    Bovine Mastitis Resistance: Novel Quantitative Trait Loci and the Role of Bovine Mammary Epithelial Cells

    Bovine mastitis, or inflammation of the mammary gland, has substantial economic and animal welfare implications. A genetic basis for mastitis resistance traits is recognized and can be used to guide selective breeding programs. The discovery of regions of the genome associated with mastitis resistance, and knowledge of the underlying molecular mechanisms responsible, can facilitate development of efficient mastitis control and therapeutic strategies. The objectives of this dissertation research were to identify sites of genetic variation associated with mastitis resistance, and to define the contributions of the milk-secreting epithelial cells to mammary gland immune responses and mastitis resistance. Twenty seven regions of the bovine genome potentially involved in mastitis resistance were identified in Holstein dairy cattle. Additionally, this research demonstrates a role of bovine mammary epithelial cells in mastitis resistance, and provides guidance for the use of an in vitro model for mastitis studies. Primary bovine mammary epithelial cells from mastitis-resistant cows have differential expression of 42 inflammatory genes compared with cells from mastitis-susceptible cows, highlighting the importance of epithelial cells in mastitis resistance. Bovine mammary epithelial cells display both similarities and differences in pro-inflammatory gene expression compared to fibroblasts, and their expression of inflammatory genes is influenced by administration of the enzyme phospholipase A2. The growth potential of milk-derived bovine mammary epithelial cells in vitro can be extended, facilitating their use in mastitis studies, by transfection with a viral protein.     By means of a numerical analysis using a non-Abelian symmetry realization of the density matrix renormalization group, we study the behavior of vector chirality correlations in isotropic frustrated chains of spin S=1 and S=1/2, subject to a strong external magnetic field. It is shown that the field induces a phase with spontaneously broken chiral symmetry, in line with earlier theoretical predictions. Phosphorus deficiency limited grassland production in Ireland and this was corrected by chemical fertiliser use in the 1960s and 1970s. The increased inputs of fertilisers led to increased intensification of grassland with a doubling of grass yield and of grazing animal numbers, from about 3 million to over 6 million livestock units. There is little information on relative contribution of increased chemical fertiliser use compared to increased grazing animal numbers on phosphorus loss to water.     Agriculture, meteorology and water quality in Ireland: a regional evaluation of pressures and pathways of nutrient loss to water

    peer-reviewedThe main environmental impact of Irish agriculture on surface and ground water quality is the potential transfer of nutrients to water. Soil water dynamics mediate the transport of nutrients to water, and these dynamics in turn depend on agro-meteorological conditions, which show large variations between regions, seasons and years. In this paper we quantify and map the spatio-temporal variability of agro-meteorological factors that control nutrient pressures and pathways of nutrient loss. Subsequently, we evaluate their impact on the water quality of Irish rivers. For nitrogen, pressure and pathways factors coincide in eastern and southern areas, which is reflected in higher nitrate levels of the rivers in these regions. For phosphorus, pathway factors are most pronounced in north-western parts of the country. In south-eastern parts, high pressure factors result in reduced biological water quality. These regional differences require that farm practices be customised to reflect the local risk of nutrient loss to water. Where pathways for phosphorus loss are present almost year-round—as is the case in most of the north-western part of the country—build-up of pressures should be prevented, or ameliorated where already high. In south-eastern areas, spatio-temporal coincidence of nutrient pressures and pathways should be prevented, which poses challenges to grassland management

    Based on the published data of pillow lava-hosted mineralized veins, this study compares post-magmatic fracturing, fluid flow, and secondary mineralization processes in the Troodos and Izu–Bonin supra-subduction zone (SSZ) and discusses the crucial factors for the development of distinct vein types. Thin section and cathodoluminescence petrography, Raman spectroscopy, fluid inclusion microthermometry, and trace element and isotope (87Sr/86Sr, δ18O, δ13C, Δ47) geochemistry indicate that most veins consist of calcite that precipitated from pristine to slightly modified seawater at temperatures  15 Myr after subduction initiation. Therefore, high-T mineralization (calcite, quartz, analcime) up to 230 °C is restricted to the Troodos SSZ

    Genetic sequencing using 16s rRNA for pathogen identification in retropharyngeal lymph nodes from wild elk

    Cervical region lymph nodes collected by hunters from 43 wild hunter-harvested elk (Cervus elaphus) in Utah were submitted to the Utah Veterinary Diagnostic Laboratory during fall 2009. We evaluated these lymph nodes as specimens for identification of bacterial pathogens using 16S rRNA genetic sequencing. Thirty-seven bacterial species were identified; each was found in 2 to 30 individual elk. Many common ruminant livestock pathogens were identified in elk; pathogens previously reported in elk were Pasteurella multocida and Streptococcus spp. Cervical region lymph nodes harvested from wild ruminants appear to be acceptable samples for genetic sequencing of bacteria

    Possible observation of phase separation near a quantum phase transition in doubly connected ultrathin superconducting cylinders of aluminum

    The kinetic energy of superconducting electrons in an ultrathin, doubly connected superconducting cylinder, determined by the applied flux, increases as the cylinder diameter decreases, leading to a destructive regime around half-flux quanta and a superconductor to normal metal quantum phase transition (QPT). Regular step-like features in resistance vs. temperature curves taken at fixed flux values were observed near the QPT in ultrathin Al cylinders. It is proposed that these features are most likely resulted from a phase separation near the QPT in which normal regions nucleate in a homogeneous superconducting cylinder.Comment: 4 pages, 4 figures, to appear in Phys.     Testing the Efficacy of OurSpace, a Brief, Group Dynamics-Based Physical Activity Intervention: A Randomized Controlled Trial

However, few interventions have provided effective means to enhance social support through these platforms. Face-to-face programs that use group dynamics-based principles of behavior change have been shown to be highly effective in enhancing social support through promoting group cohesion and PA, but to date, no studies have examined their effects in Web-based programs. Objective: The aim was to explore proof of concept and test the efficacy of a brief, online group dynamics-based intervention on PA in a controlled experiment. We expected that the impact of the intervention on PA would be moderated by perceptions of cohesion and the partner's degree of presence in the online media. Methods: Participants (n=135) were randomized into same-sex dyads and randomly assigned to one of four experimental conditions: standard social support (standard), group dynamics-based-high presence, group dynamics-based-low presence, or individual control. Participants performed two sets of planking exercises (pre-post). Between sets, participants in partnered conditions interacted with a virtual partner using either a standard social support app or a group dynamics-based app (group dynamics-based-low presence and group dynamics-based-high presence), the latter of which they participated in a series of online team-building exercises. Individual participants were given an equivalent rest period between sets. To increase presence during the second set, participants in the group dynamics-based-high presence group saw a live video stream of their partner exercising. Perceptions of cohesion were measured using a modified PA Group Environment Questionnaire. Physical activity was calculated as the time persisted during set 2 after controlling for persistence in set 1.Results: Perceptions of cohesion were higher in the group dynamics-based-low presence (overall mean 5.81, SD 1.04) condition compared to the standard (overall mean 5.04, SD 0.81) conditions (P=.006), but did not differ between group dynamics-based-low presence and group dynamics-based-high presence (overall mean 5.42, SD 1.07) conditions (P=.25). Physical activity was higher in the high presence condition (mean 64.48, SD 20.19, P=.01) than all other conditions (mean 53.3, SD 17.35).Conclusions: A brief, online group dynamics-based intervention may be an effective method of improving group cohesion in virtual PA groups.
