    Identification of novel post-transcriptional features in olfactory receptor family mRNAs.

    Olfactory receptor (Olfr) genes comprise the largest gene family in mice. Despite their importance in olfaction, how most Olfr mRNAs are regulated remains unexplored. Using RNA-seq analysis coupled with analysis of pre-existing databases, we found that Olfr mRNAs have several atypical features suggesting that post-transcriptional regulation impacts their expression. First, Olfr mRNAs, as a group, have dramatically higher average AU-content and lower predicted secondary structure than do control mRNAs. Second, Olfr mRNAs have a higher density of AU-rich elements (AREs) in their 3'UTR and upstream open reading frames (uORFs) in their 5 UTR than do control mRNAs. Third, Olfr mRNAs have shorter 3' UTR regions and with fewer predicted miRNA-binding sites. All of these novel properties correlated with higher Olfr expression. We also identified striking differences in the post-transcriptional features of the mRNAs from the two major classes of Olfr genes, a finding consistent with their independent evolutionary origin. Together, our results suggest that the Olfr gene family has encountered unusual selective forces in neural cells that have driven them to acquire unique post-transcriptional regulatory features. In support of this possibility, we found that while Olfr mRNAs are degraded by a deadenylation-dependent mechanism, they are largely protected from this decay in neural lineage cells

    Sepsis biomarkers in unselected patients on admission to intensive or high-dependency care

    Although many sepsis biomarkers have shown promise in selected patient groups, only C-reactive protein and procalcitonin (PCT) have entered clinical practice. The aim of this study was to evaluate three promising novel sepsis biomarkers in unselected patients at admission to intensive care. We assessed the performance of pancreatic stone protein (PSP), soluble CD25 (sCD25) and heparin binding protein (HBP) in distinguishing patients with sepsis from those with a non-infective systemic inflammatory response and the ability of these markers to indicate severity of illness. METHODS: Plasma levels of the biomarkers, PCT and selected inflammatory cytokines were measured in samples taken from 219 patients during the first six hours of admission to intensive or high dependency care. Patients with a systemic inflammatory response were categorized as having sepsis or a non-infective aetiology, with or without markers of severity, using standard diagnostic criteria. RESULTS: Both PSP and sCD25 performed well as biomarkers of sepsis irrespective of severity of illness. For both markers the area under the receiver operating curve (AUC) was greater than 0.9; PSP 0.927 (0.887 to 0.968) and sCD25 0.902 (0.854 to 0.949). Procalcitonin and IL6 also performed well as markers of sepsis whilst in this intensive care unit (ICU) population, HBP did not: PCT 0.840 (0.778 to 0.901), IL6 0.805 (0.739 to 0.870) and HBP 0.607 (0.519 to 0.694). Levels of both PSP and PCT reflected severity of illness and both markers performed well in differentiating patients with severe sepsis from severely ill patients with a non-infective systemic inflammatory response: AUCs 0.955 (0.909 to 1) and 0.837 (0.732 to 0.941) respectively. Although levels of sCD25 did not correlate with severity, the addition of sCD25 to either PCT or PSP in a multivariate model improved the diagnostic accuracy of either marker alone. CONCLUSIONS: PSP and sCD25 perform well as sepsis biomarkers in patients with suspected sepsis at the time of admission to intensive or high dependency care. These markers warrant further assessment of their prognostic value. Whereas previously published data indicate HBP has clinical utility in the emergency department, it did not perform well in an intensive-care population

    A time-course study of early establishment stages of parasitic angiosperm Striga asiatica on susceptible sorghum roots

    A time-course study of the early establishment stages of Striga asiatica was carried out on a susceptible sorghum hybrid, CSH 1, using polyethylene bags and whole-root clearing and staining techniques. Preconditioned Striga seeds were applied to different aged segments of primary root but the results did not differ for these different aged segments. Most of the Striga seeds (63%) germinated within 24 h of inoculation on the host roots. The attachment of Striga radicles to host root was rapid and it occurred between 36 and 48 h after inoculation. Only 9% of the germinated Striga seeds attached to the host root but 65% of these attachments successfully penetrated through the epidermis and entered the host cortex within 72 h. Penetration through the cortical cells was difficult; only 17% of attachments were able to reach the endodermis. Penetration took from 12 to 43 h after the first appearance of haustorial cells in the cortex; a total of 84 to 120 h after inoculation on the host root. Penetration through the endodermis and establishment on the host stele was relatively easier, as most of the haustoria reaching the endodermis were able to establish on the host stele. But this is a slow process taking a minimum of 24 h, and a maximum of 60 h after first contact of haustorial cells with the endodermis. The minimum time taken from inoculation of ungerminated Striga seed on the host root to establishment is about 108 h. The results are discussed in relation to published reports on other parasitic species such as Agalinis purpure

