    Herb-Induced Liver Injury in the Berlin Case-Control Surveillance Study

    Herb-induced liver injury (HILI) has recently attracted attention due to increasing reports of hepatotoxicity associated with use of phytotherapeutics. Here, we present data on HILI from the Berlin Case-Control Surveillance Study. The study was initiated in 2000 to investigate the serious toxicity of drugs including herbal medicines. Potential cases of liver injury were ascertained in more than 180 Departments of all 51 Berlin hospitals from October 2002 to December 2011. Drug or herb intake was assessed through a standardized face- to-face interview. Drug or herbal aetiology was assessed based on the updated Council for International Organizations of Medical Sciences scale. In ten of all 198 cases of hepatotoxicity included in the study, herbal aetiology was assessed as probable (once ayurvedic herb) or possible (Valeriana five times, Mentha piperita once, Pelargonium sidoides once, Hypericum perforatum once, Eucalyptus globulus once). Mean age was 56.4 ± 9.7 years, and the predominant pattern of liver injury was hepatocellular. No cases of acute liver failure or death were observed. This case series corroborates known risks for ayurvedic herbs, supports the suspected association between Valeriana use and liver injury, and indicates a hepatotoxic potential for herbs such as Pelargonium sidoides, Hypericum perforatum or Mentha piperita that were rarely associated with liver injury before. However, given that possible causality does not prove clinical significance, further studies in this field are needed

    A detailed investigation of the experimental conditions for the reversible addition fragmentation chain transfer-mediated copolymerization of acrylonitrile and butadiene

    The synthesis of acrylonitrile-butadiene rubbers (NBRs) via trithiocarbonate-mediated reversible addition fragmentation chain transfer (RAFT) polymerization of acrylonitrile (ACN) and 1,3-butadiene (BD) in solution under azeotropic conditions (38/62) was investigated for a broad range of common solvents: N,N-dimethylacetamide (DMAc), chlorobenzene, 1,4-dioxane, tert-butanol, isobutyronitrile, toluene, trimethylacetonitrile, dimethyl carbonate, acetonitrile, methyl acetate, acetone, and tert-butyl methyl ether. The gravimetrically determined conversions for the free radical polymerizations of ACN/BD after 22 h at 100 °C were in the range of 15% for methyl acetate to 35% for DMAc. The origin of the differences in conversion is attributed to the unequal decomposition behavior of the employed azo initiator 2,2â€Č-azobis(N-butyl-2-methylpropionamide) (1) in the solvents under investigation, as determined by ultraviolet-visible (UV-vis) spectroscopy. Relative decomposition of 1 in solution (0.1 mol L-1) at 100 °C was calculated from the UV-vis spectra for selected solvents. 90% of 1 in DMAc was decomposed after 22 h, 83% in tert-butanol, 57% in 1,4-dioxane, 53% in isobutyronitrile, 45% in chlorobenzene, and 21% in toluene. The evolution of molecular weight with conversion using the initiator 1 was in accordance with the theoretically expected values, regardless of the solvent studied. Moreover, the RAFT-mediated copolymerization of ACN/BD in DMAc with azo initiators 1, 1-[(1-cyano-1-methylethyl)azo]formamide (2) and 1,1â€Č- azobis(cyclohexanecarbonitrile) (3) was investigated. A strong deviation from the linear evolution of molecular weight due to a fast decomposition of these initiators - congruent with high primary radical delivery rates - at the selected temperature was observed when using 2 and 3.     High molecular weight acrylonitrile-butadiene architectures via a combination of RAFT polymerization and orthogonal copper mediated azide-alkyne cycloaddition

    α-Functional nitrile butadiene rubber (NBR) building blocks were employed in the copper mediated 1,3-dipolar Huisgen coupling upon addition of 1,4-bis(azidomethyl)benzene (4). Polymer-polymer coupling afforded linear polymers with molecular weights ranging from 2500 g mol -1 to 97000 g mol -1 and polydispersities from 1.1 to 1.6. The α-functional NBR building blocks were obtained via the reversible addition-fragmentation chain transfer (RAFT) copolymerization of acrylonitrile (AN) and 1,3-butadiene (BD) at 100 °C, utilizing the high temperature azo initiator 1,1â€Č-azobis(cyclohexane-1-carbonitrile) and chlorobenzene or acetone as solvents. A novel alkyne-functional trithiocarbonate 2 was synthesized in 64% yield via the N,Nâ€Č-dicyclohexylcarbodiimide mediated coupling of 2-((dodecylsulfanyl)carbono-thioyl)sulfanyl propanoic acid (DoPAT, 1) and propargyl alcohol. 2 was shown to be an efficient controlling agent for the controlled/living radical copolymerization of acrylonitrile and 1,3-butadiene. The use of copper mediated azide-alkyne cycloaddition was extended towards the side-chain modification of acrylonitrile-butadiene rubbers as well as applied in the synthesis of branched and cross-linked NBR structures. For this purpose an acrylonitrile-1,3-butadiene-propargyl methacrylate (PMA) terpolymer of 3900 g mol -1 with a PDI of 1.3 was synthesized by a DoPAT-mediated RAFT polymerization. Herein, monomers were employed in the ratio of 56:35:9 (BD:AN:PMA). The ability of the terpolymer to undergo side-chain modification was demonstrated upon addition of 1-undecane azide. Cross-links were established via addition of 1,4-bis(azidomethyl)benzene. The current study provides the first successful approach to employ an orthogonal conjugation technique on this technically important class of synthetic rubbers. © 2012 The Royal Society of Chemistry

