    Chromatin-sensitive cryptic promoters putatively drive expression of alternative protein isoforms in yeast

    Cryptic transcription is widespread and generates a heterogeneous group of RNA molecules of unknown function. To improve our understanding of cryptic transcription, we investigated their transcription start site (TSS) usage, chromatin organization, and posttranscriptional consequences in Saccharomyces cerevisiae We show that TSSs of chromatin-sensitive internal cryptic transcripts retain comparable features of canonical TSSs in terms of DNA sequence, directionality, and chromatin accessibility. We define the 5' and 3' boundaries of cryptic transcripts and show that, contrary to RNA degradation-sensitive ones, they often overlap with the end of the gene, thereby using the canonical polyadenylation site, and associate to polyribosomes. We show that chromatin-sensitive cryptic transcripts can be recognized by ribosomes and may produce truncated polypeptides from downstream, in-frame start codons. Finally, we confirm the presence of the predicted polypeptides by reanalyzing N-terminal proteomic data sets. Our work suggests that a fraction of chromatin-sensitive internal cryptic promoters initiates the transcription of alternative truncated mRNA isoforms. The expression of these chromatin-sensitive isoforms is conserved from yeast to human, expanding the functional consequences of cryptic transcription and proteome complexity

    Spectral functions of the Falicov-Kimball model with electronic ferroelectricity

    We calculate the angular resolved photoemission spectrum of the Falicov-Kimball model with electronic ferroelectricity where dd- and ff-electrons have different hoppings. In mix-valence regimes, the presence of strong scattering processes between dd-ff excitons and a hole, created by emission of an electron, leads to the formation of pseudospin polarons and novel electronic structures with bandwidth scaling with that of dd-ff excitons. Especially, in the two-dimensional case, we find that flat regions exist near the bottom of the quasiparticle band in a wide range of the dd- and ff-level energy difference.Comment: 5 pages, 5 figure

    Use of traditional and complementary medicine as self-care strategies in community health centers

    In China, Community Health Centers (CHCs) are major providers of primary care services, but their potential in empowering patients’ self-management capacity has not been assessed. This study aims to describe self-care practice patterns amongst CHC attendees in urban China. In this cross-sectional quantitative study, 3360 CHC patients from 6 cities within the Pearl Delta Region were sampled using multistage cluster sampling. Thirty-seven per cent had used with over-the-counter Chinese herbal medicines (OTC CHMs) in the past year and majority of respondents found OTC CHMs effective. OTC CHMs were more popular amongst those who needed to pay out of pocket for CHC services. Less than 10% used vitamins and minerals, and those with a lower socioeconomic background have a higher propensity to consume. Although doubts on their usefulness are expressed, their use by the vulnerable population may reflect barriers to access to conventional health care, cultural affinity, or a defense against negative consequences of illnesses. About 25% performed physical exercise, but the prevalence is lower amongst women and older people. Taiji seems to be an alternative for these populations with promising effectiveness, but overall only 6% of CHC attendees participated. These results suggest that CHCs should start initiatives in fostering appropriate use of OTC CHM, vitamins, and minerals. Engaging community pharmacists in guiding safe and effective use of OTC CHM amongst the uninsured is essential given their low accessibility to CHC services. Prescription of Taiji instead of physical exercises to women and older people could be more culturally appropriate, and the possibility of including this as part of the CHC services worth further exploration

    Intense high contrast femtosecond K-shell x-ray source from laser-driven Ar clusters

    Bright Ar K-shell x-ray with very little background has been generated using an Ar clustering gas jet target irradiated with an 800 mJ, 30 fs ultra-high contrast laser, with the measured flux of 1.1 x 10^4 photons/mrad^2/pulse. This intense x-ray source critically depends on the laser contrast and the laser energy and the optimization of this source with interaction is addressed. Electron driven by laser electric field directly via nonlinear resonant is proved in simulation, resulting in effective electron heating and the enhancement of x-ray emission. The x-ray pulse duration is demonstrated to be only 10 fs, as well as a source size of 20 um, posing great potential application for single-shot ultrafast x-ray imaging.Comment: 13 pages, 5 figure

