    Cross-modal conflict increases with time-on-task in a temporal discrimination task

    The modality appropriateness hypothesis argues that the auditory modality is preferred over the visual modality in tasks demanding temporal operations; hence, we predicted that responses to visual stimuli would be more sensitive to the detrimental effect of Time-on-Task. We used a bimodal temporal discrimination task. The factors were durational congruency between the modalities and the direction of modality-transmission. Participants needed to decide the duration of the cued stimulus (visual or auditory). The first five blocks of the task lasted about 1.5 h without rest [Time-on-Task (ToT) period]. The participants then had a 12-min break followed by an additional block of trials. Subjective fatigue, reaction time, error rates, and electrocardiographic data were recorded. In the visual modality, we found an enhanced congruency effect as a function of ToT. The cost of attentional shifting was higher in the auditory modality, but remained constant, suggesting that processing of auditory stimuli is robust against the effects of fatigue. Performance did not improve after the break, indicating that the effects of fatigue could not be overcome by taking a brief break. The heart rate variability (HRV) data showed that vagal inhibition increased with ToT, but this increase was not associated with the changes in performance

    Fight against cold: photosynthetic and antioxidant responses of different bell pepper cultivars (Capsicum annuum L.) to cold stress

    The special metabolites of bell pepper (Capsicum annuum L.) leaves can protect the plant under possibly damaging circum- stances, such as high light, UV, unfavorable temperatures, or other environmental effects. In this study, we examined the cold stress tolerance of three different Hungarian pepper varieties (Darina, Édesalma, Rekord), focusing on the antioxidant and photosynthetic responses. The plants were developed in growth chambers under optimal temperature conditions (day/night 25 °C/20 °C) until the leaves on the fourth node became fully developed, then half of the plants received a cold treatment (day/ night 15 °C/10 °C). Via a detailed pigment analysis, the PS II chlorophyll fluorescence responses, gas exchange parameters and total antioxidant capacities, leaf acclimation to low temperatures has been characterized. Our results display some of the developing physiological and antioxidant properties, which are among the main factors in monitoring the damaging effects of cold temperatures. Nevertheless, despite their differences, the tested pepper varieties did not show different cold responses

    Diclofenac Prolongs Repolarization in Ventricular Muscle with Impaired Repolarization Reserve

    Background: The aim of the present work was to characterize the electrophysiological effects of the non-steroidal anti- inflammatory drug diclofenac and to study the possible proarrhythmic potency of the drug in ventricular muscle. Methods: Ion currents were recorded using voltage clamp technique in canine single ventricular cells and action potentials were obtained from canine ventricular preparations using microelectrodes. The proarrhythmic potency of the drug was investigated in an anaesthetized rabbit proarrhythmia model. Results: Action potentials were slightly lengthened in ventricular muscle but were shortened in Purkinje fibers by diclofenac (20 mM). The maximum upstroke velocity was decreased in both preparations. Larger repolarization prolongation was observed when repolarization reserve was impaired by previous BaCl 2 application. Diclofenac (3 mg/kg) did not prolong while dofetilide (25 mg/kg) significantly lengthened the QT c interval in anaesthetized rabbits. The addition of diclofenac following reduction of repolarization reserve by dofetilide further prolonged QT c . Diclofenac alone did not induce Torsades de Pointes ventricular tachycardia (TdP) while TdP incidence following dofetilide was 20%. However, the combination of diclofenac and dofetilide significantly increased TdP incidence (62%). In single ventricular cells diclofenac (30 mM) decreased the amplitude of rapid (I Kr ) and slow (I Ks ) delayed rectifier currents thereby attenuating repolarization reserve. L-type calcium current (I Ca ) was slightly diminished, but the transient outward (I to ) and inward rectifier (I K1 ) potassium currents were not influenced. Conclusions: Diclofenac at therapeutic concentrations and even at high dose does not prolong repolarization markedly and does not increase the risk of arrhythmia in normal heart. However, high dose diclofenac treatment may lengthen repolarization and enhance proarrhythmic risk in hearts with reduced repolarization reserve

    Biocompatibility study of poly(vinyl alcohol)-based electrospun scaffold for hernia repair

    Abdominal hernia is a purely surgical disorder where due to a defect in the abdominal wall, tissues or organs can extrude out of the abdominal cavity. The only conclusive treatment is surgical making a mesh implantation indispensable. Tissue engineering is now a promising method for creating scaffolds that provide an adequate support for tissue ingrowth. Our purpose was to develop a non-adhesive hernia mesh, which could be used in the repair of abdominal wall hernias but concurrently a scaffold for abdominal tissue regeneration. Poly(vinyl alcohol) bulk hydrogels are promising materials in wound dressing hence, interest in electrospun poly(vinyl alcohol) meshes has emerged in the past few years for different biomedical applications. In the present paper, preparation of electrospun poly(vinyl alcohol) fiber membranes and their in vitro and in vivo behaviors were followed to study the adhesion, biocompatibility, and biodegradability of the meshes. Our results showed that the surface of PVA meshes does not favor cell adhesion in vitro. During the animal experiments, PVA meshes demonstrated good integration into the surrounding tissue with minimal inflammatory reaction and minimal adhesions to intra abdominal structures

    Efficacy and safety of the biosimilar infliximab CT-P13 treatment in inflammatory bowel diseases: a prospective, multicentre, nationwide cohort