    Drug‐induced liver injury: results from the hospital‐based Berlin Case–Control Surveillance Study

    AIM: Drug‐induced liver injury (DILI) is often responsible for acute liver failure, drug withdrawal, boxed warnings or drug non‐approval. Therefore, we conducted a case–control study to determine the hepatotoxic risk of a wide range of drugs. METHODS: The Berlin Case–Control Surveillance Study FAKOS included all 51 Berlin hospitals in a hospital network. Between 2002 and 2011, 198 patients with acute idiopathic hepatitis, 377 inpatient controls and 708 outpatient controls were ascertained. Case patients were thoroughly validated using anamnestic, clinical, laboratory and histological data. Drug exposure was obtained in a face‐to‐face interview. A possible drug aetiology was assessed in individual patients by applying the updated Council for International Organizations of Medical Sciences (CIOMS) scale. Drug risks were further quantified [odds ratios (OR) with 95% confidence intervals (CI)] in a case–control design with unconditional logistic regression analysis. Drug intake in the last 28 days before index date was considered for the analysis. RESULTS: The study corroborated hepatotoxic risks for a number of drugs, including phenprocoumon (OR 3.3, 95% CI 1.5, 6.7), amiodarone (OR 5.5, 95% CI 1.3, 21.2), clozapine (OR 34.6, 95% CI 2.8, 824.9) and flupirtine (OR 40.2, 95% CI 5.5, 856.9). Increased risks were also suggested for less commonly reported substances such as angiotensin II receptor blockers, atypical antipsychotics and for biperiden, a drug never before reported to be hepatotoxic. CONCLUSIONS: Our study identified a large number of drugs as possible causes of hepatotoxicity. The observed risk for seldom reported substances highlights the need for further post‐authorization safety studies not exclusively focusing on drugs already labelled as potentially hepatotoxic

    The Acts project: track reconstruction software for HL-LHC and beyond

    The reconstruction of trajectories of the charged particles in the tracking detectors of high energy physics (HEP) experiments is one of the most difficult and complex tasks of event reconstruction at particle colliders. As pattern recognition algorithms exhibit combinatorial scaling to high track multiplicities, they become the largest contributor to the CPU consumption within event reconstruction, particularly at current and future hadron colliders such as the LHC, HL-LHC and FCC-hh. Current algorithms provide an extremely high standard of physics and computing performance and have been tested on billions of simulated and recorded data events. However, most algorithms date back to more than 20 years ago and maintaining them has become increasingly challenging. In addition, they are challenging to adapt to modern programming paradigms and parallel architectures.Acts is based on the well-tested and highly functioning components of LHC track reconstruction algorithms, implemented with modern software concepts and inherently designed for parallel architectures. Multithreading becomes increasingly important to balance the memory usage per CPU core. However, a fully multithreaded event processing framework blurs the clear border between events, which has in the past often been used as a clearly defined validity boundary for event conditions. Acts is equipped with a full contextual conditions concept that allows to run concurrent track reconstruction even in case of multiple detector alignments, conditions or varying magnetic field being processed at the same time. It provides an experiment and, in particular, framework-independent software toolkit and light-weight, highly optimised event data model for track reconstruction. Particular care is given to thread safety and data locality. It is designed as a toolbox that allows to implement and extend widely known pattern recognition algorithms, and in addition suitable for algorithm templating and R&D. Acts has been used as the fast simulation engine for the Tracking Machine Learning (TrackML) Challenge, and will provide reference implementation of several submitted solution programs of the two phases of the challenge

    Unlike farm animals, wild animals are not subject to continuous health surveillance. Individual projects designed to screen wildlife populations for specific pathogens are, therefore, also of great importance for human health. In this context, the possible formation of a reservoir for highly pathogenic zoonotic pathogens is a focus of research. Two of these pathogens that have received particular attention during the last years are the novel severe acute respiratory syndrome coronavirus type 2 (SARS-CoV-2), due to its fast global spread and high impact to the human health, and, since its introduction into Germany, the flavivirus West Nile virus (WNV). Especially in combination with invasive vertebrate species (e.g., raccoons (Procyon lotor) and raccoon dogs (Nyctereutes procyonoides) in Germany), risk analysis must be done to enable health authorities to assess the potential for the establishment of new wild life reservoirs for pathogens. Therefore, samples were collected from raccoons and raccoon dogs and analyzed for the presence of SARS-CoV-2 and WNV infections in these populations. Molecular biological and serological data obtained imply that no SARS-CoV-2 nor WNV reservoir has been established in these two wild life species yet. Future investigations need to keep an eye on these invasive carnivore populations, especially since the close contact of these animals to humans, mainly in urban areas, would make animal–human transmission a challenge for human health
