic diversity of P. knowlesi parasites can provide insights into its evolution, population structure, diagnostics, transmission dynamics, and the emergence of drug resistance. Previous work has revealed that P. knowlesi fall into three main sub-populations distinguished by a combination of geographical location and macaque host (Macaca fascicularis and M. nemestrina). It has been shown that Malaysian Borneo groups display profound heterogeneity with long regions of high or low divergence resulting in mosaic patterns between sub-populations, with some evidence of chromosomal-segment exchanges. However, the genetic structure of non-Borneo sub-populations is less clear. By gathering one of the largest collections of P. knowlesi whole-genome sequencing data, we studied structural genomic changes across sub-populations, with the analysis revealing differences in Borneo clusters linked to mosquito-related stages of the parasite cycle, in contrast to differences in host-related stages for the Peninsular group. Our work identifies new genetic exchange events, including introgressions between Malaysian Peninsular and M. nemestrina-associated clusters on various chromosomes, including in parasite invasion genes (DBPβ, NBPXα and NBPXβ), and important proteins expressed in the vertebrate parasite stages. Recombination events appear to have occurred between the Peninsular and M. fascicularis-associated groups, including in the DBPβ and DBPγ invasion associated genes. Overall, our work finds that genetic exchange events have occurred among the recognised contemporary groups of P. knowlesi parasites during their evolutionary history, leading to apparent mosaicism between these sub-populations. These findings generate new hypotheses relevant to parasite evolutionary biology and P. knowlesi epidemiology, which can inform malaria control approaches to containing the impact of zoonotic malaria on human communities.Publisher PDFPeer reviewe

    Metabolite profiling of endophytic Streptomyces spp. and its antiplasmodial potential

    Background: Antiplasmodial drug discovery is significant especially from natural sources such as plant bacteria. This research aimed to determine antiplasmodial metabolites of Streptomyces spp. against Plasmodium falciparum 3D7 by using a metabolomics approach. Methods: Streptomyces strains’ growth curves, namely SUK 12 and SUK 48, were measured and P. falciparum 3D7 IC50 values were calculated. Metabolomics analysis was conducted on both strains’ mid-exponential and stationary phase extracts. Results: The most successful antiplasmodial activity of SUK 12 and SUK 48 extracts shown to be at the stationary phase with IC50 values of 0.8168 ng/mL and 0.1963 ng/mL, respectively. In contrast, the IC50 value of chloroquine diphosphate (CQ) for antiplasmodial activity was 0.2812 ng/mL. The univariate analysis revealed that 854 metabolites and 14, 44 and three metabolites showed significant differences in terms of strain, fermentation phase, and their interactions. Orthogonal partial least square-discriminant analysis and S-loading plot putatively identified pavettine, aurantioclavine, and 4-butyldiphenylmethane as significant outliers from the stationary phase of SUK 48. For potential isolation, metabolomics approach may be used as a preliminary approach to rapidly track and identify the presence of antimalarial metabolites before any isolation and purification can be done

    Sintesis, aktiviti antimalaria dan pendekatan dinamik molekul kompleks logam salofen

    Malaria masih merupakan salah satu penyakit berjangkit yang utama di kawasan beriklim tropika. Pelbagai ubat antimalaria telah diperkenalkan sebelum ini seperti klorokuina, primakuina dan artemisinin. Namun, keberkesanan ubat antimalaria menjadi rumit disebabkan oleh penyebaran parasit Plasmodium yang ada kerintangan terhadap ubat sedia ada. Oleh demikian, penerokaan kompleks logam salofen berasaskan ligan bes Schiff yang berpotensi sebagai agen antimalaria dijalankan. Empat ligan berasaskan salofen a-d dan lapan kompleks logam salofen 1a-4a dan 1b-4b telah disintesis melalui tindak balas kondensasi bes Schiff. Aktiviti antimalaria sebatian ini telah diuji dengan asai Plasmodium laktat dehidrogenase terhadap Plasmodium falciparum 3D7 yang menunjukkan bahawa 1a, 3a dan 4b masing-masing (IC50 = 14.74, 19.38 dan 12.13 µM) mempunyai kesan perencatan aktif terhadap parasit secara in vitro. Analisis dok molekul mendedahkan bahawa sebatian 1a, 3a dan 4b menunjukkan nilai pengikatan yang baik pada protein PfDHFR-TS iaitu nilai masingmasing adalah -9.7, -8.9 dan -9.5 kcal mol. Tambahan pula, simulasi dinamik molekul mendedahkan bahawa sebatian 1a dan 3a ditempatkan di dalam poket reseptor protein dengan sebatian dikelilingi oleh beberapa permukaan berpolar. Bagaimanapun, sebatian 4b membentuk sistem kompleks reseptor-ligan dengan konformasi yang paling stabil apabila kompleks diletakkan di dalam poket reseptor hidrofobik. Oleh itu, PfDHFR-TS dianggap sebagai sasaran protein parasit yang khusus untuk kompleks logam salofen. Kesimpulannya, kompleks logam salofen didapati mempunyai potensi tinggi untuk dibangunkan sebagai agen antimalaria dan mampu menjadi struktur templat kepada pembentukan ubat antimalaria bagi melawan kes kerintangan
