    Biocompatibility of the vital dye Acid Violet-17 on retinal pigment epithelial cells

    Purpose: To examine the viability and differentiation of retinal pigment epithelial (RPE) cells after exposure to the vital dye Acid Violet-17 (AV-17). Methods: Bovine RPE cells were incubated with AV-17 (0.0625-0.5 mg/mL) for 30 seconds or 5 minutes. Viability was determined by live/dead staining, cleaved CASP3 immunostainings, and MTT test. Actin cytoskeleton was visualized by Alexa 488-phalloidin. Immunocytochemistry was performed to determine the levels of ZO-1, CTNNB1, and KRT19. Results: Exposure to AV-17 at the concentrations of 0.25-0.5 mg/mL resulted in a dose-dependent decrease in viability, the loss of ZO-1 from tight junctions, translocation of CTNNB1 into the cytoplasm and nucleus, disarrangement of the actin cytoskeleton, and a slight increase in KRT19. Conclusion: AV-17 at a concentration. 0.125 mg/mL is likely to be well tolerated by the RPE cells, whereas the concentrations from 0.25 mg/mL onward can reduce viability and induce dedifferentiation particularly after long-term exposure

    Dynamical coupled-channel model of meson production reactions in the nucleon resonance region

    A dynamical coupled-channel model is presented for investigating the nucleon resonances in the meson production reactions induced by pions and photons. The model is based on an energy-independent Hamiltonian which is derived from a set of Lagrangians by using a unitary transformation method. By applying the projection operator techniques,we derive a set of coupled-channel equations which satisfy the unitarity conditions within the channel space spanned by the considered two-particle meson-baryon states and the three-particle ππN\pi\pi N state. We present and explain in detail a numerical method based on a spline-function expansion for solving the resulting coupled-channel equations which contain logarithmically divergent one-particle-exchange driving terms resulted from the ππN\pi\pi N unitarity cut. We show that this driving term can generate rapidly varying structure in the reaction amplitudes associated with the unstable particle channels. It also has large effects in determining the two-pion production cross sections. Our results indicate that cautions must be taken to interpret the NN^* parameters extracted from using models which do not include ππN\pi\pi N cut effects.Comment: 73 pages, 20 figure

Please click on the hyperlink in Additional Links field.BACKGROUND: Diabetes mellitus, as well as subsequent ocular complications such as cystoid macular edema (CME), are of fundametal socio-economic relevance. Therefore, we evaluated the influence of internal limiting membrane (ILM) removal on longterm morphological and functional outcome in patients with diabetes mellitus (DM) type 2 and chronic CME without evident vitreomacular traction. METHOD: Forty eyes with attached posterior hyaloid were included in this prospective trial and randomized intraoperatively. Prior focal (n = 31) or panretinal (n = 25) laser coagulation was permitted. Group I (n = 19 patients) underwent surgical induction of posterior vitreous detachment (PVD), group II (n = 20 patients) PVD and removal of the ILM. Eleven patients with detached posterior hyaloid (group III) were not randomized, and ILM removal was performed. One eye had to be excluded from further analysis. Examinations included ETDRS best-corrected visual acuity (BCVA), fluorescein angiography (FLA) and OCT at baseline, 3 and 6 months postoperatively. Main outcome measure was BCVA at 6 months, secondary was foveal thickness. RESULTS: Mean BCVA over 6 months remained unchanged in 85% of patients of group II, and decreased in 53% of patients of group I. Results were not statistically significant different [group I: mean decrease log MAR 95% CI (0.06; 0.32), group II: (-0.02; 0.11)]. OCT revealed a significantly greater reduction of foveal thickness following PVD with ILM removal [group I: mean change: 95% CI (-208.95 μm; -78.05 μm), group II: (-80.90 μm: +59.17 μm)]. CONCLUSION: Vitrectomy, PVD with or without ILM removal does not improve vision in patients with DM type 2 and cystoid diabetic macular edema without evident vitreoretinal traction. ILM delamination shows improved morphological results, and appears to be beneficial in eyes with preexisting PVD

