Refractive index measurement of hydrogen isotopologue mixture and applicability for homogeneity of hydrogen solid at cryogenic temperature in fusion fuel system
Deuterium (D)-Tritium (T) nuclear fusion reaction has potential as an energy source in the future. In both magnetic confinement and inertial confinement fusion reactors, solid D-T will generally be supplied as fusion fuel. The efficiency of the nuclear fusion reaction depends on the quality of solid D-T fuel, which is related to the composition, homogeneity, helium-3 (3He) content, and so on. However, there is no technique for in-situ examination of solid D-T fuel. In this study, we consider a simple and precise method for the characterization of solid hydrogen isotopologues at cryogenic temperature using refractive index measurement, and evaluate the distribution of hydrogen isotopologue composition and homogeneity. To evaluate without the effect of tritium decay, the homogeneity of the hydrogen (H2)-deuterium (D2) mixture is measured at first. By the in-situ refractive index measurement at cryogenic temperature, the homogeneity of solid H2-D2 mixture is roughly quantified. The phase diagram of the H2-D2 mixture shows a solid solution type. D2-rich crystal first appears from the liquid phase as a primary crystal. The composition of D2 in liquid phase ias homogeneous, whereas it reduces by obeying the liquidus line in the phase diagram with the crystallization. On the other hand, the composition of the H2-D2 mixture in solid phase is inhomogeneous because the mobility of H2 and D2 in solid phase was too slow to be homogeneous and solid. The compositions of H2-D2 mixture in liquid and solid phases could be evaluated by the in-situ refractive index measurement in time. A profile of pro-inflammatory cytokine expression in human Delta-1-induced monocyte-derived Langerhans cell-like dendritic cells after stimulation with Toll-like receptor ligands
Monocyte-derived Langerhans cell-like dendritic cells (Mo-LCs) are involved in epidermal disorders such as psoriasis in murine models. However, the roles of Mo-LCs in the pathogenesis of psoriasis in humans remain unclear. Also, the contribution of notch ligand delta-like 1 (DLL-1), expressed on keratinocytes, to Mo-LC functions requires clarification. Here, we established a new method of stimulating Mo-LCs derived from CD14+ monocytes with immobilized human DLL-1 to generate induced Mo-LCs (DI(+)Mo-LCs). The DI(+)Mo-LCs were compared to the dendritic cells derived from monocytes (Mo-DCs) cultured with interleukin-4 (IL-4) and granulocyte-macrophage colony-stimulating factor (GM-CSF), and M1 macrophages (Mφ) derived from monocytes cultured with GM-CSF. The DI(+)Mo-LCs were found to produce significant amounts of IL15, IL23A, and interferon-β (IFNB1) in response to the Toll-like receptor (TLR)3 agonist Polyinosinic-polycytidylic acid (Poly(I:C)) or TLR4 agonist lipopolysaccharide (LPS) despite their low expression of tumor necrosis factor (TNF). In conclusion, we have established a new method to generate DI(+)Mo-LCs. We have also discovered that DI(+)Mo-LCs have a unique capacity for producing IL15 and IL23A, which are related to the pathogenesis of psoriasis. Our data contribute to a better understanding of the roles of Mo-LCs in epidermal defense and pathogenesis
Refractive index measurements of solid deuterium–tritium
Physical properties of tritium (T) and deuterium (D) have been of great interest as a fuel for nuclear fusion. However, several kinds of the physical properties in a cryogenic environment have not been reported. Optical properties in liquid and solid phases are indispensable for the quality control of the DT fuel. We study the dependence of the refractive index of solid DT on temperature. A dedicated cryogenic system has been developed and forms a transparent solid DT in a prism cell. Refractive index measurements based on Snell’s law were conducted. The refractive indexes of solid DT are from 1.1618 ± 0.0002 to 1.1628 ± 0.0002 in the temperature range of 19.40 K to 17.89 K.Iwano K., Zhang J., Iwamoto A., et al. Incidence of endocrine-related immune-related adverse events in Japanese subjects with various types of cancer