    Measurement of Two-Qubit States by a Two-Island Single Electron Transistor

    We solve the master equations of two charged qubits measured by a single-electron transistor (SET) consisted of two islands. We show that in the sequential tunneling regime the SET current can be used for reading out results of quantum calculations and providing evidences of two-qubit entanglement, especially when the interaction between the two qubits is weak

    Magnetization steps in Zn_(1-x)Mn_xO: Four largest exchange constants and single-ion anisotropy

    Magnetization steps (MST's) from Mn pairs in several single crystals of Zn_(1-x)Mn_xO (0.0056<=x<=0.030, and in one powder (x=0.029), were observed. The largest two exchange constants, J1/kB=-18.2+/-0.5K and J1'/kB=-24.3+/-0.6K, were obtained from large peaks in the differential susceptibility, dM/dH, measured in pulsed magnetic fields, H, up to 500 kOe. These two largest J's are associated with the two inequivalent classes of nearest neighbors (NN's) in the wurtzite structure. The 29% difference between J1 and J1' is substantially larger than 13% in CdS:Mn, and 15% in CdSe:Mn. The pulsed-field data also indicate that, despite the direct contact between the samples and a superfluid-helium bath, substantial departures from thermal equilibrium occurred during the 7.4 ms pulse. The third- and fourth-largest J's were determined from the magnetization M at 20 mK, measured in dc magnetic fields H up to 90 kOe. Both field orientations H||c and H||[10-10] were studied. (The [10-10] direction is perpendicular to the c-axis, [0001].) By definition, neighbors which are not NN's are distant neighbors (DN's). The largest DN exchange constant (third-largest overall), has the value J/kB=-0.543+/-0.005K, and is associated with the DN at r=c. Because this is not the closest DN, this result implies that the J's do not decrease monotonically with the distance r. The second-largest DN exchange constant (fourth-largest overall), has the value J/kB=-0.080 K. It is associated with one of the two classes of neighbors that have a coordination number z=12, but the evidence is insufficient for a definite unique choice. The dependence of M on the direction of H gives D/kB=-0.039+/-0.008K, in fair agreement with -0.031 K from earlier EPR work.Comment: 12 pages, 10 figures. Submitted to PR

    Does accelerating universe indicates Brans-Dicke theory

    The evolution of universe in Brans-Dicke (BD) theory is discussed in this paper. Considering a parameterized scenario for BD scalar field ϕ=ϕ0aα\phi=\phi_{0}a^{\alpha} which plays the role of gravitational "constant" GG, we apply the Markov Chain Monte Carlo method to investigate a global constraints on BD theory with a self-interacting potential according to the current observational data: Union2 dataset of type supernovae Ia (SNIa), high-redshift Gamma-Ray Bursts (GRBs) data, observational Hubble data (OHD), the cluster X-ray gas mass fraction, the baryon acoustic oscillation (BAO), and the cosmic microwave background (CMB) data. It is shown that an expanded universe from deceleration to acceleration is given in this theory, and the constraint results of dimensionless matter density Ω0m\Omega_{0m} and parameter α\alpha are, Ω0m=0.2860.0390.047+0.037+0.050\Omega_{0m}=0.286^{+0.037+0.050}_{-0.039-0.047} and α=0.00460.01710.0206+0.0149+0.0171\alpha=0.0046^{+0.0149+0.0171}_{-0.0171-0.0206} which is consistent with the result of current experiment exploration, α0.132124\mid\alpha\mid \leq 0.132124. In addition, we use the geometrical diagnostic method, jerk parameter jj, to distinguish the BD theory and cosmological constant model in Einstein's theory of general relativity.Comment: 16 pages, 3 figure
