    In situ study of sigma phase formation in Cr-Co-Ni ternary alloys at 800°C using the long duration experiment facility at Diamond Light Source.

    The new long duration experiment facility on beamline I11 at Diamond Light Source has been used to study the kinetics of sigma phase formation in three Cr-Co-Ni alloys. Diffraction data acquired during in situ exposure at 800°C for 50 d showed progressive increases in the sigma fraction. This was accompanied by changes in the proportions of the other phases, which differed markedly between the alloys studied. These results demonstrate the capabilities of the long duration facility for the study of metallurgical phenomena over periods of months to years, a capability not previously available at a synchrotron source

    Digit-only sauropod pes trackways from China - evidence of swimming or a preservational phenomenon?

    For more than 70 years unusual sauropod trackways have played a pivotal role in debates about the swimming ability of sauropods. Most claims that sauropods could swim have been based on manus-only or manus-dominated trackways. However none of these incomplete trackways has been entirely convincing, and most have proved to be taphonomic artifacts, either undertracks or the result of differential depth of penetration of manus and pes tracks, but otherwise showed the typical pattern of normal walking trackways. Here we report an assemblage of unusual sauropod tracks from the Lower Cretaceous Hekou Group of Gansu Province, northern China, characterized by the preservation of only the pes claw traces, that we interpret as having been left by walking, not buoyant or swimming, individuals. They are interpreted as the result of animals moving on a soft mud-silt substrate, projecting their claws deeply to register their traces on an underlying sand layer where they gained more grip during progression. Other sauropod walking trackways on the same surface with both pes and manus traces preserved, were probably left earlier on relatively firm substrates that predated the deposition of soft mud and silt . Presently, there is no convincing evidence of swimming sauropods from their trackways, which is not to say that sauropods did not swim at all

    Substantial emission reductions from Chinese power plants after the introduction of ultra-low emissions standards

    In 2014, China introduced an ultra-low emissions (ULE) standards policy for renovating coal-fired power-generating units to limit SO2, NOx and particulate matter (PM) emissions to 35, 50 and 10 mg m−3, respectively. The ULE standard policy had ambitious levels (surpassing those of all other countries) and implementation timeline. We estimate emission reductions associated with the ULE policy by constructing a nationwide, unit-level, hourly-frequency emissions dataset using data from a continuous emissions monitoring systems network covering 96–98% of Chinese thermal power capacity during 2014–2017. We find that between 2014 and 2017 China’s annual power emissions of SO2, NOx and PM dropped by 65%, 60% and 72%, respectively. Our estimated emissions using actual monitoring data are 18–92% below other recent estimates. We detail the technologies used to meet the ULE standards and the determinants of compliance, underscoring the importance of ex post evaluation and providing insights for other countries wishing to reduce their power emissions

    Simultaneous determination of natural and synthetic steroid estrogens and their conjugates in aqueous matrices by liquid chromatography / mass spectrometry

    An analytical method for the simultaneous determination of nine free and conjugated steroid estrogens was developed with application to environmental aqueous matrices. Solid phase extraction (SPE) was employed for isolation and concentration, with detection by liquid chromatography/mass spectrometry (LC/MS) using electrospray ionisation (ESI) in the negative mode. Method recoveries for various aqueous matrices (wastewater, lake and drinking water) were determined, recoveries proving to be sample dependent. When spiked at 50 ng/l concentrations in sewage influent, recoveries ranged from 62-89 % with relative standard deviations (RSD) < 8.1 %. In comparison, drinking water spiked at the same concentrations had recoveries between 82-100 % with an RSD < 5%. Ion suppression is a known phenomenon when using ESI; hence its impact on method recovery was elucidated for raw sewage. Both ion suppression from matrix interferences and the extraction procedure has bearing on the overall method recovery. Analysis of municipal raw sewage identified several of the analytes of interest at ng/l concentrations, estriol (E3) being the most abundant.     Distinct Binding and Immunogenic Properties of the Gonococcal Homologue of Meningococcal Factor H Binding Protein

    Neisseria meningitidis is a leading cause of sepsis and meningitis. The bacterium recruits factor H (fH), a negative regulator of the complement system, to its surface via fH binding protein (fHbp), providing a mechanism to avoid complement-mediated killing. fHbp is an important antigen that elicits protective immunity against the meningococcus and has been divided into three different variant groups, V1, V2 and V3, or families A and B. However, immunisation with fHbp V1 does not result in cross-protection against V2 and V3 and vice versa. Furthermore, high affinity binding of fH could impair immune responses against fHbp. Here, we investigate a homologue of fHbp in Neisseria gonorrhoeae, designated as Gonococcal homologue of fHbp (Ghfp) which we show is a promising vaccine candidate for N. meningitidis. We demonstrate that Gfhp is not expressed on the surface of the gonococcus and, despite its high level of identity with fHbp, does not bind fH. Substitution of only two amino acids in Ghfp is sufficient to confer fH binding, while the corresponding residues in V3 fHbp are essential for high affinity fH binding. Furthermore, immune responses against Ghfp recognise V1, V2 and V3 fHbps expressed by a range of clinical isolates, and have serum bactericidal activity against N. meningitidis expressing fHbps from all variant groups

