    Lithium chloride reinforces the regeneration-promoting effect of chondroitinase ABC on rubrospinal neurons after spinal cord injury

    After spinal cord injury, enzymatic digestion of chondroitin sulfate proteoglycans promotes axonal regeneration of central nervous system neurons across the lesion scar. We examined whether chondroitinase ABC (ChABC) promotes the axonal regeneration of rubrospinal tract (RST) neurons following injury to the spinal cord. The effect of a GSK-3ÎČ inhibitor, lithium chloride (LiCl), on the regeneration of axotomized RST neurons was also assessed. Adult rats received a unilateral hemisection at the seventh cervical spinal cord segment (C7). Four weeks after different treatments, regeneration of RST axons across the lesion scar was examined by injection of Fluoro-Gold at spinal segment T2, and locomotor recovery was studied by a test of forelimb usage. Injured RST axons did not regenerate spontaneously after spinal cord injury, and intraperitoneal injection of LiCl alone did not promote the regeneration of RST axons. Administration of ChABC at the lesion site enhanced the regeneration of RST axons by 20%. Combined treatment of LiCl together with ChABC significantly increased the regeneration of RST axons to 42%. Animals receiving combined treatment used both forelimbs together more often than animals that received sham or single treatment. Immunoblotting and immunohistochemical analysis revealed that LiCl induced the expression of inactive GSK-3ÎČ as well as the upregulation of Bcl-2 in injured RST neurons. These results indicate that in vivo, LiCl inhibits GSK-3ÎČ and reinforces the regeneration-promoting function of ChABC through a Bcl-2-dependent mechanism. Combined use of LiCl together with ChABC could be a novel treatment for spinal cord injury.published_or_final_versio

    Itinerant ferromagnetism and intrinsic anomalous Hall effect in amorphous iron-germanium

    The amorphous iron-germanium system (a-FexGe1-x) lacks long-range structural order and hence lacks a meaningful Brillouin zone. The magnetization of a-FexGe1-x is well explained by the Stoner model for Fe concentrations x above the onset of magnetic order around x=0.4, indicating that the local order of the amorphous structure preserves the spin-split density of states of the Fe-3d states sufficiently to polarize the electronic structure despite k being a bad quantum number. Measurements reveal an enhanced anomalous Hall resistivity ρxyAH relative to crystalline FeGe; this ρxyAH is compared to density-functional theory calculations of the anomalous Hall conductivity to resolve its underlying mechanisms. The intrinsic mechanism, typically understood as the Berry curvature integrated over occupied k states but shown here to be equivalent to the density of curvature integrated over occupied energies in aperiodic materials, dominates the anomalous Hall conductivity of a-FexGe1-x (0.38≀x≀0.61). The density of curvature is the sum of spin-orbit correlations of local orbital states and can hence be calculated with no reference to k space. This result and the accompanying Stoner-like model for the intrinsic anomalous Hall conductivity establish a unified understanding of the underlying physics of the anomalous Hall effect in both crystalline and disordered systems

    Influences of crustal thickening in the Tibetan Plateau on loading modeling and inversion associated with water storage variation

    We use the average crustal structure of the CRUST1.0 model for the Tibetan Plateau to establish a realistic earth model termed as TC1P, and data from the Global Land Data Assimilation System (GLDAS) hydrology model and Gravity Recovery and Climate Experiment (GRACE) data, to generate the hydrology signals assumed in this study. Modeling of surface radial displacements and gravity variation is performed using both TC1P and the global Preliminary Reference Earth Model (PREM). Furthermore, inversions of the hydrology signals based on simulated Global Positioning System (GPS) and GRACE data are performed using PREM. Results show that crust in TC1P is harder and softer than that in PREM above and below a depth of 15 km, respectively, causing larger differences in the computed load Love numbers and loading Green's functions. When annual hydrology signals are assumed, the differences of the radial displacements are found to be as large as approximately 0.6 mm for the truncated degree of 180; while for hydrology-trend signals the differences are very small. When annual hydrology signals and the trends are assumed, the differences in the surface gravity variation are very small. It is considered that TC1P can be used to efficiently remove the hydrological effects on the monitoring of crustal movement. It was also found that when PREM is used inappropriately, the inversion of the hydrology signals from simulated annual GPS signals can only recover approximately 88.0% of the annual hydrology signals for the truncated degree of 180, and the inversion of hydrology signals from the simulated trend GPS signals can recover approximately 92.5% for the truncated degree of 90. However, when using the simulated GRACE data, it is possible to recover almost 100%. Therefore, in future, the TC1P model can be used in the inversions of hydrology signals based on GPS network data. PREM is also valid for use with inversions of hydrology signals from GRACE data at resolutions of approximately 220 km and larger.published_or_final_versio

