    Siberian crane chick calls reflect their thermal state

    Chicks can convey information about their needs with calls. But it is still unknown if there are any universal need indicators in chick vocalizations. Previous studies have shown that in some species vocal activity and/or temporal-frequency variables of calls are related to the chick state, whereas other studies did not confirm it. Here, we tested experimentally whether vocal activity and temporal-frequency variables of calls change with cooling. We studied 10 human-raised Siberian crane (Grus leucogeranus) chicks at 3–15 days of age. We found that the cooled chicks produced calls higher in fundamental frequency and power variables, longer in duration and at a higher calling rate than in the control chicks. However, we did not find significant changes in level of entropy and occurrence of non-linear phenomena in chick calls recorded during the experimental cooling. We suggest that the level of vocal activity is a universal indicator of need for warmth in precocial and semi-precocial birds (e.g. cranes), but not in altricial ones. We also assume that coding of needs via temporal-frequency variables of calls is typical in species whose adults could not confuse their chicks with other chicks. Siberian cranes stay on separate territories during their breeding season, so parents do not need to check individuality of their offspring in the home area. In this case, all call characteristics are available for other purposes and serve to communicate chicks’ vital needs

    Long-term stability in the vocal duets of the endangered Siberian Crane Leucogeranus leucogeranus

    Vocal-based monitoring is increasingly being used as a non-invasive method for identifying individuals within avian populations and is promising for the Siberian Crane, Leucogeranus leucogeranus. This is a poorly studied, long-lived, secretive and critically endangered bird species that breeds in the Arctic tundra of western and eastern regions of Siberia. We assessed between- and within-year stability of individual-specific vocal features in duets of Siberian Crane and tested the effect of pair-mate change on their stability. Previous findings showed that duets are specific to different pairs of birds; however, it is still unknown how long pair-specific traits of duets remain and if they change in the course of a year or when birds re-mate. We recorded duets of 15 reproductively active pairs in the Oka Crane Breeding Centre in 2003–2006 and 2013–2017. We found that pair-specific vocal signatures remained stable both within the year and across ~ 10 years. After a change of mate, most of the variables we measured in the call did not change in any of the birds. Our data suggest that the stability of the individually specific vocal features may enable Siberian Cranes to be reliably identified by their duets over the birds’ lifetime. We believe that our work can increase confidence in the use of acoustic recognition techniques for endangered crane monitoring programs. Our results also suggest that Siberian Cranes may use their duets to form long-term social bonds between neighbours

    Voice breaking and its relation to body mass and testosterone level in the Siberian Crane (Leucogeranus leucogeranus)

    Vocal development of cranes (Gruidae) has attracted scientifc interest due to its special stage, voice breaking. During voice breaking, chicks of diferent crane species produce calls with two fundamental frequencies that correspond to those in adult low-frequency and juvenile high-frequency vocalizations. However, triggers that afect voice breaking in cranes are mainly unknown. Here we studied the voice breaking in the Siberian Crane (Leucogeranus leucogeranus) and test its relation to the body mass and testosterone level. We analyzed 5846 calls, 39 body mass measurements and 60 blood samples from 11 Siberian Crane chicks in 8 ages from 2.5 to 18 months of life together with 90 body mass measurements and 61 blood samples from 24 Siberian Crane adults. The individual duration of voice breaking and dates of its onset, culmination and completion depended neither on the body mass nor on the testosterone level at various ages. But we found correlation between the testosterone level and mean deltas of percentages of the high and low frequency components in Siberian Crane calls between the closest recording sessions. We also observed some coincidence in time between the mean dates of voice breaking onset and the termination of body mass gain (at 7.5 months of age), and between the mean dates of voice breaking completion and the start of a new breeding season. Similar relations have been shown previously for some other crane species. We also showed for the frst time that the mean dates of voice breaking culmination correlated with the signifcant increase of the testosterone level (at 10.5 months of age). So, we suggest that voice breaking in cranes may be triggered by the end of chicks’ body growth, is stimulated by the increase of testosterone level and ends soon after adult cranes stop taking care of their chicks

    Mutational Analysis of the Poly(ADP-Ribosyl)ation Sites of the Transcription Factor CTCF Provides an Insight into the Mechanism of Its Regulation by Poly(ADP-Ribosyl)ation

