    Serological evidence for human cystic echinococcosis in Slovenia

    <p>Abstract</p> <p>Background</p> <p>Cystic echinococcosis (CE) is caused by the larva of tapeworm <it>Echinococcus granulosus</it>. Dogs and other canids are the primary definitive hosts for this parasite. CE may develop after accidental ingestion of tapeworm eggs, excreted with the feces of these animals. In the intestine, the larvae released from the eggs are nested in the liver, lungs or other organs of livestock as intermediate hosts and humans as aberrant hosts. The aim of this study was to examine serologically whether some of the patients in Slovenia, suspected of CE by imaging findings in the liver or lungs had been infected with the larva of <it>Echinococcus granulosus</it>.</p> <p>Methods</p> <p>Between January 1, 2002 and the end of December 2006, 1323 patients suspected of having echinococcosis were screened serologically by indirect haemagglutination assay (IHA). For confirmation and differentiation of <it>Echinococcus </it>spp. infection, the sera of IHA-positive patients were then retested by western blot (WB).</p> <p>Results</p> <p>Out of 127 IHA-positive sera, 34 sera were confirmed by WB and considered specific for CE. Of 34 sera of CE-positive patients sera, 32 corresponded to the characteristic imaging findings of a liver cysts and 2 to those of lung cysts. The mean age of CE-positive patients was 58.3 years. No significant differences were found between the CE-positive patients in regard to their sex.</p> <p>Conclusion</p> <p>In the study, it was found out that CE was mostly spread in the same area of Slovenia as in the past, but its prevalence decreased from 4.8 per 10<sup>5 </sup>inhabitants in the period 1956–1968 to 1.7 per 10<sup>5 </sup>inhabitants in the period 2002–2006. In spite of the decreased prevalence of CE in the last years, it is suggested that clinicians and public health authorities, especially in the eastern parts of Slovenia where the most CE patients come from, should pay greater attention to this disease in the future.</p

    Single-cell analysis of CD4+ T-cell differentiation reveals three major cell states and progressive acceleration of proliferation

    Background: Differentiation of lymphocytes is frequently accompanied by cell cycle changes, interplay that is of central importance for immunity but is still incompletely understood. Here, we interrogate and quantitatively model how proliferation is linked to differentiation in CD4+ T cells. Results: We perform ex vivo single-cell RNA-sequencing of CD4+ T cells during a mouse model of infection that elicits a type 2 immune response and infer that the differentiated, cytokine-producing cells cycle faster than early activated precursor cells. To dissect this phenomenon quantitatively, we determine expression profiles across consecutive generations of differentiated and undifferentiated cells during Th2 polarization in vitro. We predict three discrete cell states, which we verify by single-cell quantitative PCR. Based on these three states, we extract rates of death, division and differentiation with a branching state Markov model to describe the cell population dynamics. From this multi-scale modelling, we infer a significant acceleration in proliferation from the intermediate activated cell state to the mature cytokine-secreting effector state. We confirm this acceleration both by live imaging of single Th2 cells and in an ex vivo Th1 malaria model by single-cell RNA-sequencing. Conclusion: The link between cytokine secretion and proliferation rate holds both in Th1 and Th2 cells in vivo and in vitro, indicating that this is likely a general phenomenon in adaptive immunity

    Reduced Transmissibility of East African Indian Strains of Mycobacterium tuberculosis

    BACKGROUND: Mycobacterium tuberculosis (MTB) has been classified into 4 main lineages. Some reports have associated certain lineages with particular clinical phenotypes, but there is still insufficient information regarding the clinical and epidemiologic implications of MTB lineage variation. METHODS: Using large sequence polymorphisms we classified MTB isolates from a population-based study in Montreal, Canada into the 4 major lineages, and identified the associated clinical and epidemiologic features. In addition, IS6110-RFLP and spoligotyping were used as indicators of recent TB transmission. The study population was divided into a derivation cohort, diagnosed between 2001 and 2007, and a separate validation cohort, diagnosed between 1996 and 2000. RESULTS: In the derivation cohort, when compared to the other MTB lineages, the East African-Indian (EAI) lineage was associated with lower rates of TB transmission, as measured by: positive TST among close contacts of pulmonary TB cases (adjusted odds ratio 0.6: [95% confidence interval 0.4-0.9]), and clustered TB cases (0.3: [<0.001-0.6]). Severe forms of TB were also less likely among the EAI group (0.4: [<0.001-0.8]). There were no significant differences when comparing patients with the other MTB lineages. In the validation cohort, the EAI lineage was associated with lower rates of positive TST among contacts (0.5: [0.3-0.9]) and a trend towards less clustered TB cases (0.5: [0.1-1.8]) when compared to the other lineages. Disease severity among the different groups was not significantly different in the validation cohort. CONCLUSIONS: We conclude that in Montreal, EAI strains were associated with reduced transmission compared to other MTB lineages

