In Vivo Inhibition Followed by Exogenous Supplementation Demonstrates Galactopoietic Effects of Prolactin on Mammary Tissue and Milk Production in Dairy Cows
ABSTRACT: It has been previously shown that the long-term inhibition of milking-induced prolactin (PRL) release by quinagolide (QN), a dopamine agonist, reduces milk yield in dairy cows. To further demonstrate that PRL is galactopoietic in cows, we performed a short-term experiment that used PRL injections to restore the release of PRL at milking in QN-treated cows. Nine Holstein cows were assigned to treatments during three 5-d periods in a 3 × 3 Latin square design: 1) QN: twice-daily i.m. injections of 1 mg of QN; 2) QN-PRL: twice-daily i.m. injections of 1 mg of QN and twice-daily (at milking time) i.v. injections of PRL (2 μg/kg body weight); and 3) control: twice-daily injections of the vehicles. Mammary epithelial cells (MEC) were purified from milk so that their viability could be assessed, and mammary biopsies were harvested for immunohistological analyses of cell proliferation using PCNA and STAT5 staining. In both milk-purified MEC and mammary tissue, the mRNA levels of milk proteins and BAX were determined using real-time reverse-transcription PCR. Daily QN injections reduced milking-induced PRL release. The area under the PRL curve was similar in the control and PRL injection treatments, but the shape was different. The QN treatment decreased milk, lactose, protein, and casein production. Injections of PRL did not restore milk yield but tended to increase milk protein yield. In mammary tissue, the percentage of STAT5-positive cells was reduced during QN but not during QN-PRL in comparison with the control treatment. The percentage of PCNA-positive cells was greater during QN-PRL injections than during the control or QN treatment and tended to be lower during QN than during the control treatment. In milk-purified MEC, κ-casein and α-lactalbumin mRNA levels were lower during QN than during the control treatment, but during QN-PRL, they were not different from the control treatment. In mammary tissue, the BAX mRNA level was lower during QN-PRL than during QN. The number of MEC exfoliated into milk was increased by QN injections but tended to be decreased by PRL injections. Injections of PRL also increased the viability of MEC harvested from milk. Although PRL injections at milking could not reverse the effect of QN treatment on milk production, their effects on cell survival and exfoliation and on gene expression suggest that the effect of QN treatment on the mammary gland is due to QN’s inhibition of PRL secretion
Economic Analysis of Knowledge: The History of Thought and the Central Themes
Following the development of knowledge economies, there has been a rapid expansion of economic analysis of knowledge, both in the context of technological knowledge in particular and the decision theory in general. This paper surveys this literature by identifying the main themes and contributions and outlines the future prospects of the discipline. The wide scope of knowledge related questions in terms of applicability and alternative approaches has led to the fragmentation of research. Nevertheless, one can identify a continuing tradition which analyses various aspects of the generation, dissemination and use of knowledge in the economy
Focus and Intensification in the Semantics of Brow Raise
International audienceWe argue that in American Sign Language (ASL), Brow Raise has two sorts of functions that can be distinguished by timing: it may serve well-known information-theoretic functions that can, among others, realize focus; but it may also intensify gradable constructions – a far less well-known observation. While Brow Raise on an expression can fulfill both functions, Brow Raise right before an expression preferentially has an information-theoretic function. The main findings are replicated on some examples from LSF (French Sign Language). Strikingly, these two functions mirror those found for 'stress' (= emphasis) by Bergen 2016, who argued for a unified analysis of information-theoretic effects and of intensificational effects. We sketch a unified analysis within Alternative Semantics, and discuss a further possibility within a simplified version of Bergen's own theory of 'noise-reduction' (Bergen 2016). An extension of our ASL data shows that related generalizations hold when Brow Raise is applied to a highly iconic construction (here involving a helicopter path): depending on timing, Brow Raise may serve to evoke alternatives or to intensify part of the construction
Meaningful Blurs: the Sources of Repetition-based Plurals in ASL
International audienceIn several sign languages, plurals can be realized with unpunctuated or punctuated repetitions of a noun, with different semantic implications; similar repetitionbased plurals have been described in some homesigns and silent gestures. Unpunctuated repetitions often get approximate 'at least' readings while punctuated repetitions typically correspond to 'exactly' readings. The prevalence of these mechanisms could be thought to be a case in which Universal Grammar does not just specify the abstract properties of grammatical elements, but also their phonological realization, at least in the visual modality. We explore an alternative in which punctuated and unpunctuated repetitions arise from general properties of iconic representations. On an empirical level, we argue that in ASL (American Sign Language), punctuated and unpunctuated repetitions are unlikely to be an exclusively grammatical mechanism, as they can be found with purely iconic (word-free) representations. On a theoretical level, we argue for a modular account with three components. First, repetition-based plurals can create a simplified pictorial representation. Second, unpunctuated repetitions give rise to pictorial vagueness, resolved by way of quantification over precisifications. Third, a pragmatic process involving strategic reasoning maps these vague representations onto a set of candidate linguistic meanings, including some 'at least' plural readings that are best expressed by unpunctuated repetitions
Iconic Plurality
International audienceASL (American Sign Language) can express plurals by repeating a noun, in an unpunctuated fashion, in different parts of signing space. We argue that this construction may come with a rich (and at-issue) iconic component: the geometric arrangement of the repetitions provides information about the arrangement of the denoted plurality; in addition, the number and speed of the repetitions provide information about the size of the denoted plurality. Interestingly, the shape of the repetitions may introduce a new singular discourse referent when a vertex can be inferred to denote a singular object. Thus one may point towards the first or last iteration of a horizontal repetition of BOOK to denote the left-or right-edge of the corresponding row. This yields a remarkable interaction between iconic semantics and standard logical semantics. We show that our analysis extends to 'punctuated' repetitions, which involve clearly individuated iterations of a singular noun. While these may initially look like coordinated indefinites, they are better handled by the same iconic framework as plural, unpunctuated repetitions. Some repetition-based mass terms also give rise to iconic effects, and to different readings depending on whether the repetition is continuous, unpunctuated, or punctuated. Our analysis highlights the need for a formal semantics with iconicity to study the integration of such iconic and logical conditions. It also raises a question: can similar facts be found in spoken language when gestures are taken into account? We suggest that several effects can be replicated, especially when one considers examples involving 'pro-speech gestures' (= gestures that fully replace some spoken expressions)