    Inheritance of Striga seed-germination stimulant from S. asiatica seed collected at ICRISAT Centre was investigated in sorghum using 3 low-stimulant-producing (resistant) cultivars (555, Framida and SNR6496) and 6 high-stimulant-producing (susceptible) cultivars (Swarna, NJ2006, IS508, 168, 148 and M35-1). From a study of the parents, F1, F2 and backcross F2 seedling progenies of 9 crosses between resistant and susceptible parents, it was concluded that low stimulant production is under the control of a single recessive allele. It was not determined whether this allele was the same in the 3 resistant parent

    A New Rapid and Sensitive Stability-Indicating UPLC Assay Method for Tolterodine Tartrate: Application in Pharmaceuticals, Human Plasma and Urine Samples

    A new rapid, simple, sensitive, selective and accurate reversed-phase stability-indicating Ultra Performance Liquid Chromatography (RP-UPLC) technique was developed for the assay of Tolterodine Tartrate in pharmaceutical dosage form, human plasma and urine samples. The developed UPLC method is superior in technology to conventional HPLC with respect to speed, solvent consumption, resolution and cost of analysis. Chromatographic run time was 6 min in reversed-phase mode and ultraviolet detection was carried out at 220 nm for quantification. Efficient separation was achieved for all the degradants of Tolterodine Tartrate on BEH C18 sub-2-μm Acquity UPLC column using Trifluoroacetic acid and acetonitrile as organic solvent in a linear gradient program. The active pharmaceutical ingredient was extracted from tablet dosage form using a mixture of acetonitrile and water as diluent. The calibration graphs were linear and the method showed excellent recoveries for bulk and tablet dosage form. The test solution was found to be stable for 40 days when stored in the refrigerator between 2 and 8 °C. The developed UPLC method was validated and meets the requirements delineated by the International Conference on Harmonization (ICH) guidelines with respect to linearity, accuracy, precision, specificity and robustness. The intra-day and inter-day variation was found be less than 1%. The method was reproducible and selective for the estimation of Tolterodine Tartrate. Because the method could effectively separate the drug from its degradation products, it can be employed as a stability-indicating one

    Safety and disease monitoring biomarkers in Duchenne muscular dystrophy: results from a Phase II trial

    Aim: Evaluate the utility of glutamate dehydrogenase (GLDH) and cardiac troponin I as safety biomarkers, and creatine kinase and muscle injury panel as muscle health biomarkers in Duchenne muscular dystrophy. Patients & methods: Data were collected during a Phase II trial of domagrozumab. Results: GLDH was a more specific biomarker for liver injury than alanine aminotransferase. Cardiac troponin I elevations were variable and not sustained, limiting its applicability as a biomarker. Muscle injury panel biomarkers were no more informative than creatine kinase as a muscle health biomarker. Conclusion: Results support the use of GLDH as a specific biomarker for liver injury in patients with Duchenne muscular dystrophy

    Breeding Sorghum with Resistance to Striga asiatica (L) Kuntze at ICRISAT Center

    Breeding for Striga resistance at ICRISAT Center, near Hyderabad, India, has the twin objectives of identifying sorghum source lines resistant to Striga asiatica and transfering the resistance to agronomically elite lines; 14 000 sorghum germplasm lines have been screened in the laboratory for their stimulant production and 640 low-stimulant lines have been identified. Studies on the genetics of stimulant production have indicated a preponderance of additive over nonadditive genetic variance. Selection for field resistance among the derivatives from Striga-resistant source lines x adapted crosses resulted in a higher proportion of field resistants in the low-stimulant than in the high-stimulant derivatives. In multilocation testing of the source lines, field reaction indicated that the best available low susceptible lines are N-13, 555, IS-4202, IS-7471, and IS-9985. The technique of growing test plants of the host in shallow seed pans in a soil medium has been found useful in differentiating resistant from susceptible host plants. An improved, three-stage system of screening for field resistance to Striga is described. Initial studies on Striga collected from five locations in India and four sorghum varieties indicated significant strain x variety interactions and SRN-4882B gave differential reaction. Intensive studies on host-parasite relationships, environmental interactions influencing Striga, screening methodology, guidelines to manage Striga sick fields, and surveys to understand species and race complexes have been projected as some of the priority areas of Striga research
