    Intramolecular vibronic dynamics in molecular solids: C60

    Vibronic coupling in solid C60 has been investigated with a combination of resonant photoemission spectroscopy (RPES) and resonant inelastic x-ray scattering (RIXS). Excitation as a function of energy within the lowest unoccupied molecular orbital resonance yielded strong oscillations in intensity and dispersion in RPES, and a strong inelastic component in RIXS. Reconciling these two observations establishes that vibronic coupling in this core hole excitation leads to predominantly inelastic scattering and localization of the excited vibrations on the molecule on a femtosecond time scale. The coupling extends throughout the widths of the frontier valence bands.

    Multiplex PCR for detection of plasmid-mediated colistin resistance determinants, mcr-1, mcr-2, mcr-3, mcr-4 and mcr-5 for surveillance purposes

    Background and aim: Plasmid-mediated colistin resistance mechanisms have been identified worldwide in the past years. A multiplex polymerase chain reaction (PCR) protocol for detection of all currently known transferable colistin resistance genes (mcr-1 to mcr-5, and variants) in Enterobacteriaceae was developed for surveillance or research purposes. Methods: We designed four new primer pairs to amplify mcr-1, mcr-2, mcr-3 and mcr-4 gene products and used the originally described primers for mcr-5 to obtain a stepwise separation of ca 200 bp between ampli-cons. The primer pairs and amplification conditions allow for single or multiple detection of all currently described mcr genes and their variants present in Enterobacteriaceae. The protocol was validated testing 49 European Escherichia coli and Salmonella isolates of animal origin. Results: Multiplex PCR results in bovine and porcine isolates from Spain, Germany, France and Italy showed full concordance with whole genome sequence data. The method was able to detect mcr-1, mcr-3 and mcr-4 as singletons or in different combinations as they were present in the test isolates. One new mcr-4 variant, mcr-4.3, was also identified. Conclusions: This method allows rapid identification of mcr-positive bacteria and overcomes the challenges of phenotypic detection of colistin resistance. The multiplex PCR should be particularly interesting in settings or laboratories with limited resources for performing genetic analysis as it provides information on the mechanism of colistin resistance without requiring genome sequencing. © 2018, European Centre for Disease Prevention and Control (ECDC).     Northern Fennoscandia via the British Isles: evidence for a novel postglacial recolonization route by winter moth (Operophtera brumata)

    The frequency and severity of outbreaks by pestiferous insects is increasing globally, likely as a result of human-mediated introductions of non-native organisms. However, it is not always apparent whether an outbreak is the result of a recent introduction of an evolutionarily naïve population, or of recent disturbance acting on an existing population that arrived previously during natural range expansion. Here we use approximate Bayesian computation to infer the colonization history of a pestiferous insect, the winter moth, Operophtera brumata L. (Lepidoptera: Geometridae), which has caused widespread defoliation in northern Fennoscandia. We generated genotypes using a suite of 24 microsatellite loci and find that populations of winter moth in northern Europe can be assigned to five genetically distinct clusters that correspond with 1) Iceland, 2) the British Isles, 3) Central Europe and southern Fennoscandia, 4) Eastern Europe, and 5) northern Fennoscandia. We find that the northern Fennoscandia winter moth cluster is most closely related to a population presently found in the British Isles, and that these populations likely diverged around 2,900 years ago. This result suggests that current outbreaks are not the result of a recent introduction, but rather that recent climate or habitat disturbance is acting on existing populations that may have arrived to northern Fennoscandia via pre-Roman traders from the British Isles, and/or by natural dispersal across the North Sea likely using the Orkney Islands of northern Scotland as a stepping-stone before dispersing up the Norwegian coast. © 2021.     Molecular dynamics of ribosomal elongation factors G and Tu

    Translation on the ribosome is controlled by external factors. During polypeptide lengthening, elongation factors EF-Tu and EF-G consecutively interact with the bacterial ribosome. EF-Tu binds and delivers an aminoacyl-tRNA to the ribosomal A site and EF-G helps translocate the tRNAs between their binding sites after the peptide bond is formed. These processes occur at the expense of GTP. EF-Tu:tRNA and EF-G are of similar shape, share a common binding site, and undergo large conformational changes on interaction with the ribosome. To characterize the internal motion of these two elongation factors, we used 25 ns long all-atom molecular dynamics simulations. We observed enhanced mobility of EF-G domains III, IV, and V and of tRNA in the EF-Tu:tRNA complex. EF-Tu:GDP complex acquired a configuration different from that found in the crystal structure of EF-Tu with a GTP analogue, showing conformational changes in the switch I and II regions. The calculated electrostatic properties of elongation factors showed no global similarity even though matching electrostatic surface patches were found around the domain I that contacts the ribosome, and in the GDP/GTP binding region

    Consumption Style Among Young Adults Toward Their Shopping Behavior (An Empirical Study in Pakistan)

    The purpose of this study was to substantiate the consumption styles of adolescents as customer. The study was executed in Karachi, Pakistan by applying consumption style inventory scale. The data covered of 1,048 respondents who are young and educated mostly students, which belong to the different universities in Karachi. The data was collected through structured and self administered questionnaire. To test objective Independent sample t test was used. The results show that young females are more shopping influenced, fashion conscious, recreational, and confused over-choice as compare to males whereas males are more reliance on media, perfectionist, brand conscious, and impulsive as compare to females for their consumption style toward shopping behavior. This research provides understanding about adolescents’ decision making style of consumers in Karachi which would enable organizations to make more appropriate strategies to cater youth consumers market
