    Analysis of Dynamic Variation Characteristics and Influential Factors of PM2.5 on Subway Platforms under Air-Conditioning Condition and Ventilation Condition

    The purpose of this study is to investigate the main influential factors of the subway Particulate Matter (PM) pollutants under the air-conditioning condition and ventilation condition, by an on-site field measurement in a subway station in Shanghai. It is found that the dust accumulation at fresh air shaft was the secondary dusting action under the influence of fresh air flow, leading to a higher concentration of the PM in the subway station than in the outdoor air even when the outdoor air quality was good. In addition, in the morning rush hour the PM 2.5 value near the platform screen door our is highest of the day

    peer reviewedGlucocorticoids (GCs) can stimulate the appetite and AMPK in broilers. The activation of hypothalamic mTOR has been proposed as an important anorexigenic signal. However, inhibitory effect of AMPK activity on appetite and AMPK downstream signaling pathway under stress has not been reported. In this study, we performed an intracerebroventricular (icv) injection of compound C, an AMPK inhibitor, in GC-treated birds to explore the regulatory mechanism on appetite and AMPK downstream signaling pathway. A total of 48 7-day-old broilers, which had received an icv cannula, were randomly subjected to one of two treatments: subcutaneous injection of dexamethasone (DEX) or saline. After 3 days of continuous DEX injection, chicks of each group received an icv injection with either compound C (6 μg/2 μL) or vehicle (dimethyl sulfoxide, 2 μL). The results showed that body weight gain was reduced by the DEX treatment. Compared with the control, icv injection of compound C reduced feed intake at 0.5-1.5 h. In the DEX-treated group, the inhibitory effect of compound C on appetite remained apparent at 0.5-1 h. The DEX treatment increased the gene expression of liver kinase B1 (LKB1), neuropeptide Y (NPY), and decreased p-mTOR protein level. In stressed broilers, inhibition of AMPK relieved the decreased mTOR activity. A significant interaction was noted in DEX and compound C on protein expression of phospho-AMPK. Taken together, in stressed broilers, the central injection of compound C could inhibit central AMPK activity and reduce appetite, in which the AMPK/mTOR signaling pathway might be involved

    Expression Profiles of microRNAs in Drug-Resistant Non-Small Cell Lung Cancer Cell Lines Using microRNA Sequencing

    Background/Aims: Drug resistance remains a main obstacle to the treatment of non- small cell lung cancer (NSCLC). The aim of this study was to identify the expression profiles of microRNAs (miRNAs) in drug-resistant NSCLC cell lines. Methods: The expression profiles of miRNAs in drug-resistant NSCLC cell lines were examined using miRNA sequencing, and the common dysregulated miRNAs in these cell lines were identified and analyzed by bioinformatics methods. Results: A total of 29 upregulated miRNAs and 36 downregulated miRNAs were found in the drug-resistant NSCLC cell lines, of which 26 upregulated and 36 downregulated miRNAs were found to be involved in the Ras signaling pathway. The expression levels, survival analysis, and receiver operating characteristic curve of the dysregulated miRNAs based on The Cancer Genome Atlas database for lung adenocarcinoma showed that hsa-mir-192, hsa-mir-1293, hsa-mir-194, hsa-mir-561, hsa-mir-205, hsa-mir-30a, and hsa-mir-30c were related to lung cancer, whereas only hsa-mir-1293 and hsa-mir-561 were not involved in drug resistance. Conclusion: The results of this study may provide novel biomarkers for drug resistance in NSCLC and potential therapies for overcoming drug resistance, and may also reveal the potential mechanisms underlying drug resistance in this disease