    ABSTRACT Poly(ADP-ribosyl)ation of the conserved multifunctional transcription factor CTCF was previously identified as important to maintain CTCF insulator and chromatin barrier functions. However, the molecular mechanism of this regulation and also the necessity of this modification for other CTCF functions remain unknown. In this study, we identified potential sites of poly(ADP-ribosyl)ation within the N-terminal domain of CTCF and generated a mutant deficient in poly(ADP-ribosyl)ation. Using this CTCF mutant, we demonstrated the requirement of poly(ADP-ribosyl)ation for optimal CTCF function in transcriptional activation of the p 19ARF promoter and inhibition of cell proliferation. By using a newly generated isogenic insulator reporter cell line, the CTCF insulator function at the mouse Igf2-H19 imprinting control region (ICR) was found to be compromised by the CTCF mutation. The association and simultaneous presence of PARP-1 and CTCF at the ICR, confirmed by single and serial chromatin immunoprecipitation assays, were found to be independent of CTCF poly(ADP-ribosyl)ation. These results suggest a model of CTCF regulation by poly(ADP-ribosyl)ation whereby CTCF and PARP-1 form functional complexes at sites along the DNA, producing a dynamic reversible modification of CTCF. By using bioinformatics tools, numerous sites of CTCF and PARP-1 colocalization were demonstrated, suggesting that such regulation of CTCF may take place at the genome level. </jats:p

    В статье анализируются экологическая уголовная политика и нормы об экологических преступлениях уголовного законодательства Российской Федерации. На основании системного анализа санкций статей главы 26 Уголовного кодекса Российской Федерации сделан вывод, что государственная власть недостаточно учитывает общественную опасность экологических преступлений и оценивает их с экономических позиций, как если бы они совершались в сфере экономической деятельности. По мнению автора статьи, критериями правильной экологической уголовной политики является обеспеченность безопасности не только личности и общества, но и государства. Экологические преступления угрожают государственной безопасности, поскольку разрушающе воздействуют на население и территории (базовые компоненты государства). В соответствии с целью обеспечения экологической безопасности должны выполняться задачи гарантирования рационального использования природных ресурсов в соответствии с закономерностями организации жизни, а также охраны окружающей среды. The article analyzes the environmental criminal policy and norms on environmental crimes of the criminal legislation of the Russian Federation. On the basis of the system analysis of the sanctions of the articles of Chapter 26 of the Criminal code of the Russian Federation, it is concluded that the state power does not take enough into account the social danger of environmental crimes and assesses them from an economic standpoint, as if they were committed in the sphere of economic activity. According to the author, the criteria of the correct environmental criminal policy is the security of not only the individual and society, but also the state. Environmental crimes threaten state security because they have a devastating impact on the population and territories (the basic components of the state). In accordance with the purpose of ensuring environmental safety, the tasks of guaranteeing the rational use of natural resources in accordance with the laws of the organization of life, as well as environmental protection, should be carried out

    Physical and chemical processes and the morphofunctional characteristics of human erythrocytes in hyperglycaemia

    Background: This study examines the effect of graduated hyperglycaemia on the state and oxygen-binding ability of hemoglobin, the correlation of phospholipid fractions and their metabolites in the membrane, the activity of proteolytic enzymes and the morphofunctional state of erythrocytes. Methods: Conformational changes in the molecule of hemoglobin were determined by Raman spectroscopy. The structure of the erythrocytes was analyzed using laser interference microscopy (LIM). To determine the activity of NADN-methemoglobinreductase, we used the P.G. Board method. The degree of glycosylation of the erythrocyte membranes was determined using a method previously described by Felkoren et al. Lipid extraction was performed using the Bligh and Dyer method. Detection of the phospholipids was performed using V. E. Vaskovsky method. Results: Conditions of hyperglycaemia are characterized by a low affinity of hemoglobin to oxygen, which is manifested as a parallel decrease in the content of hemoglobin oxyform and the growth of deoxyform, methemoglobin and membrane-bound hemoglobin. The degree of glycosylation of membrane proteins and hemoglobin is high. For example, in the case of hyperglycaemia, erythrocytic membranes reduce the content of all phospholipid fractions with a simultaneous increase in lysoforms, free fatty acids and the diacylglycerol (DAG). Step wise hyperglycaemia in incubation medium and human erythrocytes results in an increased content of peptide components and general trypsin-like activity in the cytosol, with a simultaneous decreased activity of µ-calpain and caspase 3. Conclusions: Metabolic disorders and damage of cell membranes during hyperglycaemia cause an increase in the population of echinocytes and spherocytes. The resulting disorders are accompanied with a high probability of intravascular haemolysis.</p
