Interaction and uptake of exosomes by ovarian cancer cells
<p>Abstract</p> <p>Background</p> <p>Exosomes consist of membrane vesicles that are secreted by several cell types, including tumors and have been found in biological fluids. Exosomes interact with other cells and may serve as vehicles for the transfer of protein and RNA among cells.</p> <p>Methods</p> <p>SKOV3 exosomes were labelled with carboxyfluoresceine diacetate succinimidyl-ester and collected by ultracentrifugation. Uptake of these vesicles, under different conditions, by the same cells from where they originated was monitored by immunofluorescence microscopy and flow cytometry analysis. Lectin analysis was performed to investigate the glycosylation properties of proteins from exosomes and cellular extracts.</p> <p>Results</p> <p>In this work, the ovarian carcinoma SKOV3 cell line has been shown to internalize exosomes from the same cells via several endocytic pathways that were strongly inhibited at 4°C, indicating their energy dependence. Partial colocalization with the endosome marker EEA1 and inhibition by chlorpromazine suggested the involvement of clathrin-dependent endocytosis. Furthermore, uptake inhibition in the presence of 5-ethyl-N-isopropyl amiloride, cytochalasin D and methyl-beta-cyclodextrin suggested the involvement of additional endocytic pathways. The uptake required proteins from the exosomes and from the cells since it was inhibited after proteinase K treatments. The exosomes were found to be enriched in specific mannose- and sialic acid-containing glycoproteins. Sialic acid removal caused a small but non-significant increase in uptake. Furthermore, the monosaccharides D-galactose, α-L-fucose, α-D-mannose, D-N-acetylglucosamine and the disaccharide β-lactose reduced exosomes uptake to a comparable extent as the control D-glucose.</p> <p>Conclusions</p> <p>In conclusion, exosomes are internalized by ovarian tumor cells via various endocytic pathways and proteins from exosomes and cells are required for uptake. On the other hand, exosomes are enriched in specific glycoproteins that may constitute exosome markers. This work contributes to the knowledge about the properties and dynamics of exosomes in cancer.</p

Effect of microirrigation on occurrence of asparagus beetles in asparagus cultivation on the very light soil
Doświadczenia polowe zostały przeprowadzone w latach 2005-2006 we wsi Kruszyn Krajeński koło Bydgoszczy. Celem przeprowadzonego doświadczenia było określenie wpływu różnych sposobów nawadniania wybranych mieszańców oraz odmian szparaga uprawianego na glebie bardzo lekkiej na występowanie poskrzypek. Badania prowadzono jako dwuczynnikowe w układzie losowanych podbloków, w czterech replikacjach. Czynnikiem pierwszego rzędu było nawadnianie: O - bez nawadniania (kontrola), K - nawadnianie kroplowe,
M - mikrozraszanie. Drugim czynnikiem prowadzonych doświadczeń były dwa mieszańce szparaga literowo oznaczone przez producenta: Ap, Gr oraz odmiana Schwetzinger Meisterschuss. Trzykrotnie w okresie wzrostu pędów asymilacyjnych (w lipcu i sierpniu, co 3 tygodnie) przeprowadzono makroskopową obserwację stanu liczebności owadów doskonałych i larw poskrzypki szparagowej i dwunastokropkowej przebywających na wszystkich roślinach rosnących na poszczególnych poletkach doświadczalnych. Powierzchnia poletka doświadczalnego wynosiła 14,5 m2 (23 rośliny x 0,35 m x 1,8 m). Mieszańce szparaga: Ap, Gr oraz ‘Schwetzinger Meisterschuss', chętniej zasiedlane były przez imagines i larwy poskrzypki dwunastokropkowej, w porównaniu do poskrzypki szparagowej. Mikrozraszanie testowanych roślin szparaga: Ap, Gr i ‘Schwetzinger Meisterschuss' nie różnicowało ich podatności na zasiedlenie przez owady doskonałe oraz larwy poskrzypki dwunastokropkowej i szparagowej. Podobna sytuacja miała miejsce w przypadku stosowania nawadniania kroplowego. Mieszaniec szparaga Ap był preferowany bardziej przez imagines oraz larwy poskrzypki dwunastokropkowej i szparagowej, w porównaniu do mieszańca Gr oraz odmiany ‘Schwetzinger Meisterschuss', niezależnie od stosowanego wariantu nawadniania.The field experiments were carried out in two consecutive years (2005-
2006) at Kruszyn Krajeński near Bydgoszcz. The aim of the study was the influence
of different systems of irrigation in asparagus hybrids cultivation on the very
light soil on the occurrence of asparagus beetles. The experiments were conducted
in a randomized block design of a two-factorial system with four replications. The
first tested factor was irrigation in three following variants: O – non-irrigated
plots (control), K – drip-irrigated plots and M – microirrigated plots. The second
factor was associated with two asparagus hybrids (called by producer): Ap, Gr
and one cultivar ‘Schwetzinger Meisterschuss’. The number of adults and larvae
