    Investigation of Database Models for Evolving Graphs

    We deal with the efficient implementation of storage models for time-varying graphs. To this end, we present an improved approach for the HiNode vertex-centric model based on MongoDB. This approach, apart from its inherent space optimality, exhibits significant improvements in global query execution times, which is the most challenging query type for entity-centric approaches. Not only significant speedups are achieved but more expensive queries can be executed as well, when compared to an implementation based on Cassandra due to the capability to exploit indices to a larger extent and benefit from in-database query processing

    A general strategy to optimize immunogenicity of HLA-B*0702 restricted cryptic peptides from tumor associated antigens: Design of universal neo-antigen like tumor vaccines for HLA-B*0702 positive patients

    International audienceTumor Associated Antigens (TAAs) are the privileged targets of almost all the cancer vaccines tested to date. Unfortunately all these vaccines failed to show a clinical efficacy. The main reason for this failure is the immune tolerance to TAAs that are self-proteins expressed by normal and cancer cells. Self-tolerance to TAAs is directed against their dominant rather than against their cryptic epitopes. The best way to overcome self-tolerance to TAAs would therefore be to target their cryptic epitopes. However, because of their low HLA-I affinity, cryptic peptides are non-immunogenic and cannot be used to stimulate an antitumor immune response unless their immunogenicity has been previously enhanced. In this paper we describe a general approach to enhance immunogenicity of almost all the HLA-B*0702 restricted cryptic peptides derived from TAAs. It consists in substituting residues at position 1 or 9 of low HLA-B*0702 affinity cryptic peptides by an Alanine or a Leucine respectively. These substitutions increase affinity of peptides for HLA-B*0702. These optimized cryptic peptides are strongly immunogenic and very importantly CTL they stimulate recognize their native counterparts. TAAs derived optimized cryptic peptides can be considered as universal antitumor vaccine since they escape self-tolerance, are immunogenic and are not patient specific

    Adaptive Fine-Tuning for Large-Scale Complex Nonlinear Systems

    Practical large-scale nonlinear control systems (PLSNCS) require an intensive and time-consuming fine-tuning process in order to achieve a satisfactory - or, even, acceptable - performance. In the majority of PLSNCS the fine-tuning process is performed by experienced personnel based on field observations and by experimenting with different combinations of controller parameters, without the use of a systematic approach. Adaptive Optimization (AO) methods such as the SPSA or AFT1 provide probably the most promising approach for the development of a systematic methodology for automatic and efficient fine-tuning of PLSNCS. However, despite the success of AO methodologies in particular fine-tuning control applications, these methods suffer from the serious problem of not guaranteeing efficient transient behaviour due to the use of random perturbations. A new algorithm (AFT2) has been developed and analyzed for alleviating this problem. This paper presents a comparative evaluation of AFT2 (versus SPSA and AFT1) when applied to three different traffic control applications: urban traffic control, network-wide motorway ramp metering and motorway variable speed control. It is demonstrated by means of simulation investigations that AFT2 not only overcomes the aforementioned problem of poor transient performance but also achieves significantly faster convergence than SPSA and AFT1.
