    Meta-analysis of studies on biochemical marker tests for the diagnosis of premature rupture of membranes: comparison of performance indexes

    BACKGROUND: Premature rupture of the membranes (PROM) is most commonly diagnosed using physical examination; however, accurate decision making in ambiguous cases is a major challenge in current obstetric practice. As this may influence a woman's subsequent management, a number of tests designed to assist with confirming a diagnosis of PROM are commercially available. This study sought to evaluate the published data for the accuracy of two amniotic fluid-specific biomarker tests for PROM: insulin-like growth factor binding protein-1 (IGFBP-1 - Actim® PROM) and placental alpha microglobulin-1 (PAMG-1 - AmniSure®). METHODS: Main analysis included all PubMed referenced studies related to Actim® PROM and AmniSure® with available data to extract performance rates. To compare accuracy, a comparison of pooled indexes of both rapid tests was performed. Studies in which both tests were used in the same clinical population were also analysed. Membrane status, whether it was known or a suspected rupture, and inclusion or not of women with bleeding, were considered. RESULTS: All the available studies published in PubMed up to April 2013 were reviewed. Data were retrieved from 17 studies; 10 for Actim® PROM (n = 1066), four for AmniSure® (n = 1081) and three studies in which both biomarker tests were compared directly. The pooled analysis found that the specificity and positive predictive value were significantly higher for AmniSure® compared with Actim® PROM. However, when 762 and 1385 women with known or suspected rupture of membranes, respectively, were evaluated, AmniSure® only remained significantly superior in the latter group. Furthermore, when the two tests were compared directly in the same study no statistically significant differences were observed. Remarkably, women with a history or evidence of bleeding were excluded in all four studies for AmniSure®, in two Actim® PROM studies and in two of the three studies reporting on both tests. CONCLUSIONS: No differences were observed in the performance of the two tests in studies where they were used under the same clinical conditions or in women with known membrane status. Although AmniSure® performed better in suspected cases of PROM, this may need further analysis as exclusion of bleeding may not be representative of the real clinical presentation of women with suspected PROM

    FCNC Top Quark Decays in Extra Dimensions

    The flavor changing neutral top quark decay t -> c X is computed, where X is a neutral standard model particle, in a extended model with a single extra dimension. The cases for the photon, X= \gamma,andaStandardModelHiggsboson,X=H,areanalyzedindetailinanon−linear, and a Standard Model Higgs boson, X = H, are analyzed in detail in a non-linearR_\xi gauge. We find that the branching ratios can be enhanced by the dynamics originated in the extra dimension. In the limit where 1/R >> ->, we have found Br(t -> c \gamma) \simeq 10^{-10} for 1/R = 0.5 TeV. For the decay t -> c H, we have found Br(t -> cH) \simeq 10^{-10} for a low Higgs mass value. The branching ratios go to zero when 1/R -> \infty.Comment: Accepted to be published in the Europ. Phys.     The ArcB Leucine Zipper Domain Is Required for Proper ArcB Signaling

    The Arc two-component system modulates the expression of numerous genes in response to respiratory growth conditions. This system comprises ArcA as the response regulator and ArcB as the sensor kinase. ArcB is a tripartite histidine kinase whose activity is regulated by the oxidation of two cytosol-located redox-active cysteine residues that participate in intermolecular disulfide bond formation. Here, we report that the ArcB protein segment covering residues 70–121, fulfills the molecular characteristics of a leucine zipper containing coiled coil structure. Also, mutational analyses of this segment reveal three different phenotypical effects to be distributed along the coiled coil structure of ArcB, demonstrating that this motif is essential for proper ArcB signaling

    Emerging planetary nebulae within 3D spiral patterns

    We present the first 3D radiation-hydrodynamic simulations of the formation of planetary nebulae (PNe) emerging from 3D spiral patterns. We use the GUACHO code to create 3D spiral structures as a consequence of the distortions on the geometry of the intrinsically isotropic wind of an asymptotic giant branch (AGB) star produced by a companion star in a circular orbit. We found that the orbital period of the binary producing the 3D spiral pattern has consequences on the formation and shaping of the PN itself. Stellar systems with longer period create less entwined 3D spirals, producing PNe with rounder inner cavities, and prevent the expansion of jet towards the polar directions. The spiral fitting procedure used in the literature to predict the binary's orbital period may be misleading in the case of proto-PNe and PNe as spiral patterns are diluted by their own thermal expansion down to the average AGB density profile within a few hundred years and are further disrupted by the action of jets. By adopting a phase of jet ejections between the AGB and post-AGB stages, we are able to recover the morphologies of proto-PNe and PNe that exhibit ring-like structures in their halos.Comment: 12 pages, 12 figures, 1 table; Accepted to MNRA

    Rapid Bacterial and Fungal Successional Dynamics in First Year After Chaparral Wildfire

    The rise in wildfire frequency and severity across the globe has increased interest in secondary succession. However, despite the role of soil microbial communities in controlling biogeochemical cycling and their role in the regeneration of post-fire vegetation, the lack of measurements immediately post-fire and at high temporal resolution has limited understanding of microbial secondary succession. To fill this knowledge gap, we sampled soils at 17, 25, 34, 67, 95, 131, 187, 286, and 376 days after a southern California wildfire in fire-adapted chaparral shrublands. We assessed bacterial and fungal biomass with qPCR of 16S and 18S and richness and composition with Illumina MiSeq sequencing of 16S and ITS2 amplicons. Fire severely reduced bacterial biomass by 47%, bacterial richness by 46%, fungal biomass by 86%, and fungal richness by 68%. The burned bacterial and fungal communities experienced rapid succession, with 5-6 compositional turnover periods. Analogous to plants, turnover was driven by fire-loving pyrophilous microbes, many of which have been previously found in forests worldwide and changed markedly in abundance over time. Fungal secondary succession was initiated by the Basidiomycete yeast Geminibasidium, which traded off against the filamentous Ascomycetes Pyronema, Aspergillus, and Penicillium. For bacteria, the Proteobacteria Massilia dominated all year, but the Firmicute Bacillus and Proteobacteria Noviherbaspirillum increased in abundance over time. Our high-resolution temporal sampling allowed us to capture post-fire microbial secondary successional dynamics and suggest that putative tradeoffs in thermotolerance, colonization, and competition among dominant pyrophilous microbes control microbial succession with possible implications for ecosystem function