    Objective Breast cancer is one of the most common malignant tumors that threaten the physical and mental health and even life-threatening of women worldwide.Chronic inflammation plays a key role in the occurrence, progression and recurrence of cancer.Several sources of evidence indicate that exercise during and after breast cancer could positively modulate the tumor microenvironment. The purpose of this meta-analysis is to determine the impact of exercise training on inflammatory factors in breast cancer patients. Methods We systematically searched the relevant randomized controlled trials published from January 1990 to June 2017 and analyzed them by Pubmed、Embase、Web of Science and Cochrane Library.The combination of Breast Cancer and Exercise is used for retrieval.The search term used for Pub Med is:(Exercise[Me SH] OR Physical Activity[Text Word] OR Training [Text Word] OR Activity [Text Word] OR Sport[Text Word] AND Breast Cancer[Text Word] OR Breast Tumor [Text Word] OR Breast Neoplasm [Text Word] OR Mammary Cancer [Text Word] OR Malignant Neoplasm of Breast [Text Word] OR Breast Carcinoma [Text Word] AND Interleukins [Text Word] OR IL [Text Word]).And manually check the references in the article to identify additional articles. Examination of titles and abstracts of papers based on pre-set inclusion criteria.Eleven high-quality trials were included. Results Pooled analyses revealed compared with the control group, the exercise group significantly improved the serum concentration of IL-8(Z=0.07,SMD=-0.02, 95%CI:[-0.47,-0.44],p=0.946)and TNF-α(Z=2.10, SMD=-0.60,95%CI:[-1.16,-0.04],p=0.036). No significant differences were found in the serum concentrations of IL-2(Z=1.96,SMD=-0.69,95%CI:[-1.37,0.00],p=0.05),IL-6(Z=0.40,SMD=-0.12, 95%CI: [-0.69,0.45],p=0.686),IL-10 (Z=1.73,SMD=-0.45,95%CI:[-0.95,0.06],p=0.084)or C-reactive protein(Z=0.18, SMD=-0.03,95%CI:[-0.35,0.41],p=0.861). Conclusions Exercise training can effectively improve some inflammatory factors in breast cancer patients and may affect tumor microenvironment. These findings provide a theoretical basis for the promotion of sports in this population

    Case Report: A rare case of non-small cell lung cancer with STRN-ALK fusion in a patient in very poor condition treated with first-line ensartinib

    Several cases of STRN-ALK fusion have been reported, and some anaplastic lymphoma kinase (ALK) inhibitors have been shown to be effective for treatment. Nevertheless, no cases of COVID-19 leading to heart failure and respiratory failure have been reported in people older than 70 years treated with ALK inhibitors. The present case report describes a 70-year-old patient with usual chronic obstructive pulmonary disease, diabetes, depression, and carotid plaque disease. Next-generation sequencing of tissue obtained by puncture biopsy revealed a STRN-ALK mutation accompanied by a TP53 mutation. The patient was treated with ensartinib and developed COVID-19 leading to heart failure and respiratory failure; nevertheless, he had a good clinical outcome and exhibited high treatment tolerability

    Effects of Dietary Energy Level on Performance, Plasma Parameters, and Central AMPK Levels in Stressed Broilers.

    peer reviewedThis study aimed to characterize the effects of diets with different energy levels on the growth performance, plasma parameters, and central AMPK signaling pathway in broilers under dexamethasone (DEX)-induced stress. A total of 216 1-day-old male broiler chickens were allocated to groups fed with high (HED), National Research Council-recommended (control), or low (LED) energy diets. At 10 days old, chickens were treated with or without dexamethasone (DEX, 2 mg/kg body weight) for 3 consecutive days. HED increased broiler average daily gain (ADG) at 10 days old, compared with the LED (P < 0.05), while average daily feed intake (ADFI) and feed conversion rate (FCR) decreased as the dietary energy level increased (P < 0.05). Chickens fed a HED had higher total protein (TP) content, albumin (ALB), glucose (GLU), total cholesterol (TCHO), high-density lipoprotein (HDL) cholesterol, and low-density lipoprotein (LDL) cholesterol, compared with the control group (P < 0.05). At 13 days old, DEX decreased ADG and increased FCR in broilers fed with different energy diets (P < 0.05). The DEX-HED group had a higher ADFI than non-DEX treated HED group chickens. In addition, TP, ALB, triglycerides (TG), TCHO, HDL, and LDL content levels in the DEX group were higher than those in the control group (P < 0.05). The uric acid (UA) content of the LED group was higher than that of the HED group (P < 0.05). Further, gene expression levels of liver kinase B1, AMP-activated protein kinase α1, neuropeptide Y, and GC receptor in the hypothalamus were increased in chickens treated with DEX (P < 0.05). There was a trend toward interaction between plasma TCHO and hypothalamic LKB1 expression (0.05 < P < 0.1). In conclusion, this study suggests that HED improves growth performance, plasma glucose and total cholesterol at 10 days old broilers, but had no significant effect on performance, plasma parameters, and central AMPK in stressed broilers

