    Manipulating transgenes using a chromosome vector

    Recent technological advances have enabled us to visualize the organization and dynamics of local chromatin structures; however, the comprehensive mechanisms by which chromatin organization modulates gene regulation are poorly understood. We designed a human artificial chromosome vector that allowed manipulation of transgenes using a method for delivering chromatin architectures into different cell lines from human to fish. This methodology enabled analysis of de novo construction, epigenetic maintenance and changes in the chromatin architecture of specific genes. Expressive and repressive architectures of human STAT3 were established from naked DNA in mouse embryonic stem cells and CHO cells, respectively. Delivery of STAT3 within repressive architecture to embryonic stem cells resulted in STAT3 activation, accompanied by changes in DNA methylation. This technology for manipulating a single gene with a specific chromatin architecture could be utilized in applied biology, including stem cell science and regeneration medicine

    Factors associated with a measles outbreak in children admitted at Mahosot Hospital, Vientiane, Laos

    <p>Abstract</p> <p>Background</p> <p>In 2002 and 2003 there were large outbreaks of measles in many provinces of Laos, including in Vientiane. We therefore conducted a study to determine risk factors associated with measles amongst children admitted at Mahosot Hospital, Vientiane.</p> <p>Methods</p> <p>A retrospective case-control study was conducted in 50 children with clinical measles who were matched by age and sex with 50 healthy children (who had never had a febrile rash) living in the same villages as the cases.</p> <p>Results</p> <p>The proportion of children with complete immunizations was significantly lower in the group with clinical measles compared to the controls [13/50 (26%) vs 34/50 (68%), <it>P </it>< 0.001). The percentage of children who had received measles vaccine at 9ā€“23 months of age was significantly lower in the group with clinical measles compared to the healthy controls [12/50 (24%) vs 24/50 (48%), <it>P </it>= 0.01). The family educational and socio-economic status did not differ significantly (<it>P </it>> 0.05) between cases and controls.</p> <p>Conclusion</p> <p>These results emphasize the importance of intensification of measles immunization coverage in Laos. The strengthening of campaigns with large, widespread high second dose coverage is likely to be a key measure to prevent further measles outbreaks in Laos (192 words).</p

    Origin of Middle-Infrared Peaks in Cerium Compounds

    We have demonstrated that the middle-infrared (mid-IR) peaks in the optical conductivity spectra of CeX3X_3 (XX = Pd, Sn, In) can be explained by first-principle band structure calculation with the spin-orbit interaction. The mid-IR peak shapes in these materials are not identical to one another: CePd3_3, CeSn3_3, and CeIn3_3 have a triple-peak structure, double-peak structure and broad single-peak structure, respectively. These peaks can be theoretically explained by the optical transition from the occupied state to the spin-orbit splitted Ce 4f4f state. This result indicates that the mid-IR peaks originate from the simple band picture with the Ce 4f4f state near the Fermi level, not from the conventional cf hybridization gap based on the periodic Anderson model.Comment: 5 pages, 6 figures.     Among patients who visited our hospital with a chief complaint of disorders involving the oromandibular region, with suspected carotid artery calcification based on panoramic radiography, and who underwent computed tomography for a detailed examination of these disorders, there were four patients in whom a definitive diagnosis of carotid artery calcification was made. We herein provide an overview of these cases, along with a briefreview of the relevant literature. When imaging findings suggestive of carotid artery calcification are obtained on panoramic radiographs, dentists can request detailed examinations and treatment at specialized medical institutions in the early stage by providing patients with information about the risks of atherosclerotic diseases. Early detection and treatment may reduce the severity or risks of complications and damage caused by atherosclerotic diseases, as well as possibly contributing to improved quality of life. In this study, we evaluated carotid artery calcification visible on panoramic radiographs. However, it seems that further detailed analysis is needed to examine the association between carotid artery calcification and a past history of risk factors for arteriosclerosis

    JAXA EARTH OBSERVATION DASHBOARD WITH COG AND WMS/WMTSS

    JAXA has developed and implemented earth observation (EO) dashboard jointly with ESA and NASA. The development of the JAXA dashboard, along with the "Earth-graphy" website and the newly developed "JAXA Earth API" service, demonstrate JAXA's commitment to providing climate change and earth science information to users worldwide. The EO dashboard serves as a platform to deliver valuable data and information related to climate change. The WMS/WMTS technology allows users to visualize and interact with geospatial information by providing web-based mapping services. This technology enhances the user experience by enabling the display of satellite imagery, overlays, and other geospatial data layers within the EO dashboard. To further facilitate the efficient use of satellite data, JAXA has developed the JAXA Earth API service. This service offers a user-friendly interface for accessing and utilizing JAXA's Earth observation satellite image data. By providing an easy-to-use format, JAXA aims to promote the effective utilization of satellite data and encourage its widespread use. Overall, the development and operation of the JAXA dashboard, with its integration of COG format data, WMS/WMTS technology, Python-based API.     A 100%-complete sequence reveals unusually simple genomic features in the hot-spring red alga Cyanidioschyzon merolae

    <p>Abstract</p> <p>Background</p> <p>All previously reported eukaryotic nuclear genome sequences have been incomplete, especially in highly repeated units and chromosomal ends. Because repetitive DNA is important for many aspects of biology, complete chromosomal structures are fundamental for understanding eukaryotic cells. Our earlier, nearly complete genome sequence of the hot-spring red alga <it>Cyanidioschyzon merolae </it>revealed several unique features, including just three ribosomal DNA copies, very few introns, and a small total number of genes. However, because the exact structures of certain functionally important repeated elements remained ambiguous, that sequence was not complete. Obviously, those ambiguities needed to be resolved before the unique features of the <it>C. merolae </it>genome could be summarized, and the ambiguities could only be resolved by completing the sequence. Therefore, we aimed to complete all previous gaps and sequence all remaining chromosomal ends, and now report the first nuclear-genome sequence for any eukaryote that is 100% complete.</p> <p>Results</p> <p>Our present complete sequence consists of 16546747 nucleotides covering 100% of the 20 linear chromosomes from telomere to telomere, representing the simple and unique chromosomal structures of the eukaryotic cell. We have unambiguously established that the <it>C. merolae </it>genome contains the smallest known histone-gene cluster, a unique telomeric repeat for all chromosomal ends, and an extremely low number of transposons.</p> <p>Conclusion</p> <p>By virtue of these attributes and others that we had discovered previously, <it>C. merolae </it>appears to have the simplest nuclear genome of the non-symbiotic eukaryotes. These unusually simple genomic features in the 100% complete genome sequence of <it>C. merolae </it>are extremely useful for further studies of eukaryotic cells.</p
