    There are over 18.000 ha of irrigated mango in the São Francisco River Valley, and 90% of the fruit produced in this semi-arid region is exported to the european and american markets. With the expansion of the mango areas in the last decade, the intensity of malformation has been increasing, and there are many unresolved questions on the dynamics of the disease in the region, such as the role of the mango bud mite (Aceria mangiferae) or mechanical transmission in the dissemination of the disease.     The Role of Experts across Two Different Arenas in a Deliberative System

    The notion of a “deliberative system” has become central to debates on deliberation. The plea to regard deliberative processes from a system-wide perspective is genuinely innovative and attractive, but little has been done to understand how deliberation in one arena or a separate institution relates to other arenas. This study investigates the role that experts play in public communication in two arenas that have distinct systemic functions. It compares how experts express and justify their opinions on a controversial public policy in legislative public hearings and when they are quoted in the news media. Our findings, based on an empirical case study, revealed that experts played a similar role in different contexts in micro- and macro arenas; and most debate participants appealed to technical knowledge to compel a particular decision. Our analysis concludes by reflecting upon the interconnectivities of the aforementioned arenas; and the systemic approach implications on empirical research

    Genotype-phenotype correlations and BH4 estimated responsiveness in patients with phenylketonuria from Rio de Janeiro, Southeast Brazil

    Background: Genetic heterogeneity and compound heterozygosis give rise to a continuous spectrum of phenylalanine hydroxylase deficiency and metabolic phenotypes in phenylketonuria (PKU). The most used parameters for evaluating phenotype in PKU are pretreatment phenylalanine (Phe) levels, tolerance for dietary Phe, and Phe overloading test. Phenotype can vary from a "classic" (severe) form to mild hyperphenylalaninemia, which does not require dietary treatment. A subset of patients is responsive to treatment by the cofactor tetrahydrobiopterin (BH4 ). Genotypes of PKU patients from Rio de Janeiro, Brazil, were compared to predicted and observed phenotypes. Genotype-based estimations of responsiveness to BH4 were also conducted. Methods: Phenotype was defined by pretreatment Phe levels. A standard prediction system based on arbitrary assigned values was employed to measure genotype-phenotype concordance. Patients were also estimated as BH4 -responders according to the responsiveness previously reported for their mutations and genotypes. Results: A 48.3% concordance rate between genotype-predicted and observed phenotypes was found. When the predicted phenotypes included those reported at the BIOPKU database, the concordance rate reached 77%. A total of 18 genotypes from 30 patients (29.4%) were estimated as of potential or probable BH4 responsiveness. Inconsistencies were observed in genotypic combinations including the common "moderate" mutations p.R261Q, p.V388M, and p.I65T and the mild mutations p.L48S, p.R68S, and p.L249F. Conclusion: The high discordance rate between genotype-predicted and observed metabolic phenotypes in this study seems to be due partially to the high frequency of the so-called "moderate" common mutations, p.R261Q, p.V388M, and p.I65T, which are reported to be associated to erratic or more severe than expected metabolic phenotypes.     Structural mechanical simulation to optimize the sensor arm geometry to be implemented on cranial remodeling orthosis

    For the treatment of moderate and severe cases of deformational plagiocephaly, an asymmetrical deformation of the skull, a cranial remodeling orthosis (CRO) is used. For the development of a new CRO concept, a pressure sensor grid is placed inside the orthosis that will allow the monitoring of excessive pressures and incorrect CRO positioning throughout the treatment. To implement the sensor grid in the CRO, high mobility of the sensor arms structure that joins the several sensors of this grid is required, however, it is intended that this procedure does not damage the printed copper tracks on the sensor structure. In this study, computer simulations were performed to optimize the sensor arm structure geometry, minimizing undesirable mechanical behavior in the sensor structure when subjected to tensile forces and displacements applied during its placement in the CRO. It was observed that the different sensor arm structures geometries have different effects on the mechanical behavior of the sensor structure when subjected to tensile forces and tensile displacement.     Mesenchymal stem/stromal cells : disrupting cell therapy storage and distribution with hypothermic preservation of adipose-derived mesenchymal stromal cells

    Background & Aim: Cell and gene therapies (CGT) have reached new therapeutic targets but have noticeably high prices. Solutions to reduce production costs might be found in CGT storage and transportation since they typically involve cryopreservation, which is a heavily burdened process. Encapsulation at hypothermic temperatures (e.g.,2–8°C) could be a feasible alternative. In this study, we aim to determine the ability of alginate encapsulation to maintain cell viability, identity, and function in the context of MSC-based therapy manufacturing. Methods, Results & Conclusion: Adipose tissue-derived mesenchymal stromal cells (MSC(AT)) expanded using fetal bovine serum (FBS)- (MSC-FBS) or human platelet lysate (HPL)-supplemented mediums (MSC-HPL) were encapsulated in alginate beads (BeadReady™ kits kindly provided by Atelerix) for 30 min, 5 days, and 12 days. After bead release, cell recovery and viability were determined to assess encapsulation performance. MSC identity and functional immunophenotype, MSC tri-lineage differentiation potential, metabolic activity, and hematopoietic support capacity were determined and compared between timepoints. MSC(AT) were able to survive encapsulated for a standard transportation period of 5 days, with recovery values of 56 ± 5% for MSC-FBS and 77 ± 6% for MSC-HPL (which is a negligible drop compared to earlier timepoints). Importantly, MSC function did not suffer from encapsulation, with recovered cells showing robust differentiation potential, expression of immunomodulatory molecules, and hematopoietic support capacity. MSC(AT) encapsulation was proven possible for a remarkable 12 day period. There is currently no solution to completely replace cryopreservation in CGT logistics and supply chain, although encapsulation has shown potential to act as a serious competitor.info:eu-repo/semantics/publishedVersio