    RELICS: spectroscopy of gravitationally lensed z ≃ 2 reionization-era analogues and implications for C III] detections at z > 6

    Recent observations have revealed the presence of strong C III] emission (EWCIII]>20 Å) in z > 6 galaxies, the origin of which remains unclear. In an effort to understand the nature of these line emitters, we have initiated a survey targeting C III] emission in gravitationally lensed reionization-era analogues identified in Hubble Space Telescope imaging of clusters from the Reionization Lensing Cluster Survey. Here, we report initial results on four galaxies selected to have low stellar masses (2–8 × 107 M⊙) and J125-band flux excesses indicative of intense [O III] + H β emission (EW[OIII]+Hβ = 500–2000 Å), similar to what has been observed at z > 6. We detect C III] emission in three of the four sources, with the C III] EW reaching values seen in the reionization era (EWCIII]≃17−22 Å) in the two sources with the strongest optical line emission (EW[OIII]+Hβ≃2000 Å). We have obtained a Magellan/FIRE (Folded-port InfraRed Echellette) near-infrared spectrum of the strongest C III] emitter in our sample, revealing gas that is both metal poor and highly ionized. Using photoionization models, we are able to simultaneously reproduce the intense C III] and optical line emission for extremely young (2–3 Myr) and metal-poor (0.06–0.08 Z⊙) stellar populations, as would be expected after a substantial upturn in the star formation rate of a low-mass galaxy. The sources in this survey are among the first for which C III] has been used as the primary means of redshift confirmation. We suggest that it should be possible to extend this approach to z > 6 with current facilities, using C III] to measure redshifts of objects with IRAC excesses indicating EW[OIII]+Hβ≃2000 Å, providing a method of spectroscopic confirmation independent of Ly α

    Genome-wide copy number variation study in anorectal malformations

    Anorectal malformations (ARMs, congenital obstruction of the anal opening) are among the most common birth defects requiring surgical treatment (2-5/10 000 live-births) and carry significant chronic morbidity. ARMs present either as isolated or as part of the phenotypic spectrum of some chromosomal abnormalities or monogenic syndromes. The etiology is unknown. To assess the genetic contribution to ARMs, we investigated single-nucleotide polymorphisms and copy number variations (CNVs) at genome-wide scale. A total of 363 Han Chinese sporadic ARM patients and 4006 Han Chinese controls were included. Overall, we detected a 1.3-fold significant excess of rare CNVs in patients. Stratification of patients by presence/absence of other congenital anomalies showed that while syndromic ARM patients carried significantly longer rare duplications than controls (P = 0.049), non-syndromic patients were enriched with both rare deletions and duplications when compared with controls (P = 0.00031). Twelve chromosomal aberrations and 114 rare CNVs were observed in patients but not in 868 controls nor 11 943 healthy individuals from the Database of Genomic Variants. Importantly, these aberrations were observed in isolated ARM patients. Gene-based analysis revealed 79 genes interfered by CNVs in patients only. In particular, we identified a de novo DKK4 duplication. DKK4 is a member of the WNT signaling pathway which is involved in the development of the anorectal region. In mice, Wnt disruption results in ARMs. Our data suggest a role for rare CNVs not only in syndromic but also in isolated ARM patients and provide a list of plausible candidate genes for the disorder.postprin

    Probing and controlling fluorescence blinking of single semiconductor nanoparticles

    In this review we present an overview of the experimental and theoretical development on fluorescence intermittency (blinking) and the roles of electron transfer in semiconductor crystalline nanoparticles. Blinking is a very interesting phenomenon commonly observed in single molecule/particle experiments. Under continuous laser illumination, the fluorescence time trace of these single nanoparticles exhibit random light and dark periods. Since its first observation in the mid-1990s, this intriguing phenomenon has attracted wide attention among researchers from many disciplines. We will first present the historical background of the discovery and the observation of unusual inverse power-law dependence for the waiting time distributions of light and dark periods. Then, we will describe our theoretical modeling efforts to elucidate the causes for the power-law behavior, to probe the roles of electron transfer in blinking, and eventually to control blinking and to achieve complete suppression of the blinking, which is an annoying feature in many applications of quantum dots as light sources and fluorescence labels for biomedical imaging
