Outerwall and Beyond
In 2013, Outerwall (parent company of Coinstar and RedBox) acquired ecoATM, an automated e-waste recycling station. As a disruptive technology in the e-recycling business, ecoATM has demonstrated significant potential for triple-bottom-line success. Outerwall has struggled with negative press related to law-enforcement belief that these kiosks are “a motivator for the criminal element.” This CSR mini-case provides students an opportunity to work creatively to solve a public relations and perception problem in order to fully realize a company’s full potential for positive social, environmental and economic impact
Integrating Human Dimensions Studies and Human-Wildlife Conflict Data to Develop a Targeted Awareness Campaign About Coyotes in Florida
The Florida Fish and Wildlife Conservation Commission (FWC) uses qualitative and quantitative data on people’s values, beliefs, attitudes, and understanding of wildlife species to optimize outreach efforts to reduce human-wildlife conflict. These data also help the agency develop and implement effective and acceptable programs on fish and wildlife management by reducing adverse impacts of fish and wildlife on Florida’s environment, economy, and human health and safety. Combining human dimensions information with human wildlife conflict incident data can serve as a powerful tool to help prioritize outreach efforts.
In last few years, FWC has supported several research projects to gain a better understanding of human-coyote (Canis latrans) conflict across the state and public opinions across various demographics and geographic regions. The agency maintains a Wildlife Incident Management System (WIMS) database that records species-specific data related to human wildlife conflicts (including location, date, nature of incident, etc.). Human dimensions studies were conducted using focus groups, citizen surveys, and message testing. The research results in combination with the data collected in WIMS has helped FWC prioritize and target audiences for messaging related to coyote management. As a result, FWC has developed brochures, infographics, and hosted workshops in targeted areas in Florida as part of an on-going urban coyote and pet safety campaign
Acinetobacter baumannii Infection Inhibits Airway Eosinophilia and Lung Pathology in a Mouse Model of Allergic Asthma
Allergic asthma is a dysregulation of the immune system which leads to the development of Th2 responses to innocuous antigens (allergens). Some infections and microbial components can re-direct the immune response toward the Th1 response, or induce regulatory T cells to suppress the Th2 response, thereby inhibiting the development of allergic asthma. Since Acinetobacter baumannii infection can modulate lung cellular and cytokine responses, we studied the effect of A. baumannii in modulating airway eosinophilia in a mouse model of allergic asthma. Ovalbumin (OVA)-sensitized mice were treated with live A. baumannii or phosphate buffered saline (PBS), then intranasally challenged with OVA. Compared to PBS, A. baumannii treatment significantly reduced pulmonary Th2 cytokine and chemokine responses to OVA challenge. More importantly, the airway inflammation in A. baumannii-treated mice was strongly suppressed, as seen by the significant reduction of the proportion and the total number of eosinophils in the bronchoalveolar lavage fluid. In addition, A. baumannii-treated mice diminished lung mucus overproduction and pathology. However, A. baumannii treatment did not significantly alter systemic immune responses to OVA. Serum OVA-specific IgE, IgG1 and IgG2a levels were comparable between A. baumannii- and PBS-treated mice, and tracheobronchial lymph node cells from both treatment groups produced similar levels of Th1 and Th2 cytokines in response to in vitro OVA stimulation. Moreover, it appears that TLR-4 and IFN-Îł were not directly involved in the A. baumannii-induced suppression of airway eosinophilia. Our results suggest that A. baumannii inhibits allergic airway inflammation by direct suppression of local pulmonary Th2 cytokine responses to the allergen
Public acceptability of congestion charging in Beijing, China: How transferrable are Western ideas of public acceptability?
Public acceptability is a major concern for road pricing schemes in Western countries but has not yet been sufficiently studied in the context of Chinese cities, a number of which are considering the introduction of such travel constraint measures. This study explores factors influencing public acceptability of a proposed congestion charge in the City of Beijing. In so doing, the study focuses on understanding the appropriateness of Western frameworks for assessing public acceptability in the Chinese context.
Through literature review and focus groups a survey to test different public acceptability constructs was developed (N = 1104). A Structural Equation Model was used to analyze relationships that exist among the different aspects of public acceptability. The results demonstrate that public acceptability is dominantly influenced by the level of trust toward the Government and experts. Various determinants in the Western context, such as access to information and perceived effectiveness were not found to have a significant impact on public acceptability. The results imply that public acceptability of congestion charging in the Chinese context has a stronger resonance with wider social issues such as equity than more specific transport problems such as congestion. As such, attempting to present evidence on the anticipated effectiveness of the policy in alleviating congestion and smog may not make the policy more acceptable to the public. The overall inference of the study is that contextual factors are more important than has been previously considered within public acceptability studies

Background
The immunological escape of tumors represents one of the main ob- stacles to the treatment of malignancies. The blockade of PD-1 or CTLA-4 receptors represented a milestone in the history of immunotherapy. However, immune checkpoint inhibitors seem to be effective in specific cohorts of patients. It has been proposed that their efficacy relies on the presence of an immunological response. Thus, we hypothesized that disruption of the PD-L1/PD-1 axis would synergize with our oncolytic vaccine platform PeptiCRAd.
Methods
We used murine B16OVA in vivo tumor models and flow cytometry analysis to investigate the immunological background.
Results
First, we found that high-burden B16OVA tumors were refractory to combination immunotherapy. However, with a more aggressive schedule, tumors with a lower burden were more susceptible to the combination of PeptiCRAd and PD-L1 blockade. The therapy signifi- cantly increased the median survival of mice (Fig. 7). Interestingly, the reduced growth of contralaterally injected B16F10 cells sug- gested the presence of a long lasting immunological memory also against non-targeted antigens. Concerning the functional state of tumor infiltrating lymphocytes (TILs), we found that all the immune therapies would enhance the percentage of activated (PD-1pos TIM- 3neg) T lymphocytes and reduce the amount of exhausted (PD-1pos TIM-3pos) cells compared to placebo. As expected, we found that PeptiCRAd monotherapy could increase the number of antigen spe- cific CD8+ T cells compared to other treatments. However, only the combination with PD-L1 blockade could significantly increase the ra- tio between activated and exhausted pentamer positive cells (p= 0.0058), suggesting that by disrupting the PD-1/PD-L1 axis we could decrease the amount of dysfunctional antigen specific T cells. We ob- served that the anatomical location deeply influenced the state of CD4+ and CD8+ T lymphocytes. In fact, TIM-3 expression was in- creased by 2 fold on TILs compared to splenic and lymphoid T cells. In the CD8+ compartment, the expression of PD-1 on the surface seemed to be restricted to the tumor micro-environment, while CD4 + T cells had a high expression of PD-1 also in lymphoid organs. Interestingly, we found that the levels of PD-1 were significantly higher on CD8+ T cells than on CD4+ T cells into the tumor micro- environment (p < 0.0001).
Conclusions
In conclusion, we demonstrated that the efficacy of immune check- point inhibitors might be strongly enhanced by their combination with cancer vaccines. PeptiCRAd was able to increase the number of antigen-specific T cells and PD-L1 blockade prevented their exhaus- tion, resulting in long-lasting immunological memory and increased median survival
