    The effect of matured date palm tree (Phoenix dactylifera L.) fruit extract on mite- induced allergic symptoms in NC/Nga mice

    The effect of a hot water extract of matured date palm tree fruit on allergic responses was investigated in NC/Nga mice. The allergic score was significantly reduced in the mice fed a date palm tree fruit extract-added diet than the fruit extract-free diet. The levels of serum antigen-specific immunoglobulin E and spleen interleukin (IL)-4+CD4+ cells were significantly lowered in the mice fed the fruit extract-added diet. Bruton's tyrosine kinase (Btk) and IL-2-inducible T cell kinase (Itk) mRNA expressions in spleen cells were significantly lowered in the mice fed the fruit extract-added diet. Moreover, chlorogenic acid and pelargonin decreased these mRNA expressions in C3H/HeN mouse spleen cell cultures.     The effects of oral ingestion of a hot water extract of matured fruit of the date palm tree (Phoenix dactylifera L.) on allergic responses were investigated in mite-sensitized mice. Sneezing and nose rubbing events in mice given a date extract-added diet were significantly lower than in those given an extract-free (control) diet. The serum total and mite antigen-specific immunoglobulin (Ig) E levels, and the number of spleen interleukin-4(+)CD4(+), IgE(+)B220(+) and Fc epsilon RI alpha(+)CD117(+) cells was significantly lower in mice given the date extract- added diet than in those given the control diet. Chlorogenic acid, pelargonin and ferulic acid significantly reduced the number of IgE(+)B220(+) cells, while chlorogenic acid and pelargonin significantly decreased the number of Fc epsilon RI alpha(+)CD117(+) cells in mouse spleen cell cultures.     Anti-allergic Properties of a Fruit Extract of Prune (Prunus domestica L.), in Mite-sensitized BALB/c Mice

    The effects of oral ingestion of a hot water extract of prune (Prunus domestica L.) fruit on allergic responses were investigated in mite-sensitized BALB/c mice. The number of sneezing events in mice given the extract-added diet was significantly lower than in mice given the extract-free diet. Serum mite allergen-specific immunoglobulin E levels were significantly lower in mice given the extract-added diet than in mice given the extract-free diet. In contrast, the ratio of splenic IFN-gamma(+)CD4(+) cells/IL-4(+)CD4(+) cells was higher in mice given the extract-added diet. Expression levels of Stat6, Btk and Stim1 mRNAs in spleen cells of mice given the extract-added diet were significantly lower than in mice given the extract-free diet.     Fermented Soybean Powder with Rice Mold in the Absence of Salt Stimulates the Cellular Immune System and Suppresses the Humoral Immune Response in Mice

    The immunomodulatory effect of fermented non-salty soybean powder (NSBP) was investigated in C3H/HeN mice. The number of splenic CD11b(+), CD49b(+), and interferon (IFN)-gamma(+)CD4(+) cells increased significantly, while that of interleukin (IL)-4(+)CD4(+) and CD19(+) cells decreased significantly in cultures containing NSBP. Similarly, in the spleen and Peyer's patches of mice fed a diet containing NSBP, the number of IL-12(+)CD11b(+), CD49b(+), and IFN-gamma(+)CD4(+) cells increased noticeably, whereas the number of splenic IL-4(+)CD4(+) and CD19b(+) cells was lower compared to mice fed an NSBP-free diet. Superoxide production by peritoneal macrophages was significantly higher in mice fed an NSBP-containing diet. Both intestinal total IgA and serum total IgG levels declined in mice fed the NSBP-containing diet. Microarray analysis of mRNAs extracted from Peyer's patch cells of mice fed the NSBP-containing diet indicated an increase in the expression of several genes related to cellular immune responses, while the expression of genes related to immunoglobulin production decreased.     Orally administered low-molecular weight agaro-oligosaccharides are absorbed into the plasma of healthy humans