BackgroundImmune checkpoint inhibitors (ICIs), such as cytotoxic T lymphocyte antigen-4 (CTLA-4) inhibitors, programmed cell death protein 1 (PD-1) inhibitors, and programmed cell death protein 1 ligand 1 (PD-L1) inhibitors, are often used to treat a variety of malignancies. ICIs are known to cause endocrine-related immune-related adverse events (irAEs), but the incidence varies among reports and/or agents. This study evaluated the incidence of endocrine-related irAEs in patients who were treated with ICIs in Japan.MethodThis single-center, retrospective, observational study examined the incidence and clinical characteristics of endocrine-related irAEs in 466 participants who were treated with ICIs at Kawasaki Medical School Hospital.ResultThe mean age of participants with and without endocrine-related irAEs was 69.1 ± 1.8 years and 68.1 ± 1.1 years, respectively, with no difference between them. The overall incidence of any endocrine-related irAEs among the participants was 25.5%. Hypothyroidism was prevalent in 24.3%, hypoadrenocorticism in 3.2%, hypopituitarism in 0.9%, and insulin-dependent diabetes mellitus in 1.1%. Participants receiving combination therapy with CTLA-4 and PD-1 inhibitors had a significantly higher incidence of endocrine-related irAEs than those receiving monotherapy.ConclusionEndocrine-related irAEs correlated significantly with survival and mean observation period. There was substantial difference in the incidence of endocrine-related irAEs among various types of ICIs and types of cancer. We should bear in mind that endocrine testing is necessary during the treatment with ICIs
Recurring radiation-induced angiosarcoma of the breast that was treated with paclitaxel chemotherapy: a case report
Background Angiosarcoma of the breast is very rare and can be divided into primary and secondary angiosarcoma. Radiation-induced angiosarcoma (RIAS) is classified as secondary angiosarcoma. Diagnosis of RIAS is difficult due to its rarity, and the interpretation of pathological imaging is complicated. In the National Comprehensive Care Network (NCCN) guidelines, the first choice of treatment is surgery with negative margins. Adjuvant radiotherapy (RT) for close soft tissue margins should be considered. Preoperative or adjuvant chemotherapy of nonmetastatic disease is not recommended for angiosarcoma. We report a case of RIAS, which was impossible to diagnose with core needle biopsy (CNB) but was diagnosed by excisional biopsy. The patient was then administered adjuvant chemotherapy using conjugated paclitaxel (PTX).
Case presentation A 62-year-old woman noticed a tumor in her right breast. She had a history of right breast cancer and had undergone breast-conserving surgery, RT, and tamoxifen therapy 8 years previously. CNB, which was performed twice, was inconclusive. The tumor was surgically excised and pathological analysis yielded a diagnosis of angiosarcoma. She then underwent a right mastectomy. One month after she underwent right mastectomy, a nodule reappeared on the skin of her right breast, and excisional biopsy revealed recurrence of angiosarcoma. A few weeks later another nodule reappeared near the post-operative scar and excisional biopsy revealed recurrence of angiosarcoma. We assumed that surgical therapy was insufficient because the patient experienced relapse of angiosarcoma after complete mastectomy. After the second recurrence, we treated her with systemic chemotherapy using PTX. There was no evidence of recurrence 8 months after chemotherapy.