    A novel 16-channel wireless system for electroencephalography measurements with dry spring-loaded sensors

    Understanding brain function using electroencephalography (EEG) is an important issue for cerebral nervous system diseases, especially for epilepsy and Alzheimer's disease. Many EEG measurement systems are used reliably to study these diseases, but their bulky size and the use of wet sensors make them uncomfortable and inconvenient for users. To overcome the limitations of conventional EEG measurement systems, a wireless and wearable multichannel EEG measurement system is proposed in this paper. This system includes a wireless data acquisition device, dry spring-loaded sensors, and a sizeadjustable soft cap. We compared the performance of the proposed system using dry versus conventional wet sensors. A significant positive correlation between readings from wet and dry sensors was achieved, thus demonstrating the performance of the system. Moreover, four different features of EEG signals (i.e., normal, eye-blinking, closed-eyes, and teeth-clenching signals) were measured by 16 dry sensors to ensure that they could be detected in real-life cognitive neuroscience applications. Thus, we have shown that it is possible to reliably measure EEG signals using the proposed system. This paper presents novel insights into the field of cognitive neuroscience, showing the possibility of studying brain function under real-life conditions. © 2014 IEEE

    Single channel wireless EEG device for real-time fatigue level detection

    © 2015 IEEE. Driver fatigue problem is one of the important factors of traffic accidents. Recent years, many research had investigated that using EEG signals can effectively detect driver's drowsiness level. However, real-time monitoring system is required to apply these fatigue level detection techniques in the practical application, especially in the real-road driving. Therefore, it required less channels, portable and wireless, real-time monitoring and processing techniques for developing the real-time monitoring system. In this study, we develop a single channel wireless EEG device which can real-time detect driver's fatigue level on the mobile device such as smart phone or tablet. The developed device is investigated to obtain a better and precise understanding of brain activities of mental fatigue under driving, which is of great benefit for devolvement of detection of driving fatigue system. This system consists of a Bluetooth-enabled one channel EEG, a regression model, and smartphone, which was a platform recording and transforming the raw EEG data to useful driving status. In the experiment, this was a sustained-attention driving task to implement in a virtual-reality (VR) driving simulator. To training model and develop the system, we were performed for 15 subjects to study Electroencephalography (EEG) brain dynamics by using a mobile and wireless EEG device. Based on the outstanding training results, the leave-one-subject-out cross validation test obtained 90% fatigue detection accuracy. These results indicate that the combination of a smartphone and wireless EEG device constitutes an effective and easy wearable solution for detecting and preventing driver fatigue in real driving environments

    Error Arising from Original Approximation in Photothermal Measurement of Weak Absorption of Optical Thin Film

    The technique of photothermal deflection spectroscopy (PTDS) for absorption measurement of optical thin film is simple in installation and fast in detection. However, while measurable absorbance reaches the order of magnitude of 10-7, e.g. in studies of optical thin film and investigation of laser damage mechanisms [1,2], we are still doubtful of its precision because of the lack of independent evaluation with available instruments whose precision could reach this lower limit. This work aims at giving a theoretical investigation on the source of error in order to obtain a realistic estimate of measurement error and hence to find optimal way to minimize measurement error in applying PTDS to optical thin film.</p

    Massive expansion and cryopreservation of functional human induced pluripotent stem cell-derived cardiomyocyte

    Since the discovery of human induced pluripotent stem cells (hiPSCs), numerous strategies have been established to efficiently derive cardiomyocytes from hiPSCs (hiPSC-CMs). Here, we describe a cost-effective strategy for the subsequent massive expansion (>250-fold) of high-purity hiPSC-CMs relying on two aspects: removal of cell-cell contacts and small-molecule inhibition with CHIR99021. The protocol maintains CM functionality, allows cryopreservation, and the cells can be used in downstream assays such as disease modeling, drug and toxicity screening, and cell therapy.     TEMPRANILLO is a regulator of juvenility in plants

    Many plants are incapable of flowering in inductive daylengths during the early juvenile vegetative phase (JVP). Arabidopsis mutants with reduced expression of TEMPRANILLO (TEM), a repressor of FLOWERING LOCUS T (FT) had a shorter JVP than wild-type plants. Reciprocal changes in mRNA expression of TEM and FT were observed in both Arabidopsis and antirrhinum, which correlated with the length of the JVP. FT expression was induced just prior to the end of the JVP and levels of TEM1 mRNA declined rapidly at the time when FT mRNA levels were shown to increase. TEM orthologs were isolated from antirrhinum (AmTEM) and olive (OeTEM) and were expressed most highly during their juvenile phase. AmTEM functionally complemented AtTEM1 in the tem1 mutant and over-expression of AmTEM prolonged the JVP through repression of FT and CONSTANS (CO). We propose that TEM may have a general role in regulating JVP in herbaceous and woody species