    Background and Aims: Biosimilar infliximab CT-P13 is approved for all indications of the originator product in Europe. Prospective data on its efficacy, safety, and immunogenicity in inflammatory bowel diseases are lacking. Methods: A prospective, nationwide, multicentre, observational cohort was designed to examine the efficacy, safety, and immunogenicity of CT-P13 infliximab biosimilar in the induction treatment of Crohn’s disease [CD] and ulcerative colitis [UC]. Demographic data were collected and a harmonised monitoring strategy was applied. Early clinical remission, response, and early biochemical response were evaluated at Week 14, steroid-free clinical remission was evaluated at Week 30. Therapeutic drug level was monitored using a conventional enzyme-linked immunosorbent assay. Results: In all, 210 consecutive inflammatory bowel disease [126 CD and 84 UC] patients were included in the present cohort. At Week 14, 81.4% of CD and 77.6% of UC patients showed clinical response and 53.6% of CD and 58.6% of UC patients were in clinical remission. Clinical remission rates at Week 14 were significantly higher in CD and UC patients who were infliximab naïve, compared with those with previous exposure to the originator compound [ p < 0.05]. Until Week 30, adverse events were experienced in 17.1% of all patients. Infusion reactions and infectious adverse events occurred in 6.6% and 5.7% of all patients, respectively. Conclusions: This prospective multicentre cohort shows that CT-P13 is safe and effective in the induction of clinical remission and response in both CD and UC. Patients with previous infliximab exposure exhibited decreased response rates and were more likely to develop allergic reactions

    Deep Investigation of Neutral Gas Origins (DINGO): HI stacking experiments with early science data

    We present early science results from Deep Investigation of Neutral Gas Origins (DINGO), an HI survey using the Australian Square Kilometre Array Pathfinder (ASKAP). Using ASKAP sub-arrays available during its commissioning phase, DINGO early science data were taken over \sim 60 deg2^{2} of the Galaxy And Mass Assembly (GAMA) 23 h region with 35.5 hr integration time. We make direct detections of six known and one new sources at z<0.01z < 0.01. Using HI spectral stacking, we investigate the HI gas content of galaxies at 0.04<z<0.090.04 < z< 0.09 for different galaxy colours. The results show that galaxy morphology based on optical colour is strongly linked to HI gas properties. To examine environmental impacts on the HI gas content of galaxies, three sub-samples are made based on the GAMA group catalogue. The average HI mass of group central galaxies is larger than those of satellite and isolated galaxies, but with a lower HI gas fraction. We derive a variety of HI scaling relations for physical properties of our sample, including stellar mass, stellar mass surface density, NUVrNUV-r colour, specific star formation rate, and halo mass. We find that the derived HI scaling relations are comparable to other published results, with consistent trends also observed to \sim0.5 dex lower limits in stellar mass and stellar surface density. The cosmic HI densities derived from our data are consistent with other published values at similar redshifts.     High rate of in-stent restenosis after coronary intervention in carriers of the mutant mannose-binding lectin allele

    BACKGROUND: In-stent restenosis occurs in 10-30% of patients following bare metal stent (BMS) implantation and has various risk factors. Mannose-binding lectin (MBL) is known to have effect on the progression of atherosclerosis. Single nucleotide polymorphisms (SNP) of the MBL2 gene intron 1 (codon 52, 54, 57) are known to modulate the bioavailability of the MBL protein. Our aim was to identify the association of these polymorphisms of the MBL gene in the occurrence of in-stent restenosis after coronary artery bare metal stent implantation. METHODS: In a non-randomized prospective study venous blood samples were collected after recoronarography from 225 patients with prior BMS implantation. Patients were assigned to diffuse restenosis group and control group based on the result of the coronarography. MBL genotypes were determined using quantitative real-time PCR. Proportion of different genotypes was compared and adjusted with traditional risk factors using multivariate logistic regression. RESULTS: Average follow-up time was 1.0 (+ - 1.4) year in the diffuse restenosis group (N = 117) and 2.7 (+ - 2.5) years in the control group (N = 108). The age, gender distribution and risk status was not different between study groups. Proportion of the MBL variant genotype was 26.8% (29 vs. 79 normal homozygous) in the control group and 39.3% (46 vs. 71 normal homozygous) in the restenosis group (p = 0.04). In multivariate analysis the mutant allele was an independent risk factor (OR = 1.96, p = 0.03) of in-stent restenosis. CONCLUSIONS: MBL polymorphisms are associated with higher incidence of development of coronary in-stent restenosis. The attenuated protein function in the mutant allelic genotype may represent the underlying mechanism

    Differentiation and Glucocorticoid Regulated Apopto-Phagocytic Gene Expression Patterns in Human Macrophages. Role of Mertk in Enhanced Phagocytosis

    The daily clearance of physiologically dying cells is performed safely mainly by cells in the mononuclear phagocyte system. They can recognize and engulf dying cells utilizing several cooperative mechanisms. In our study we show that the expression of a broad range of apopto-phagocytic genes is strongly up-regulated during differentiation of human monocytes to macrophages with different donor variability. The glucocorticoid dexamethasone has a profound effect on this process by selectively up-regulating six genes and down-regulating several others. The key role of the up-regulated mer tyrosine kinase (Mertk) in dexamethasone induced enhancement of phagocytosis could be demonstrated in human monocyte derived macrophages by gene silencing as well as blocking antibodies, and also in a monocyte-macrophage like cell line. However, the additional role of other glucocorticoid induced elements must be also considered since the presence of autologous serum during phagocytosis could almost completely compensate for the blocked function of Mertk