    Inwieweit Verbitterung mit Suizidgedanken assoziiert ist, wurde bislang nur selten untersucht. Darüber hinaus stellt sich die Frage, ob Weisheit bzw. Weisheitskomponenten den Zusammenhang zwischen Verbitterung und Suizidgedan­ken moderieren, Weisheit also als ein Resilienzfaktor gegenüber Suizidgedanken zu verstehen ist. Insgesamt nahmen 311 Proband*innen (n\it n = 179, 57,6% weiblich; Alter: Mittelwert = 32,63 Jahre, SD = 14,06) an einer internetbasierten Fragebogenstudie teil. Es zeigte sich sowohl, dass Verbitte­rung und Suizidgedanken eng assoziiert sind, als auch, dass dieser Zusammenhang durch die Weisheitskomponente Humor moderiert wird. In Bezug auf weitere Weisheitskomponenten fanden sich keine erwartungskonformen Zusammenhänge. Die Ergebnisse unterstützen aktuelle Befunde zur Bedeutung von Humor als Resilienzfaktor gegenüber suizidalem Erleben.The extent to which embitterment is associated with suicide ideation has only rarely been investigated. Moreover, it is unclear whether wisdom or wisdom components may moderate the association between embitterment and suicide ideation. A total of 311 participants (n\it n = 179, 57.6% women; age: mean = 32.63 years, SD = 14.06) took part in an online survey. Results revealed a close positive association between embitterment and suicide ideation. Furthermore, this association was moderated by the wisdom component humor. With regard to other wisdom components, no associations were found that were in line with expectations. The results support current findings on the importance of humor as a resilience factor against suicide ideation

    Metagenomic potential for and diversity of N-cycle driving microorganisms in the Bothnian Sea sediment

    The biological nitrogen cycle is driven by a plethora of reactions transforming nitrogen compounds between various redox states. Here, we investigated the metagenomic potential for nitrogen cycle of the in situ microbial community in an oligotrophic, brackish environment of the Bothnian Sea sediment. Total DNA from three sediment depths was isolated and sequenced. The characterization of the total community was performed based on 16S rRNA gene inventory using SILVA database as reference. The diversity of diagnostic functional genes coding for nitrate reductases (napA;narG), nitrite:nitrate oxidoreductase (nxrA), nitrite reductases (nirK;nirS;nrfA), nitric oxide reductase (nor), nitrous oxide reductase (nosZ), hydrazine synthase (hzsA), ammonia monooxygenase (amoA), hydroxylamine oxidoreductase (hao), and nitrogenase (nifH) was analyzed by blastx against curated reference databases. In addition, Polymerase chain reaction (PCR)-based amplification was performed on the hzsA gene of anammox bacteria. Our results reveal high genomic potential for full denitrification to N2, but minor importance of anaerobic ammonium oxidation and dissimilatory nitrite reduction to ammonium. Genomic potential for aerobic ammonia oxidation was dominated by Thaumarchaeota. A higher diversity of anammox bacteria was detected in metagenomes than with PCR-based technique. The results reveal the importance of various N-cycle driving processes and highlight the advantage of metagenomics in detection of novel microbial key players

    Объект исследования – молекулы типа ассиметричного волчка C2D4 и H2MS (M = 32, 33 и 34). Спектры были зарегистрированы на Фурье-спектрометрах Bruker IFS120HR и Bruker IFS 125HR. В работе исследованы спектры молекулы C2D4 в ИК диапазоне 600-1150 см-1 и спектры молекулы H2MS (M = 32, 33 и 34) в ИК диапазонах 5650-6660 см-1 и 4500-5000 см-1. В результате были улучшены параметры основного состояния молекулы C2D4. Исследована вращательная структура четырех колебательных состояний (для состояния (v4=1) структура определена впервые). Проведено исследование второй гексады молекулы H2MS (M = 32, 33 и 34) и полосы 4?2 молекул H232S и H234S. Определены интенсивности переходов полосы 4?2 молекулы H232S, и в результате определены параметры эффективного дипольного момента.The object of research is molecules of the asymmetric top type, C2D4 and H2MS (M = 32, 33 and 34). Spectra were recorded with a FT-spectrometers Bruker IFS120HR and Bruker IFS 125HR. We studied spectra of the C2D4 molecule in the IR range of 600-1150 cm-1, spectra of the H2MS molecule (M = 32, 33 and 34) in the IR range of 5650-6660 cm-1 and 4500-5000 cm-1. As a result, parameters of the ground state of the C2D4 molecule were improved. Study of the second hexade of H2MS (M = 32, 33 and 34) and the 4?2 band of the H232S and H234S molecules was fulfilled. The intensities of transitions of the 4?2 band of the H232S molecule were determined and, as a result, parameters of the effective dipole moment were determined