of both asparagus beetles was observed on every single plot area; three times
during the vegetation period from July to August (beginning from the first decade
of July and continuing every third week). The harvest plot area was 14.5 m2
(23 plants x 35 cm x 180 cm). Generally, the adults and larvae of twelve-spotted
asparagus beetles were more numerous than common asparagus beetles on asparagus
hybrids plants: Ap, Gr and ‘Schwetzinger Meisterschuss’. Microirrigation
applied on asparagus hybrids wasn’t related with these two asparagus beetles did
not susceptible to. Much the same situation was observed when the drip irrigation
was used. The adults and larvae of both asparagus beetles preferred Ap than Gr
and ‘Schwetzinger Meisterschuss’ hybrids plants, irrespectively from irrigation

Effect of dried material from peppermint (Mentha piperita L.) on the development and the fecundity of grain weevil (Sitophilus granarius L.)
W prezentowanej pracy oceniano potencjalne możliwości wykorzystywania jednego z alternatywnych sposobów obniżania liczebności szkodników magazynowych, jakim jest bezpośrednie dodawanie naturalnych substancji roślinnych do przechowywanego ziarna zbóż. Podjęto problem dotyczący oddziaływania produktu pochodzenia roślinnego - proszku z mięty pieprzowej (Mentha piperita L.) na populację wołka zbożowego. Doświadczenia prowadzono w klimatyzowanym pomieszczeniu, na ziarnie pszenicy konsumpcyjnej. Proszek stosowano w dwóch dawkach (1,0 i 0,5 g na 100 g pszenicy) oraz w dwóch formach: zmieszany z ziarnem i zamknięty w osłonce z gazy młyńskiej. Na podstawie uzyskanych wyników stwierdzono zależność długości cyklu rozwojowego szkodnika i jego płodności od wysokości dawki suszu oraz sposobu jego aplikacji. Notowano większą efektywność proszku zmieszanego z ziarnem. Wraz ze wzrostem dawki testowanego suszu z mięty pieprzowej obserwowano: wydłużenie czasu rozwoju pokolenia, skracanie trwania pojawu chrząszczy i spadek płodności szkodnika.Combating the storage pests has been a serious economic problem for years. So far pest control involved mostly chemical fumigation of storage places. Today, since January 1, 2014 we have been obliged to obey the legal regulations of the European Parliament and the Council of the European Union which order a blanket application of integrated methods, with the priority of the use of non-chemical and natural methods, which is connected with limiting the application of chemicals to those with a selective effect and demonstrating a lack of negative effect on the human and animal health and on the environment. The present paper provides an analysis of one of the alternative methods of decreasing the number of storage pests; direct adding natural plant substances (i.e. dried and plant parts crushed to powder) to the grain stored. There has been investigated the problem of the effect of the product of plant origin: powder from peppermint (Mentha piperita L.) on the population of grain weevil. The experiments were performed in an air-conditioned room, at the moisture of 70-80%, at the temperature of 25°C, and involved milling grain. The powder, made from peppermint, was tested at two rates (1.0 and 0.5 g per 100 g) of wheat as well as in two forms: mixed with grain and closed in a mill gauze cover. Each series consisted of 10 replications. One replication corresponded to one glass flask 250 ml in capacity filled with 100 g of wheat, in which 10 males and 10 females at the age of 2-5 days were placed. To monitor the after-effect of the plant dried material tested, the second experiment was performed, without peppermint added, which involved the use of weevils derived from respective series of the first experiment. For each series the following were calculated: the generation development time, hatching time as well as the fertility rate which was determined based on the ratio of the number of F1 individuals to the number of P generation females. The results demonstrated a varied reaction of grain weevil to the dried peppermint material. There was shown the dependence of the pest development cycle time and the pest fertility on the rate of the dose of dried material as well as its application method. In the experiments analyzing a direct effect, with increasein of the dose of the dried peppermint material tested, the following were observed: a prolonged generation development time, shortening of the period of beetle hatching and a decreased pest fertility. There was noted a higher effectiveness of dried material mixed with grain, as compared with that added in covers of mill gauze. However, no significant after-effect was reported
Influence of drip irrigation on insects occurence in cultivation of two cultivars of summer squash (Cucurbita pepo L.)