    BRCAA1 monoclonal antibody conjugated fluorescent magnetic nanoparticles for in vivo targeted magnetofluorescent imaging of gastric cancer

    <p>Abstract</p> <p>Background</p> <p>Gastric cancer is 2th most common cancer in China, and is still the second most common cause of cancer-related death in the world. How to recognize early gastric cancer cells is still a great challenge for early diagnosis and therapy of patients with gastric cancer. This study is aimed to develop one kind of multifunctional nanoprobes for <it>in vivo </it>targeted magnetofluorescent imaging of gastric cancer.</p> <p>Methods</p> <p>BRCAA1 monoclonal antibody was prepared, was used as first antibody to stain 50 pairs of specimens of gastric cancer and control normal gastric mucous tissues, and conjugated with fluorescent magnetic nanoparticles with 50 nm in diameter, the resultant BRCAA1-conjugated fluorescent magnetic nanoprobes were characterized by transmission electron microscopy and photoluminescence spectrometry, as-prepared nanoprobes were incubated with gastric cancer MGC803 cells, and were injected into mice model loaded with gastric cancer of 5 mm in diameter via tail vein, and then were imaged by fluorescence optical imaging and magnetic resonance imaging, their biodistribution was investigated. The tissue slices were observed by fluorescent microscopy, and the important organs such as heart, lung, kidney, brain and liver were analyzed by hematoxylin and eosin (HE) stain method.</p> <p>Results</p> <p>BRCAA1 monoclonal antibody was successfully prepared, BRCAA1 protein exhibited over-expression in 64% gastric cancer tissues, no expression in control normal gastric mucous tissues, there exists statistical difference between two groups (<it>P </it>< 0.01). The BRCAA1-conjugated fluorescent magnetic nanoprobes exhibit very low-toxicity, lower magnetic intensity and lower fluorescent intensity with peak-blue-shift than pure FMNPs, could be endocytosed by gastric cancer MGC803 cells, could target <it>in vivo </it>gastric cancer tissues loaded by mice, and could be used to image gastric cancer tissues by fluorescent imaging and magnetic resonance imaging, and mainly distributed in local gastric cancer tissues within 12 h post-injection. HE stain analysis showed that no obvious damages were observed in important organs.</p> <p>Conclusions</p> <p>The high-performance BRCAA1 monoclonal antibody-conjugated fluorescent magnetic nanoparticles can target <it>in vivo </it>gastric cancer cells, can be used for simultaneous magnetofluorescent imaging, and may have great potential in applications such as dual-model imaging and local thermal therapy of early gastric cancer in near future.</p

    Grover search algorithm accelerates the key search on the symmetric key cipher and the pre-image attack on the hash function. In order to perform Grover search algorithm, the target algorithm should be implemented in a quantum circuit. For this reason, we propose an optimal SM3 hash function (Chinese standard) in a quantum circuit. We focused on minimizing the use of qubits together with reducing the use of quantum gates. To do this, the on-the-fly approach is utilized for message expansion and compression functions. In particular, the previous value is restored and used without allocating new qubits in the permutation operation. Finally, we estimate quantum resources required for the quantum pre-image attack based on the proposed SM3 hash function implementation in the quantum circuit