Conclusion Although angiosarcoma is difficult to diagnose, many patients have a poor prognosis. Therefore, prompt treatment intervention is desired. Moreover, there is little evidence regarding adjuvant therapy of angiosarcoma since it is a rare disease. We consider that adjuvant therapy helped to effectively prevent recurrence in the patient after complete excision
Unified Total Synthesis, Stereostructural Elucidation, and Biological Evaluation of Sarcophytonolides
Sarcophytonolides are cembranolide diterpenes isolated from the soft corals of genus Sarcophyton. Unified total synthesis of sarcophytonolides C, E, F, G, H, and J and isosarcophytonolide D was achieved. The synthetic routes feature NaHMDS- or SmI2-mediated fragment coupling, alkoxycarbonylallylation, macrolactonization, and transannular ring-closing metathesis. These total syntheses led to the absolute configurational confirmation of sarcophytonolide H, elucidation of sarcophytonolides C, E, F, and G, and revision of sarcophytonolide J and isosarcophytonolide D. We also evaluated the antifouling activity and toxicity of the synthetic sarcophytonolides H and J and their analogues as well as the cytotoxicity of the synthetic sarcophytonolides and the key synthetic intermediates
Fabrication of high-concentration Cu-doped deuterated targets for fast ignition experiments
In high-energy-density physics, including inertial fusion energy using high-power lasers, doping tracer atoms and deuteration of target materials play an important role in diagnosis. For example, a low-concentration Cu dopant acts as an x-ray source for electron temperature detection while a deuterium dopant acts as a neutron source for fusion reaction detection. However, the simultaneous achievement of Cu doping, a deuterated polymer, mechanical toughness and chemical robustness during the fabrication process is not so simple. In this study, we report the successful fabrication of a Cu-doped deuterated target. The obtained samples were characterized by inductively coupled plasma optical emission spectrometry, differential scanning calorimetry and Fourier transform infrared spectroscopy. Demonstration of a spherical plasma mirror for the counter-propagating kilojoule-class petawatt LFEX laser system
A counter-propagating laser-beam platform using a spherical plasma mirror was developed for the kilojoule-class petawatt LFEX laser. The temporal and spatial overlaps of the incoming and redirected beams were measured with an optical interferometer and an x-ray pinhole camera. The plasma mirror performance was evaluated by measuring fast electrons, ions, and neutrons generated in the counter-propagating laser interaction with a Cu-doped deuterated film on both sides. The reflectivity and peak intensity were estimated as ∼50% and ∼5 × 1018 W/cm2, respectively. A Single Nucleotide Polymorphism within the Acetyl-Coenzyme A Carboxylase Beta Gene Is Associated with Proteinuria in Patients with Type 2 Diabetes
It has been suggested that genetic susceptibility plays an important role in the pathogenesis of diabetic nephropathy. A large-scale genotyping analysis of gene-based single nucleotide polymorphisms (SNPs) in Japanese patients with type 2 diabetes identified the gene encoding acetyl-coenzyme A carboxylase beta (ACACB) as a candidate for a susceptibility to diabetic nephropathy; the landmark SNP was found in the intron 18 of ACACB (rs2268388: intron 18 +4139 C > T, p = 1.4×10−6, odds ratio = 1.61, 95% confidence interval [CI]: 1.33–1.96). The association of this SNP with diabetic nephropathy was examined in 9 independent studies (4 from Japan including the original study, one Singaporean, one Korean, and two European) with type 2 diabetes. One case-control study involving European patients with type 1 diabetes was included. The frequency of the T allele for SNP rs2268388 was consistently higher among patients with type 2 diabetes and proteinuria. A meta-analysis revealed that rs2268388 was significantly associated with proteinuria in Japanese patients with type 2 diabetes (p = 5.35×10−8, odds ratio = 1.61, 95% Cl: 1.35–1.91). Rs2268388 was also associated with type 2 diabetes–associated end-stage renal disease (ESRD) in European Americans (p = 6×10−4, odds ratio = 1.61, 95% Cl: 1.22–2.13). Significant association was not detected between this SNP and nephropathy in those with type 1 diabetes. A subsequent in vitro functional analysis revealed that a 29-bp DNA fragment, including rs2268388, had significant enhancer activity in cultured human renal proximal tubular epithelial cells. Fragments corresponding to the disease susceptibility allele (T) had higher enhancer activity than those of the major allele. These results suggest that ACACB is a strong candidate for conferring susceptibility for proteinuria in patients with type 2 diabetes
