    Genetic variation between Phytophthora cactorum isolates differing in their ability to cause crown rot in strawberry

    Analysis of 44 isolates of Phytophthora cactorum, isolated from strawberry and other hosts, by AFLP showed that the crown rot pathotype is different from leather rot isolates and from P. cactorum isolated from other hosts. 16 of 23 crown rot isolates, including isolates from Europe, Japan, Australia, and New Zealand, were identical in an analysis based on 96 polymorphic bands from seven primer combinations. Leather rot isolates of strawberry could not be distinguished from isolates from other hosts. The pathogenicity test of all 44 isolates on strawberry plants mostly gave unambiguous results, except for three American isolates, which seemed to have reduced aggressiveness compared to the crown rot isolates. These isolates also differed in the AFLP analysis. Comparing information on the origin of the isolates with results from the pathogenicity test, showed that isolates from strawberry fruits or petioles could be either leather rot or crown rot pathotypes. None of the isolates from hosts other than strawberry caused crown rot symptoms in strawberr

    Mycotoxin contamination of cereal grain commidities in relation to climate in North West Europe

    This study aimed to investigate mycotoxin contamination of cereal grain commodities for feed and food production in North Western Europe during the last two decades, including trends over time and co-occurrence between toxins, and to assess possible effects of climate on the presence of mycotoxins. For these aims, analytical results related to mycotoxin contamination of cereal grain commodities, collected in the course of national monitoring programmes in Finland, Sweden, Norway and the Netherlands during a 20-year period, were gathered. Historical observational weather data, including daily relative humidity, rainfall and temperature, were obtained from each of these four countries. In total 6382 records, referring to individual sample results for mycotoxin concentrations (one or more toxins) in cereal grains were available. Most records referred to wheat, barley, maize and oats. The most frequently analysed mycotoxins were deoxynivalenol, 3-acetyl-deoxynivalenol, nivalenol, T-2 toxin, HT-2 toxin and zearalenone. Deoxynivalenol had the highest overall incidence of 46%, and was mainly found in wheat, maize and oats. Mycotoxins that showed co-occurrence were: deoxynivalenol and 3-acetyl-deoxynivalenol in oats; deoxynivalenol and zearalenone in maize and wheat; and T-2 toxin and HT-2 toxin in oats. The presence of both deoxynivalenol and zearalenone in wheat increased with higher temperatures, relative humidity and rainfall during cultivation, but the presence of nivalenol was negatively associated with most of these climatic factors. The same holds for both nivalenol and deoxynivalenol in oats. This implies that climatic conditions that are conducive for one toxin may have a decreasing effect on the other. The presence of HT-2 toxin in oats showed a slight decreasing trends over time, but significant trends for other toxins showed an increasing presence during the last two decades. It is therefore useful to continue monitoring of mycotoxins. Obtained results can be used for development of predictive models for presence of mycotoxins in cereal grains

    Associations between Fusarium species and mycotoxins in oats and spring wheat from farmers fields in Norway over a six-year period

    During the last ten years, Norwegian cereal grain industry has experienced large challenges due to Fusarium spp.and Fusarium mycotoxin contamination of small-grained cereals. To prevent severely contaminated grain lotsfrom entering the grain supply chain, it is important to establish surveys for the most prevalent Fusarium spp. andmycotoxins. The objective of our study was to quantify and calculate the associations between Fusarium spp. andmycotoxins prevalent in oats and spring wheat. In a 6-year period from 2004-2009, 178 grain samples of springwheat and 289 samples of oats were collected from farmers fields in South East Norway. The grains were analysedfor 18 different Fusarium-mycotoxins by liquid chromatography mass spectrometry. Generally, the medianmycotoxin levels were higher than reported in Norwegian studies covering previous years. The DNA content ofFusarium graminearum, Fusarium culmorum, Fusarium langsethiae, Fusarium poae and Fusarium avenaceumwere determined by quantitative PCR. We identified F. graminearum as the main deoxynivalenol (DON) producerin oats and spring wheat, and F. langsethiae as the main HT-2 and T-2-toxins producer in oats. No association wasobserved between quantity of F. graminearum DNA and quantity of F. langsethiae DNA nor for their respectivemycotoxins, in oats. F. avenaceum was one of the most prevalent Fusarium species in both oats and spring wheat.The following ranking of Fusarium species was made based on the DNA concentrations of the Fusarium spp.analysed in this survey (from high to low): F. graminearum = F. langsethiae = F. avenaceum > F. poae > F. culmorum(oats); F. graminearum = F. avenaceum > F. culmorum > F. poae = F. langsethiae (spring wheat). Our results are inagreement with recently published data indicating a shift in the relative prevalence of Fusarium species towardsmore F. graminearum versus F. culmorum in Norwegian oats and spring wheat

    Antiinflammatory therapy with canakinumab for atherosclerotic disease

    BACKGROUND: Experimental and clinical data suggest that reducing inflammation without affecting lipid levels may reduce the risk of cardiovascular disease. Yet, the inflammatory hypothesis of atherothrombosis has remained unproved. METHODS: We conducted a randomized, double-blind trial of canakinumab, a therapeutic monoclonal antibody targeting interleukin-1β, involving 10,061 patients with previous myocardial infarction and a high-sensitivity C-reactive protein level of 2 mg or more per liter. The trial compared three doses of canakinumab (50 mg, 150 mg, and 300 mg, administered subcutaneously every 3 months) with placebo. The primary efficacy end point was nonfatal myocardial infarction, nonfatal stroke, or cardiovascular death. RESULTS: At 48 months, the median reduction from baseline in the high-sensitivity C-reactive protein level was 26 percentage points greater in the group that received the 50-mg dose of canakinumab, 37 percentage points greater in the 150-mg group, and 41 percentage points greater in the 300-mg group than in the placebo group. Canakinumab did not reduce lipid levels from baseline. At a median follow-up of 3.7 years, the incidence rate for the primary end point was 4.50 events per 100 person-years in the placebo group, 4.11 events per 100 person-years in the 50-mg group, 3.86 events per 100 person-years in the 150-mg group, and 3.90 events per 100 person-years in the 300-mg group. The hazard ratios as compared with placebo were as follows: in the 50-mg group, 0.93 (95% confidence interval [CI], 0.80 to 1.07; P=0.30); in the 150-mg group, 0.85 (95% CI, 0.74 to 0.98; P=0.021); and in the 300-mg group, 0.86 (95% CI, 0.75 to 0.99; P=0.031). The 150-mg dose, but not the other doses, met the prespecified multiplicity-adjusted threshold for statistical significance for the primary end point and the secondary end point that additionally included hospitalization for unstable angina that led to urgent revascularization (hazard ratio vs. placebo, 0.83; 95% CI, 0.73 to 0.95; P=0.005). Canakinumab was associated with a higher incidence of fatal infection than was placebo. There was no significant difference in all-cause mortality (hazard ratio for all canakinumab doses vs. placebo, 0.94; 95% CI, 0.83 to 1.06; P=0.31). CONCLUSIONS: Antiinflammatory therapy targeting the interleukin-1β innate immunity pathway with canakinumab at a dose of 150 mg every 3 months led to a significantly lower rate of recurrent cardiovascular events than placebo, independent of lipid-level lowering.