    Agaro-oligosaccharides (AOSs) are known to have biological activities, such as anti-inflammatory, anti-tumor, and anti-obesity effects. Although existing evidence suggests the presence of AOSs in peripheral tissues after oral administration, whether AOSs permeate into the blood circulation remains unknown. Thus, we hypothesized that AOSs with low-molecular weight can permeate the human gastrointestinal tract. To test this hypothesis, the time course of absorption was examined by analyzing plasma samples before and 1, 2, and 4 h after ingestion. Analysis was performed using liquid chromatography/mass spectrometry after labeling with p-aminobenzoic ethyl ester. Our results showed that the plasma concentration of agarobiose (Abi) was higher than that of agarotetraose (Ate); however, agarohexaose was not detected. Additionally, plasma levels of Abi and Ate were proportional to the dose. These results suggest that permeation efficiency is dependent on the molecular weight and that the systemic absorption of Abi via the gastrointestinal tract is better than that of Ate. These findings will contribute to a better understanding of the bioactivity of orally administered AOSs in peripheral tissues

    The selective continued linkage of centromeres from mitosis to interphase in the absence of mammalian separase

    Separase is an evolutionarily conserved protease that is essential for chromosome segregation and cleaves cohesin Scc1/Rad21, which joins the sister chromatids together. Although mammalian separase also functions in chromosome segregation, our understanding of this process in mammals is still incomplete. We generated separase knockout mice, reporting an essential function for mammalian separase. Separase-deficient mouse embryonic fibroblasts exhibited severely restrained increases in cell number, polyploid chromosomes, and amplified centrosomes. Chromosome spreads demonstrated that multiple chromosomes connected to a centromeric region. Live observation demonstrated that the chromosomes of separase-deficient cells condensed, but failed to segregate, although subsequent cytokinesis and chromosome decondensation proceeded normally. These results establish that mammalian separase is essential for the separation of centromeres, but not of the arm regions of chromosomes. Other cell cycle events, such as mitotic exit, DNA replication, and centrosome duplication appear to occur normally. We also demonstrated that heterozygous separase-deficient cells exhibited severely restrained increases in cell number with apparently normal mitosis in the absence of securin, which is an inhibitory partner of separase

    Results of the search for inspiraling compact star binaries from TAMA300's observation in 2000-2004

    We analyze the data of TAMA300 detector to search for gravitational waves from inspiraling compact star binaries with masses of the component stars in the range 1-3Msolar. In this analysis, 2705 hours of data, taken during the years 2000-2004, are used for the event search. We combine the results of different observation runs, and obtained a single upper limit on the rate of the coalescence of compact binaries in our Galaxy of 20 per year at a 90% confidence level.     Observation results by the TAMA300 detector on gravitational wave bursts from stellar-core collapses

    We present data-analysis schemes and results of observations with the TAMA300 gravitational-wave detector, targeting burst signals from stellar-core collapse events. In analyses for burst gravitational waves, the detection and fake-reduction schemes are different from well-investigated ones for a chirp-wave analysis, because precise waveform templates are not available. We used an excess-power filter for the extraction of gravitational-wave candidates, and developed two methods for the reduction of fake events caused by non-stationary noises of the detector. These analysis schemes were applied to real data from the TAMA300 interferometric gravitational wave detector. As a result, fake events were reduced by a factor of about 1000 in the best cases. The resultant event candidates were interpreted from an astronomical viewpoint. We set an upper limit of 2.2x10^3 events/sec on the burst gravitational-wave event rate in our Galaxy with a confidence level of 90%. This work sets a milestone and prospects on the search for burst gravitational waves, by establishing an analysis scheme for the observation data from an interferometric gravitational wave detector

    Diethylstilbestrol (DES), an estrogen agonist, increases prolactin (PRL) cells through transdifferentiation of follicle-stimulating hormone (FSH) and luteinizing hormone (LH) cells to PRL cells as well as proliferation of PRL cells in adult male mouse pituitary. Since hyperacetylation of histone H3 is implicated in the regulation of activation of various genes, we examined the effect of DES on the state of histone H3 acetylation. DES significantly reduced the immunohistochemical signal for acetylated histone H3 at lysine 9 (H3K9ac) in PRL, LH and FSH cells, but not for H3K18ac or H3K23ac. DES-treated mice were injected intraperitoneally with HDAC inhibitors (HDACi), sodium phenylbutyrate (NaPB) or valproic acid (VPA), to mimic the acetylation level of histone H3. As expected, HDACi treatment restored the level of H3K9ac expression in these cells, and also inhibited DES-induced increase in PRL cells. Furthermore, NaPB and VPA also abrogated the effects of DES on the population density of both LH and FSH cells. Similarly, the numbers of proliferating and apoptotic cells in the pituitary in NaPB- or VPA-treated mice were comparable to those of the control mice.
