Finishing the euchromatic sequence of the human genome
The sequence of the human genome encodes the genetic instructions for human physiology, as well as rich information about human evolution. In 2001, the International Human Genome Sequencing Consortium reported a draft sequence of the euchromatic portion of the human genome. Since then, the international collaboration has worked to convert this draft into a genome sequence with high accuracy and nearly complete coverage. Here, we report the result of this finishing process. The current genome sequence (Build 35) contains 2.85 billion nucleotides interrupted by only 341 gaps. It covers ∼99% of the euchromatic genome and is accurate to an error rate of ∼1 event per 100,000 bases. Many of the remaining euchromatic gaps are associated with segmental duplications and will require focused work with new methods. The near-complete sequence, the first for a vertebrate, greatly improves the precision of biological analyses of the human genome including studies of gene number, birth and death. Notably, the human enome seems to encode only 20,000-25,000 protein-coding genes. The genome sequence reported here should serve as a firm foundation for biomedical research in the decades ahead
First Time Expectant Fathers’ Attitudes, Actions and Well-being in Regard to Work & Family Issues
Expectant fathers’ attitudes, decision-making processes, and sense of well-being relating to the challenges
of work and family life as well as their conceptualizations of the role of father were explored
with a purposive sample. In 2000, twenty-five married first time expectant fathers from central
Texas in the U.S.A. completed self-administered questionnaires and fourteen were interviewed.
While all fathers placed the highest priority on ‘breadwinning’, the majority believed that childcare
and domestic duties should be shared equally when both parents are employed full time. Only two
interviewed fathers described truly egalitarian household practices. Seven interviewees were ‘traditional’
in expectations and actions. The remaining five held traditional expectations for fathers but
reported increased responsibility as helpers with domestic responsibilities and child rearing. The
fathers’ often conflicting expectations with practice suggest that the challenges couples face after the
baby is born will impact their well-being and health. Supportive programs and policies would help
Long-term management of ovarian neoplasia in two cockatiels (Nymphicus hollandicus)
Cockatiels (Nymphicus hollandicus) are commonly diagnosed with ovarian neoplasia. However, there is very little information regarding medical management of this disease condition and subsequent patient response. Long-term medical therapy of 2 cockatiels eventually diagnosed with ovarian neoplasia is described along with responses to the treatment regimens. Each bird had initial signs consistent with reproductive disease (chronic egg laying, ascites, and lethargy) and respiratory distress. The diagnosis of ovarian adenocarcinoma was confirmed on postmortem examination of both birds. The birds were conservatively managed by periodic coelomocentesis and gonadotropin-releasing hormone (GnRH) agonist administration for 9 and 25 months, respectively. A positive response to GnRH agonist therapy was documented in 1 of the 2 birds. These 2 cases demonstrate that periodic coelomocentesis with or without GnRH agonist therapy may be a viable option for the long-term management of ovarian neoplasia and reproductive-organ-associated ascites in cockatiels
Reconciling biodiversity conservation and food security: scientific challenges for a new agriculture
Production ecology and conservation biology have long focused on providing the knowledge base for intensive food production and biodiversity conservation, respectively. With increasing global food insecurity and continuing biodiversity decline, we show that the largely separate development of these fields is counterproductive. Scenario analyses suggest that feeding the world is possible without further encroachment of agriculture into natural ecosystems. Without ignoring the necessary demographic, socio-economic, institutional and governance requirements, we make the case for a science that develops the best ecological means to produce food in a way that has substantially less negative effects on biodiversity and associated ecosystem services and, indeed, should be able to contribute to their persistence and enhancement. Recent developments in trait-based ecology should soon make it possible to adapt and (re-)design agroecosystems to meet both goals of biodiversity conservation and food security. However, there are real tensions between, on the one hand, the opportunity costs of biodiversity conservation (for direct use and for conversion to agriculture) and on the other hand, the ecosystem service values and option values associated with biodiversity. We elaborate the management of plant genetic resources as a metaphor of the tensions between such values of biodiversity and ecosystem services in general. We conclude that significant changes in policies, institutions and practices are necessary to make advances in ecology work for reconciling biodiversity conservation and food security
Short-Cycle Therapy in Adolescents after Continuous Therapy with Established Viral Suppression: The Impact on Viral Load Suppression
This was a proof-of-principle study to evaluate the impact of short cycle therapy (SCT; 4 days on/3 days off) in adolescents and young adults with good viral suppression on a protease inhibitor-based antiretroviral regimen. Subjects were recruited by the Adolescent Trials Network for HIV/AIDS Interventions and the Pediatric AIDS Clinical Trials Group. Subjects were infected either through perinatal/early childhood transmission or later via risk behaviors. All subjects were required to have at least 6 months of documented viral suppression below 400 copies/ml plus a preentry value below 200 copies/ml and an entry CD4+ T cell count above 350 cells/mm3. Of the 32 subjects enrolled, 12 (37.5%) had confirmed viral load rebound >400 copies, with 18 subjects (56%) coming off for any reason. The majority of subjects resuppressed when placed back onto continuous therapy using the same agents. Although no difference was found in virologic rebound rates between the early and later transmission groups, those infected early in life had higher rates of coming off SCT for any reason. There was no impact of SCT on the CD4+ T cell counts in those who remained on study or those who came off SCT for any reason. Subjects demonstrated good adherence to the SCT regimen. This study suggests that further evaluation of SCT may be warranted in some groups of adolescents and young adults infected with HIV