Doświadczenie polowe prowadzono w Kruszynie Krajeńskim koło Bydgoszczy
w latach 2005–2006. Badano wpływ nawadniania kroplowego w uprawie
dwóch odmian dyni zwyczajnej: ‘Danka’ i ‘Sweet Dumpling’ na zasiedlenie przez
owady.
Na podstawie otrzymanych wyników stwierdzono, że na dyni zwyczajnej
odmiany ‘Danka’ występowało istotnie więcej owadów szkodliwych. Z entomofauny
fitofagicznej najwięcej wystąpiło przedstawicieli rzędu Homoptera, a wśród
nich należących do rodziny skoczkowate. Istotnie więcej osobników wystąpiło na
roślinach nawadnianych (44 sztuk x poletko-1), w porównaniu do kontrolnych
(34 sztuk x poletko-1). Skoczek ziemniaczak stanowił 65 %, skoczek czarnoplamek
– 17 %, a skoczek sześciorek – 7 % wszystkich oznaczonych skoczkowatych. Spośród
rzędu Heteroptera wystąpiły zmienik lucernowiec i wysmułek paskorogi oraz
pożyteczne (dziubałkowate i zażartkowate).
Odmiana ‘Sweet Dumpling’ była również w znacznym stopniu zasiedlana
przez owady należące do rzędu pluskwiaków równoskrzydłych – Homoptera.
Szczególnie licznie wystąpiły tu skoczkowate: skoczek ziemniaczak stanowił
42%, skoczek czarnoplamek – 32%, a skoczek sześciorek i zgłobik smużkowany
po 10 % odłowionej fauny Cicadellidae. Heteroptera pożyteczne i szkodliwe
zasiedlały analizowaną dynię nielicznie.The field experiments were carried out in Kruszyn Krajeński near
Bydgoszcz during the 2005–2006. The influence of drip irrigation on insects occupying
of two cultivars of summer squash ‘Danka’ and ‘Sweet Dumpling’ were
investigated.
On the base of the received results was proved that on the squash ‘Danka’
phytophagous insects significantly more appeard. From phytophagous insects the
largest group was Homoptera, and among them Cicadellidae. The significantly
large number of insects were found on the drip irrigated plants (44 pcs x plot-1), in
comparison to non-irrigated (34 pcs x plot-1). The Empoasca pteridis Dahlbom.,
Eupteryx atropunctata Goeze and Macrosteles laevis Ribaut, were represented in
total number of all Cicadellidae in 65, 17 and 7%, respectively. Heteroptera
was represented by Lygus rugulipennis Popp., Trigonotylus coelestialium Kirk.
and beneficial Anthocoridae as well as Nabidae.
The ‘Sweet Dumpling’ cultivar was also occupying more often by the Homoptera.
The most abundant insects were Empoasca pteridis Dahlbom (42%),
Eupteryx atropunctata Goeze (32%), Macrosteles laevis Ribaut and Psammotettix
alienus Dahlbom (10%) of all Cicadellidae. Beneficial and phytophagous Heteroptera
appeared as less abundant
Effect of nitrogen subsurface drip irrigation of chosen asparagus cultivars on the occurrence of asparagus beetles
Celem przeprowadzonego doświadczenia było określenie wpływu stosowania fertygacji podpowierzchniowej azotem różnych odmian szparaga na występowanie poskrzypek.

Imagines poskrzypki dwunastokrokowej preferowały fertygowane podpowierzchniowo odmiany szparaga: ‘Atlas', ‘UC-157', ‘Grande' i ‘Rapsody', niż rośliny nienawadniane tych odmian. Liczebność larw gatunku nie różnicowały, zarówno stosowanie fertygacji, jak i właściwości odmianowe szparagów. Stadium doskonałe poskrzypki szparagowej zasiedlało chętniej nienawadniane odmiany szparaga: ‘Grande' i ‘Purple Passion' a larwy odmianę Rapsody, w porównaniu do roślin fertygowanych podpowierzchniowo.The aim of this study was the effect of the subsurface drip irrigation with

with hand sowing nitrogen fertilizer and subsurface drip irrigated plots with nitrogen
fertigation. The second factor was associated with asparagus cultivars: 5 from
USA: Apollo, Atlas, UC-157, Grande and Purple Passion, and 3 from Germany:
Ramada, Rapsody and Ravel. The number of adults and larvae of both asparagus
beetles was observed on every single plot area; three times during the vegetation
period from July to August (beginning from the first decade of July and continuing
every third week). The harvest plot area was 14,7 m2 (20 plants x 0,35m x 2,1m).
