    Dynamic interactions of a conserved enterotoxigenic Escherichia coli adhesin with intestinal mucins govern epithelium engagement and toxin delivery

    At present, there is no vaccine for enterotoxigenic Escherichia coli (ETEC), an important cause of diarrheal illness. Nevertheless, recent microbial pathogenesis studies have identified a number of molecules produced by ETEC that contribute to its virulence and are novel antigenic targets to complement canonical vaccine approaches. EtpA is a secreted two-partner adhesin that is conserved within the ETEC pathovar. EtpA interacts with the tips of ETEC flagella to promote bacterial adhesion, toxin delivery, and intestinal colonization by forming molecular bridges between the bacteria and the epithelial surface. However, the nature of EtpA interactions with the intestinal epithelium remains poorly defined. Here, we demonstrate that EtpA interacts with glycans presented by transmembrane and secreted intestinal mucins at epithelial surfaces to facilitate pathogen-host interactions that culminate in toxin delivery. Moreover, we found that a major effector molecule of ETEC, the heat-labile enterotoxin (LT), may enhance these interactions by stimulating the production of the gel-forming mucin MUC2. Our studies suggest, however, that EtpA participates in complex and dynamic interactions between ETEC and the gastrointestinal mucosae in which host glycoproteins promote bacterial attachment while simultaneously limiting the epithelial engagement required for effective toxin delivery. Collectively, these data provide additional insight into the intricate nature of ETEC interactions with the intestinal epithelium that have potential implications for rational approaches to vaccine design

    The impact of new neutrino DIS and Drell-Yan data on large-x parton distributions

    New data sets have recently become available for neutrino and antineutrino deep inelastic scattering on nuclear targets and for inclusive dimuon production in pp pd interactions. These data sets are sensitive to different combinations of parton distribution functions in the large-x region and, therefore, provide different constraints when incorporated into global parton distribution function fits. We compare and contrast the effects of these new data on parton distribution fits, with special emphasis on the effects at large x. The effects of the use of nuclear targets in the neutrino and antineutrino data sets are also investigated.Comment: 24 pages, 13 figure

    A derivational model of discontinuous parsing

    The notion of latent-variable probabilistic context-free derivation of syntactic structures is enhanced to allow heads and unrestricted discontinuities. The chosen formalization covers both constituent parsing and dependency parsing. The derivational model is accompanied by an equivalent probabilistic automaton model. By the new framework, one obtains a probability distribution over the space of all discontinuous parses.     Comparison of <i>S</i>-Parameter Measurement Methods for Adapters

    In this work two methods for the characterization of adapters meaning reciprocal two-ports with different connector styles on both ports are analyzed and compared in detail. The focus is on the comparison of these approaches in terms of required effort and obtained measurement uncertainty. The first method is based on two one-port calibrations and it is compared to a two-port approach. Both methods are carried out in simulation as well as in measurement under very comparable conditions using a 2.4 to 3.5 mm adapter in the frequency range up to 33 GHz as an example. The obtained measurement uncertainties of the results are presented and analyzed, the required measurement time in terms of the necessary number of separate connections is compared, and the influence of cable movements is discussed.</p

    Micro-plasticity and intermittent dislocation activity in a simplified micro structural model

    Here we present a model to study the micro-plastic regime of a stress-strain curve. In this model an explicit dislocation population represents the mobile dislocation content and an internal shear-stress field represents a mean-field description of the immobile dislocation content. The mobile dislocations are constrained to a simple dipolar mat geometry and modelled via a dislocation dynamics algorithm, whilst the shear-stress field is chosen to be a sinusoidal function of distance along the mat direction. The latter, defined by a periodic length and a shear-stress amplitude, represents a pre-existing micro-structure. These model parameters, along with the mobile dislocation density, are found to admit a diversity of micro-plastic behaviour involving intermittent plasticity in the form of a scale-free avalanche phenomenon, with an exponent for the strain burst magnitude distribution similar to those seen in experiment and more complex dislocation dynamics simulations.Comment: 30 pages, 12 figures, to appear in "Modelling and Simulation in Materials Science and Engineering

    Initial conditions, Discreteness and non-linear structure formation in cosmology

    In this lecture we address three different but related aspects of the initial continuous fluctuation field in standard cosmological models. Firstly we discuss the properties of the so-called Harrison-Zeldovich like spectra. This power spectrum is a fundamental feature of all current standard cosmological models. In a simple classification of all stationary stochastic processes into three categories, we highlight with the name ``super-homogeneous'' the properties of the class to which models like this, with P(0)=0P(0)=0, belong. In statistical physics language they are well described as glass-like. Secondly, the initial continuous density field with such small amplitude correlated Gaussian fluctuations must be discretised in order to set up the initial particle distribution used in gravitational N-body simulations.     Blood Group O-Dependent cellular responses to cholera toxin: Parallel clinical and epidemiological links to severe cholera

    Because O blood group has been associated with more severe cholera infections, it has been hypothesized that cholera toxin (CT) may bind non-O blood group antigens of the intestinal mucosae, thereby preventing efficient interaction with target GM1 gangliosides required for uptake of the toxin and activation of cyclic adenosine monophosphate (cAMP) signaling in target epithelia. Herein, we show that after exposure to CT, human enteroids expressing O blood group exhibited marked increase in cAMP relative to cells derived from blood group A individuals. Likewise, using CRISPR/Cas9 engineering, a functional group O line (HT-29-A(−/−)) was generated from a parent group A HT-29 line. CT stimulated robust cAMP responses in HT-29-A(−/−) cells relative to HT-29 cells. These findings provide a direct molecular link between blood group O expression and differential cellular responses to CT, recapitulating clinical and epidemiologic observations