    Origin of Polar Order in Dense Suspensions of Phototactic Micro-Swimmers

    A main question for the study of collective motion in living organisms is the origin of orientational polar order, i.e., how organisms align and what are the benefits of such collective behaviour. In the case of micro-organisms swimming at a low Reynolds number, steric repulsion and long-range hydrodynamic interactions are not sufficient to explain a homogeneous polar order state in which the direction of motion is aligned. An external symmetry-breaking guiding field such as a mechanism of taxis appears necessary to understand this phonemonon. We have investigated the onset of polar order in the velocity field induced by phototaxis in a suspension of a motile micro-organism, the algae Chlamydomonas reinhardtii, for density values above the limit provided by the hydrodynamic approximation of a force dipole model. We show that polar order originates from a combination of both the external guiding field intensity and the population density. In particular, we show evidence for a linear dependence of a phototactic guiding field on cell density to determine the polar order for dense suspensions and demonstrate the existence of a density threshold for the origin of polar order. This threshold represents the density value below which cells undergoing phototaxis are not able to maintain a homogeneous polar order state and marks the transition to ordered collective motion. Such a transition is driven by a noise dominated phototactic reorientation where the noise is modelled as a normal distribution with a variance that is inversely proportional to the guiding field strength. Finally, we discuss the role of density in dense suspensions of phototactic micro-swimmers

    Phase Synchronization of fluid-fluid interfaces as hydrodynamically coupled oscillators

    Hydrodynamic interactions play a role in synchronized motions of coupled oscillators in fluids, and understanding the mechanism will facilitate development of applications in fluid mechanics. For example, synchronization phenomenon in two-phase flow will benefit the design of future microfluidic devices, allowing spatiotemporal control of microdroplet generation without additional integration of control elements. In this work, utilizing a characteristic oscillation of adjacent interfaces between two immiscible fluids in a microfluidic platform, we discover that the system can act as a coupled oscillator, notably showing spontaneous in-phase synchronization of droplet breakup. With this observation of in-phase synchronization, the coupled droplet generator exhibits a complete set of modes of coupled oscillators, including out-of-phase synchronization and nonsynchronous modes. We present a theoretical model to elucidate how a negative feedback mechanism, tied to the distance between the interfaces, induces the in-phase synchronization. We also identify the criterion for the transition from in-phase to out-of-phase oscillations

    Hydrodynamic coupling in polygonal arrays of colloids : Experimental and analytical results

    Colloidal particles are trapped harmonically on the vertices of planar regular polygons, using optical tweezers. The particles interact with each other via hydrodynamic coupling, which can be described adequately by Oseen’s tensor. Because of the interaction, the dynamics of any individual sphere is complex. Thermal motion results in a spectrum of relaxation times. The configuration of a system of N particles can be decomposed into 2N normal modes. In this work it is shown how to calculate these modes and their relaxation time scale analytically. The mathematical structure of the matrix of interaction leads to general properties for the symmetry of the normal modes and their dynamics, differing between the cases of even and odd N. The theory is compared to experiments performed on a range of rings with 3≤N≤10, varying also the trap stiffness and the distance between particles

    Efficiency of regional agriculture state support

    The study analyzed data on state support for agricultural production in the Samara region. The regional level is the main source of budgetary funding for the agro-industrial complex. The Samara region is a pilot region in the research field into the results impact effectiveness of support measures on the agricultural organizations activities. The study purpose is to substantiate the need to improve state support in agriculture in the Samara region. It is necessary to solve the following tasks: to analyze the experience of state regulation in the Samara region agro-industrial complex; to classify the existing directions of state support for agriculture; to substantiate the need to correct the existing approach when improving state support for agricultural production. As a result of the consistent policy implementation in the field of agro-industrial complex supporting, agricultural production in the Samara region was able to become quite effective. The twofold excess of taxes in 2019 over the amount of support indicates that agricultural producers were able to switch to expanded reproduction using their resources. If the current situation persists in the long term, then the state role will be reduced to stimulating extremely promising directions for the region's agro-industrial complex development. © Published under licence by IOP Publishing Ltd
