© 2016 Kirilova and Vlasova.Modern university education is aimed to provide a future expert with knowledge and experience of professional activity in the context of information society development and innovative knowledge-intensive production. At the same time information the teacher should be furnished with to implement own designing activity in the context of new education content formation is getting actual. Such designing is gaining an integrative character and becomes essential in the environment of controlled information streams. The paper reveals the phenomenon of education content integrative designing, features and invariant requirements to its content adequate to current trends of university education development, and ensures the unity and specificity of educational, scientific, production and social spheres of education content implementation realized within the selection of a variable logistic trajectory of its designing. The leading approaches to research the ways of integrative designing were system analysis of dynamic information educational environment and modeling of a logistic structure of information streams of educational purpose. The paper reveals the double-circuit organization of data exchange in the course of education content designing within which internal streams provide disciplinary differentiation, and external streams provide integration of disciplines, training profiles and educational structures. The paper develops subject - subject and subject – object exchange of information between educational organizations and society, between governing structures and divisions, between students and teachers in information educational environment of education content integrative designing. The paper proves that integration of information streams is a key idea to solve problems of timely and high-quality integrative designing of innovative education content. Practical significance of materials from the paper is that on the basis of described procedures the realization of developed functions and established interactions can be transferred to links of the information environment, and exchange of information becomes mediated by information resources and agents of educational purpose. The practical importance consists in the detailed and experimentally approved organization of an integration link regulating information exchanges in the course of designing and educational activity of subjects and agents of educational process

Quantitative analysis of the benzanthrone aminoderivative binding to amyloid fibrils of lysozyme

The accumulation of amyloid fibrils in different tissues is associated with a number of neurodegenerative diseases. Despite a huge variety of amyloid-specific probes, all of them suffer from many drawbacks, highlighting the necessity of searching for more preferable dyes. In the present work, the potential of new fluorescent probe AM3 for selective detection of fibrillar protein aggregates, formed from lysozyme, has been evaluated. To quantify the affinity of this dye for amyloid fibrils, the isotherms of dye binding to the fibrillar lysozyme have been derived from fluorimetric titration. Parameters of the dye-protein complexation: association constant, molar fluorescence and binding stoichiometry, calculated from the Langmuir adsorption model, revealed that AM3 interacts strongly with protein insoluble aggregates. High values of the binding parameters make AM3 an alternative to a widely-used amyloid-specific probe Thioflavin T. We also investigated the effects of polarity and viscosity on AM3 fluorescence properties. The binding of AM3 to the protein hydrophobic cavities has been followed by red shift of the dye emission spectra, which can be explained by H-bonding between proton-donating groups of the protein and carbonyl moiety of the probe. Long-wavelength shift of emission maximum was observed also upon increasing the excitation wavelength. This finding suggests that reorientation time of solvent molecules is higher, than the dye fluorescence lifetime. Fluorescence anisotropy studies revealed slow rotation diffusion of the probe, bound to amyloid fibrils being indicative of high viscosity of AM3 microenvironment. The observed photophysical properties of the new aminobenzanthrone derivative make AM3 a perspective probe for basic research and medical diagnostics

© 2015 by the authors. The article substantiates the meta dynamic approach that determines ways for information educational environment analysis. The implementation of the said approach includes a versatile analysis of information with the application of offered meta dynamic methods and tools. Tools of meta dynamics allow to regard changes in the rate of the information process, to analyze structural modifications of the information environment. The change of the information process can be analyzed, for example, if the rate of new information emegence and the rate of its processing are taken into account. These and other similar measuring instruments are based on indicators of the second order. The modification of the information structure is analyzed on the basis of absolute and relative indexes of the moved and changeable information volume, and the dynamics of the environment structure. There the process of information restructuring is implied, including, for example, the emergence of new structural components, their association, removal, specification. In this context there has been given a theoretical justification for the set of meta dynamic tools of the second order application. There have been presented some results of their experimental approbation. The significance of the designated approach has a predictive value in the development of a vocational school and its information educational environment

© Authors. Relevance of the research problem stems from the need to meet the challenges of personal growth of each participant of the educational process, a productive exchange of information and personalized contribution to the overall result of the conducted educational research. The aim of this paper is to improve joint training activities as the basis for future studies of masters that will be implemented in an open information and education space. The key approach to the study of the problems of the joint educational researches relies on the conceptual ideas of experiments conducting in joint environment in which access to individual and shared data in an open information education environment is restricted. Theoretical and technological tools were developed for working with factorial data of collaborative research in open educational environment. As a result these conceptual ideas were formulated for joint training of master's studies: the stages to improve joint research activities were grounded, the technology of forming joint bank of comparable research materials was created, the system of algorithms for collaborative (joint) working with experimental data was proposed, the strategy ensuring the adequacy of the joint pilot materials for attaining general and private purposes was worked out. This strategy also should be applied to the value and usefulness of collected data. The collected, studied, processed and presented in the paper unique experimental material can be useful both for solving current individual research tasks of undergraduates and for the development of new roles in research

