    Inertial reference unit

    The inertial reference unit is a high performance gyro attitude reference system for use on the OAO spacecraft. The IRU is a three axis system, which provides both rate and attitude information for spacecraft control. The purpose of the IRU is to reduce the dependency on gimballed startrackers and, in turn, simplify the OAO ground operations by eliminating the need for the continual programming of gimballed startracker assignments in accordance with computed occultation schedules. During normal operations, it is used to control the pitch and yaw axes during experiment occultations and during spacecraft reorientations. The roll axis is continuously under control of the IRU except during brief periods for attitude update. To provide for these capabilities the IRU must be able to perform two basic functions. One is to maintain an inertially fixed reference for spacecraft control and the second is to accurately reorient the reference upon command

    Development of an advanced high-temperature fastener system for advanced aerospace vehicle application

    The results of a program to develop a lightweight high temperature reusable fastening system for aerospace vehicle thermal protection system applications are documented. This feasibility program resulted in several fastener innovations which will meet the specific needs of the heat shield application. Three systems were designed from Hayes 188 alloy and tested by environmental exposure and residual mechanical properties. The designs include a clinch stud with a collar retainer, a weld stud with a split ring retainer, and a caged stud with a collar retainer. The results indicated that a lightweight, reusable, high temperature fastening system can be developed for aerospace vehicle application

    Force Generation by Kinesin and Myosin Cytoskeletal Motor Proteins

    Kinesins and myosins hydrolyze ATP, producing force that drives spindle assembly, vesicle transport and muscle contraction. How do motors do this? Here we discuss mechanisms of motor force transduction, based on their mechanochemical cycles and conformational changes observed in crystal structures. Distortion or twisting of the central β-sheet - proposed to trigger actin-induced Pi and ADP release by myosin, and microtubule-induced ADP release by kinesins - is shown in a movie depicting the transition between myosin ATP-like and nucleotide-free states. Structural changes in the switch I region form a tube that governs ATP hydrolysis and Pi release by the motors, explaining the essential role of switch I in hydrolysis. Comparison of the motor power strokes reveals that each stroke begins with the force-amplifying structure oriented opposite to the direction of rotation or swing. Motors undergo changes in their mechanochemical cycles in response to small-molecule inhibitors, several of which bind to kinesins by induced fit, trapping the motors in a state that resembles a force-producing conformation. An unusual motor activator specifically increases mechanical output by cardiac myosin, potentially providing valuable information about its mechanism of function. Further study is essential to understand motor mechanochemical coupling and energy transduction, and could lead to new therapies to treat human disease

    The Effect of Hot Gas in WMAP's First Year Data

    By cross-correlating templates constructed from the 2 Micron All Sky Survey (2MASS) Extended Source (XSC) catalogue with WMAP's first year data, we search for the thermal Sunyaev-Zel'dovich signature induced by hot gas in the local Universe. Assuming that galaxies trace the distribution of hot gas, we select regions on the sky with the largest projected density of galaxies. Under conservative assumptions on the amplitude of foreground residuals, we find a temperature decrement of -35 ±\pm 7 μ\muK (5σ\sim 5\sigma detection level, the highest reported so far) in the \sim 26 square degrees of the sky containing the largest number of galaxies per solid angle. We show that most of the reported signal is caused by known galaxy clusters which, when convolved with the average beam of the WMAP W band channel, subtend a typical angular size of 20--30 arcmins. Finally, after removing from our analyses all pixels associated with known optical and X-ray galaxy clusters, we still find a tSZ decrement of -96 ±\pm 37 μ\muK in pixels subtending about \sim 0.8 square degrees on the sky. Most of this signal is coming from five different cluster candidates in the Zone of Avoidance (ZoA), present in the Clusters In the ZoA (CIZA) catalogue. We found no evidence that structures less bound than clusters contribute to the tSZ signal present in the WMAP data.Comment: 10 pages, 4 figures, matches accepted version in ApJ Letter

    Dictyostelium Myosin-IE Is a Fast Molecular Motor Involved in Phagocytosis

    Class I myosins are single-headed motor proteins, implicated in various motile processes including organelle translocation, ion-channel gating, and cytoskeleton reorganization. Here we describe the cellular localization of myosin-IE and its role in the phagocytic uptake of solid particles and cells. A complete analysis of the kinetic and motor properties of Dictyostelium discoideum myosin-IE was achieved by the use of motor domain constructs with artificial lever arms. Class I myosins belonging to subclass IC like myosin-IE are thought to be tuned for tension maintenance or stress sensing. In contrast to this prediction, our results show myosin-IE to be a fast motor. Myosin-IE motor activity is regulated by myosin heavy chain phosphorylation, which increases the coupling efficiency between the actin and nucleotide binding sites tenfold and the motile activity more than fivefold. Changes in the level of free Mg(2+) ions, which are within the physiological range, are shown to modulate the motor activity of myosin-IE by inhibiting the release of adenosine diphosphate

    OAO-3 end of mission tests report

    Twelve engineering type tests were performed on several subsystems and experiment(s) of the OAO 3 spacecraft near its end of mission. The systems tested include: Princeton experiment package (PEP), fine error system guidance, inertial reference unit, star trackers, heat pipes, thermal control coatings, command and data handling, solar array; batteries, and onboard processor/power boost regulator. Generally, the systems performed well for the 8 1/2 years life of OAO 3, although some degradation was noted in the sensitivity of PEP and in the absorptivity of the skin coatings. Battery life was prolonged during the life of the mission in large part by carefully monitoring the charge-discharge cycle with careful attention not to overcharge

    A kinesin motor in a force-producing conformation

    <p>Abstract</p> <p>Background</p> <p>Kinesin motors hydrolyze ATP to produce force and move along microtubules, converting chemical energy into work by a mechanism that is only poorly understood. Key transitions and intermediate states in the process are still structurally uncharacterized, and remain outstanding questions in the field. Perturbing the motor by introducing point mutations could stabilize transitional or unstable states, providing critical information about these rarer states.</p> <p>Results</p> <p>Here we show that mutation of a single residue in the kinesin-14 Ncd causes the motor to release ADP and hydrolyze ATP faster than wild type, but move more slowly along microtubules in gliding assays, uncoupling nucleotide hydrolysis from force generation. A crystal structure of the motor shows a large rotation of the stalk, a conformation representing a force-producing stroke of Ncd. Three C-terminal residues of Ncd, visible for the first time, interact with the central β-sheet and dock onto the motor core, forming a structure resembling the kinesin-1 neck linker, which has been proposed to be the primary force-generating mechanical element of kinesin-1.</p> <p>Conclusions</p> <p>Force generation by minus-end Ncd involves docking of the C-terminus, which forms a structure resembling the kinesin-1 neck linker. The mechanism by which the plus- and minus-end motors produce force to move to opposite ends of the microtubule appears to involve the same conformational changes, but distinct structural linkers. Unstable ADP binding may destabilize the motor-ADP state, triggering Ncd stalk rotation and C-terminus docking, producing a working stroke of the motor.</p
