    DNA Vaccines Encoding Antigen Targeted to MHC Class II Induce Influenza-Specific CD8+ T Cell Responses, Enabling Faster Resolution of Influenza Disease

    Current influenza vaccines are effective but imperfect, failing to cover against emerging strains of virus and requiring seasonal administration to protect against new strains. A key step to improving influenza vaccines is to improve our understanding of vaccine induced protection. Whilst it is clear that antibodies play a protective role, vaccine induced CD8+ T cells can improve protection. To further explore the role of CD8+ T cells we used a DNA vaccine that encodes antigen dimerised to an immune cell targeting module. Immunising CB6F1 mice with the DNA vaccine in a heterologous prime boost regime with the seasonal protein vaccine improved the resolution of influenza disease compared to protein alone. This improved disease resolution was dependent on CD8+ T cells. However, DNA vaccine regimes that induced CD8+ T cells alone were not protective and did not boost the protection provided by protein. The MHC targeting module used was an anti-I-Ed single chain antibody specific to the BALB/c strain of mice. To test the role of MHC targeting we compared the response between BALB/c, C57BL/6 mice and an F1 cross of the two strains (CB6F1). BALB/c mice were protected, C57BL/6 were not and the F1 had an intermediate phenotype; showing that the targeting of antigen is important in the response. Based on these findings, and in agreement with other studies using different vaccines, we conclude that in addition to antibody, inducing a protective CD8 response is important in future influenza vaccines

    Experimental Identification of the Kink Instability as a Poloidal Flux Amplification Mechanism for Coaxial Gun Spheromak Formation

    The magnetohydrodynamic kink instability is observed and identified experimentally as a poloidal flux amplification mechanism for coaxial gun spheromak formation. Plasmas in this experiment fall into three distinct regimes which depend on the peak gun current to magnetic flux ratio, with (I) low values resulting in a straight plasma column with helical magnetic field, (II) intermediate values leading to kinking of the column axis, and (III) high values leading immediately to a detached plasma. Onset of column kinking agrees quantitatively with the Kruskal-Shafranov limit, and the kink acts as a dynamo which converts toroidal to poloidal flux. Regime II clearly leads to both poloidal flux amplification and the development of a spheromak configuration.Comment: accepted for publication in Physical Review Letter

    Acoustic Emission Multi-Parameter Analyzer—AEMPA

    A multi-parameter measurement system was designed and built to process signals from acoustic emission (AE) transducers in real time. The system makes selected measurements of each detected AE event as it occurs and stores 23 measured parameters which describe that event in digital form on a standard diskette with IBM format. Measurements recorded on the diskette include information on the shape, magnitude, and frequency content of each AE burst, its timing with respect to location on the specimen under test and its timing with respect to load conditions, pressure, test time, and event count. Over 8000 AE events can be stored on a single diskette at a maximum instantaneous rate of 6000 events/sec and a maximum average rate of 1000 events/sec. Two floppy disk units are included in the system so one can be operational while the diskette is being changed in the other. After a specimen test, the data are transmitted to a remote minicomputer with a standard RS232 interface. Rapid analysis and display of the data by the computer has been demonstrated using up to 8 of the AE parameters at one time in a pattern recognition routine

    The uniting of Europe and the foundation of EU studies: revisiting the neofunctionalism of Ernst B. Haas

    This article suggests that the neofunctionalist theoretical legacy left by Ernst B. Haas is somewhat richer and more prescient than many contemporary discussants allow. The article develops an argument for routine and detailed re-reading of the corpus of neofunctionalist work (and that of Haas in particular), not only to disabuse contemporary students and scholars of the normally static and stylized reading that discussion of the theory provokes, but also to suggest that the conceptual repertoire of neofunctionalism is able to speak directly to current EU studies and comparative regionalism. Neofunctionalism is situated in its social scientific context before the theory's supposed erroneous reliance on the concept of 'spillover' is discussed critically. A case is then made for viewing Haas's neofunctionalism as a dynamic theory that not only corresponded to established social scientific norms, but did so in ways that were consistent with disciplinary openness and pluralism

    Water-ice driven activity on Main-Belt Comet P/2010 A2 (LINEAR) ?

    The dust ejecta of Main-Belt Comet P/2010 A2 (LINEAR) have been observed with several telescopes at the at the Observatorio del Roque de los Muchachos on La Palma, Spain. Application of an inverse dust tail Monte Carlo method to the images of the dust ejecta from the object indicates that a sustained, likely water-ice driven, activity over some eight months is the mechanism responsible for the formation of the observed tail. The total amount of dust released is estimated to be 5E7 kg, which represents about 0.3% of the nucleus mass. While the event could have been triggered by a collision, this cannot be decided from the currently available data.Comment: Accepted for ApJ Letter