Early anti-VEGF treatment in CRVO assures better prognosis

Introduction: Central retinal vein occlusion (CRVO) is the second most common cause of visual impairment due to retinal vascular disease in developed countries. The prevalence of CRVO is ranging from 0.1 to 0.5%. Risk factors are numerous, and fluctuations of IOP and thrombophilic conditions are considered to be the most important. Recently, extremely high levels of intraocular vascular endothelial growth factor (VEGF) were demonstrated in CRVO, leading to increased vascular permeability and leakage. Our objective is to present our approach in the management of treatment-naïve patients with CRVO. Methods: All consecutive patients with CRVO referred to our tertiary eye hospital for a period of 2 years were evaluated. Inclusion criteria: retinal and optic disk edema, dilatation and turtuosity of all retinal veins, widespread deep and superficial hemorrhages, cotton wool spots. We performed comprehensive eye exam, detailed medical history, specialized imaging methods, glaucoma evaluation, (AS-OCT, gonioscopy, IOP control), as well as hematological investigation in all patients. Treatment included intravitreal anti-VEGF application with personalized regimen. Glaucoma patients were treated with medications, laser and trabeculectomy (TE). We analyzed several variables: the period between the CRVO diagnosis and the start of anti-VEGF treatment, number of injections, changes in central macular thickness (CMT) and in visual acuity (VA). Results: Our pool of patients consisted of 38 persons, mean age 66.4 (29 -86). In 21 (56%) the CRVO is ischemic type, in 16 (42%) patients we diagnosed initial glaucoma damage (in 15 patients exfoliation syndrome was present). An interesting observation was the high number of patients - 13 (35%) with narrow, closed or occludable anterior chamber angle, diagnosed soon after the first CRVO symptoms. This finding was associated with history of accidental transient vision blurring and pain. In patients with start of treatment at the very beginning of macular edema we achieved immediate and excellent response after the first anti-VEGF application, and fast disappearance of exudations and hemorrhages. Hematologic studies demonstrated different types of hereditary thrombophilia in 7 patients. Conclusions: Our clinical experience demonstrate that best results in CRVO treatment could be obtained at early diagnosis, close follow up and early start of anti-VEGF treatment before pronounced macular edema occurs. Detailed evaluation of anterior chamber angle configuration and glaucoma management are mandatory for successful outcome in patients with initial CRVO symptoms

Interaction of novel benzanthrone derivative with amyloid lysozyme

A novel benzanthrone derivative AM18 was investigated with respect to its photophysical properties when bound to native, oligomeric and fibrillar hen egg white lysozyme. As shown by fluorimetric titration AM18 is more sensitive to pathogenic protein aggregates than Thioflavin T, however has no ability to differentiate between mature and immature lysozyme fibrils. The recovered affinity and fluorescence response of the novel probe to amyloid protein appeared to be similar to those of recently developed amyloid lysozyme-sensitive dyes like e. g. Nile Red and cyanine dye 7515. Despite the high increase of the probe emission in the presence of amyloid lysozyme compared to its fluorescence in buffer, the minimal amount that could be detected by 1 μM AM18 was 10 times lower for amyloid-native protein solutions due to high affinity of the dye for lysozyme monomers. In general, because of high quantum yields and “signal-to-noise” ratios in the presence of pathogenic protein aggregates AM18 appeared to be an effective tool for amyloid detection and characterization in vitro, being however unable to detect pathogenic protein aggregates in vivo like e.g. recently reported p-FTAA because of the sensitivity to lipids. Compared to previously reported AM3 a novel dye showed 2-fold lower “signal-to- noise” ratio in the presence of fibrillar lysozyme, and 2 fold lower blue shift of emission maximum. This tendency was explained in terms of decreased charge transfer from the donor to acceptor groupes of AM18 compared to AM3. Finally, as concluded from the comparison of AM18 and previously studied benzanthrone derivatives, the 5 nm – red edge excitation shift of AM18 is indicative of its possible binding to fibril “deep cavities”, containing no water. High anisotropy values of amyloid-bound dye led us to conclusion that the enhanced fluorescence of the probe is associated with the decrease of the rotational motion of the amino-substitute about the benzanthrone unit. This is a sign of AM18 behaviour as a molecular rotor