The adults and larvae of twelve-spotted asparagus beetles were more numerous
than common asparagus beetles on tested asparagus cultivars. The adults
of twelve-spotted asparagus beetles prefered subsurface drip irrigation asparagus
plants: Atlas, UC-157, Grande and Rapsody, than non-irrigated plants of these
cultivars. Both subsurface drip irrigation and asparagus cultivars did not affect
the occurrence of larvae of this species. The adults of common asparagus beetles
were significantly more numerous on the non-irrigated asparagus cultivars:
Grande and Purple Passion and the larvae of this species on the cultivar Rapsody,
in comparison with subsurface drip irrigation plants of these cultivars
Effect of sprinkling irrigation and varied nitrogen fertilization of millet cv. Gierczyckie on occurrence of phytophagous entomofauna
Badano wpływ nawadniania oraz nawożenia azotowego prosa odmiany
‘Gierczyckie’ na występowanie fitofagicznej entomofauny o kłująco-ssącym aparacie
gębowym. Doświadczenie polowe prowadzono w latach 2005-2006 w Kruszynie
Krajeńskim koło Bydgoszczy.
Stwierdzono, że przylżeńce oraz pluskwiaki stanowiły najliczniejszą grupę,
która zasiedlała rośliny prosa. Odławiano zdecydowanie najmniejszą liczebność
thysanopterofauny, gdy rośliny traktowano średnimi dawkami nawożenia azotowego.
Owady te preferowały rośliny nawadniane. Kwietniczkowate (Phlaeothripidae)
stanowiły zdecydowanie najliczniejszą grupę owadów odławianych spośród
przylżeńców. Liczebność odławianych pluskwiaków była niższa, w porównaniu
do przylżeńców. Hemiptera najliczniej występowały na roślinach nawadnianych,
które traktowano wysokim poziomem nawożenia azotowego. Reprezentowane one
były przez kilka rodzin, takich jak: skoczkowate, tasznikowate, mszycowate i szydlakowate.
Najliczniejszym przedstawicielem Cicadellidae był zgłobik smużkowany
– Psammotettix alienus Dahlbom. Skoczek sześciorek – Macrosteles laevis
Ribaut oraz skoczek ziemniaczak – Empoasca pteridis Dahlbom wystąpiły w nieco
mniejszym nasileniu. Spośród rodziny mszycowatych najliczniej odławiano
mszycę zbożową i mszycę czeremchowo-zbożową. Stosowanie zróżnicowanego
nawadniania oraz nawożenia azotowego w uprawie prosa wpływało na liczebność
fitofagicznych pluskwiaków z rodziny tasznikowatych: zmienika lucernowca - Lygus
rugulipennis Popp. i wysmułka paskorogiego - Trigonotylus coelestialiumKirk. Pierwszy z wymienionych Miridae zdecydowanie preferował rośliny nawadniane
oraz nawożone wyższą dawką azotu.The sprinkling irrigation and varied nitrogen fertilisation of millet cv.

The entomological experiments were conducted in 2005 and 2006 in
Kruszyn Krajeński near Bydgoszcz.
The largest group of insects were Thysanoptera and Hemiptera. When we
used the middle rate of nitrogen fertilization, Thysanoptera was significantly less
numerous. They preffered the irrigation plants. From Thysanoptera the most
abundant were Phlaeothripidae. Number of Hemiptera was significantly less than
Thysanoptera. When the high rate of nitrogen fertilization was used, the numer of
Hemiptera was high, too. They were represented by Cicadellidae, Miridae,
Aphididae and Delphacidae families. Psammotettix alienus Dahlbom. was the
most numerous insects from Cicadellidae. The numbers of Macrosteles laevis
Ribaut and Empoasca pteridis Dahlbom was lower. The most frequently caught of
Aphididae were the following insects: Sitobion avenae (F.) H.R.L. and Rhopalosiphum
padi L. The usage of irrigation and varied nitrogen fertilization in millet
cultivation, affected on numerous of phytophagous insects such as: Lygus rugulipennis
Popp. and Trigonotylus coelestialium Kirk. First of theirs preffered irrigation
plants with higher rate of nitrogen fertilization
