A study of the biological effect of continuous inhalation exposure of 1, 1, 1-trichloroethene (methyl chloroform) on animals
The effects of continuous exposure to 1,1,1-trichloroethane on hepatic morphology and function are evaluated and compared with those produced by methylene chloride (dichloromethane) to determine environmental concentrations of each compound that would produce a similar biological response, i.e., a comparable increase in liver triglycerides over control levels. Experimental findings on mice, rats, dogs, and monkeys indicate that the pathological alternations observed with 1,1,1-trichloroethane are similar to those observed with dichloromethane except for different time courses of the effects and different degrees of recovery. A ten fold greater atmospheric concentration of 1,1,1-trichloroethane is required to produce the minimal liver changes found at 100 ppm dichloromethane

Laser-sheet imaging of HE-driven interfaces
This is the final report of a three-year, Laboratory Directed Research and Development (LDRD) project at Los Alamos National Laboratory (LANL). The authors made substantial progress in developing the MILSI (Multiple Imaging of Laser-Sheet Illumination) technique for high explosive (HE)-driven fluid interfaces. They observed the instability, but have not yet measured the instability growth rate. They developed suitable sample containers and optical systems for studying the Rightmyer-Meshkov instability of perturbed water/bromoform interfaces and they successfully fielded the new MILSI diagnostic at two firing-site facilities. The problem continues to be of central importance to the inertial confinement fusion (ICF) and weapons physics communities
High power testing of a fused quartz-based dielectricloaded accelerating structure
We report on the most recent results from a series of high power tests being carried out on rf-driven dielectric loaded accelerating (DLA) structures. The purpose of these tests is to determine the viability of the DLA as a traveling-wave accelerator and is a collaborative effort between Argonne National Laboratory (ANL), Naval Research Laboratory (NRL), and Stanford Linear Accelerator Center (SLAC). In this paper, we report on the recent high power tests of a fused quartz-based DLA structure that was carried out at incident powers of up to 12 MW at NRL and 37 MW at SLAC. We also report on test results of a TiN coated quartz structure, that exhibits good multipactor suppression
The research-teaching nexus: A case study of students' awareness, experiences and perceptions of research
This paper presents a case study of students' awareness, experiences and perceptions of research in a 'new' university in the UK. The findings are based on a questionnaire of almost 200 students and five small group interviews. Many of the students participating in this research perceived clear benefits to their learning from staff research, including being taught by enthusiastic staff, enhanced staff credibility, and the reflected glory of being taught by well-known researchers. However, they also perceived disadvantages, particularly with regard to staff availability, and did not believe that staff research should take priority over their needs as learners. They recognised that their awareness of the nature of research and the development of research skills increased most when they were actively involved in undertaking research projects. Several students also perceived benefits for future employment from their participation in research activities. The questionnaire has been used by several other universities around the world to benchmark their practices. Β© 2010 Taylor & Francis

Development of a 20 MeV Dielectric-Loaded Test Accelerator
This paper presents a progress report on a joint project by the Naval Research Laboratory (NRL) and Argonne National Laboratory (ANL), in collaboration with the Stanford Linear Accelerator Center (SLAC), to develop a dielectric-loaded test accelerator in the magnicon facility at NRL. The accelerator will be powered by an experimental 11.424-GHz magnicon amplifier that presently produces 25 MW of output power in a {approx}250-ns pulse at up to 10 Hz. The accelerator will include a 5-MeV electron injector originally developed at the Tsinghua University in Beijing, China, and can incorporate DLA structures up to 0.5 m in length. The DLA structures are being developed by ANL, and shorter test structures fabricated from a variety of dielectric materials have undergone testing at NRL at gradients up to {approx}8 MV/m. SLAC has developed components to distribute the power from the two magnicon output arms to the injector and to the DLA accelerating structure with separate control of the power ratio and relative phase. RWBruce Associates, Inc., working with NRL, has investigated means to join short ceramic sections into a continuous accelerator tube by a brazing process using an intense 83-GHz beam. The installation and testing of the first dielectric-loaded test accelerator, including injector, DLA test structure, and spectrometer, should take place within the next year
Habits of Mind: Designing Courses for Student Success
Although content knowledge remains at the heart of college teaching and learning, forward-thinking instructors recognize that we must also provide 21st-century college students with transferable skills (sometimes called portable intellectual abilities) to prepare them for their futures (Vazquez, 2020; Ritchhart, 2015; Venezia & Jaeger, 2013; Hazard, 2012). To βgrow their capacity as efficacious thinkers to navigate and thrive in the face of unprecedented changeβ (Costa et al., 2023), students must learn and improve important study skills and academic dispositions throughout their educational careers. If we do not focus on skills-building in college courses, students will not be prepared for the challenges that await them after they leave institutions of higher education. If students are not prepared for these postsecondary education challenges, then it is fair to say that college faculty have failed them
Expression and Function of Serotonin 2A and 2B Receptors in the Mammalian Respiratory Network
Neurons of the respiratory network in the lower brainstem express a variety of serotonin receptors (5-HTRs) that act primarily through adenylyl cyclase. However, there is one receptor family including 5-HT2A, 5-HT2B, and 5-HT2C receptors that are directed towards protein kinase C (PKC). In contrast to 5-HT2ARs, expression and function of 5-HT2BRs within the respiratory network are still unclear. 5-HT2BR utilizes a Gq-mediated signaling cascade involving calcium and leading to activation of phospholipase C and IP3/DAG pathways. Based on previous studies, this signal pathway appears to mediate excitatory actions on respiration. In the present study, we analyzed receptor expression in pontine and medullary regions of the respiratory network both at the transcriptional and translational level using quantitative RT-PCR and self-made as well as commercially available antibodies, respectively. In addition we measured effects of selective agonists and antagonists for 5-HT2ARs and 5-HT2BRs given intra-arterially on phrenic nerve discharges in juvenile rats using the perfused brainstem preparation. The drugs caused significant changes in discharge activity. Co-administration of both agonists revealed a dominance of the 5-HT2BR. Given the nature of the signaling pathways, we investigated whether intracellular calcium may explain effects observed in the respiratory network. Taken together, the results of this study suggest a significant role of both receptors in respiratory network modulation

Functional Stability of Unliganded Envelope Glycoprotein Spikes among Isolates of Human Immunodeficiency Virus Type 1 (HIV-1)
The HIV-1 envelope glycoprotein (Env) spike is challenging to study at the molecular level, due in part to its genetic variability, structural heterogeneity and lability. However, the extent of lability in Env function, particularly for primary isolates across clades, has not been explored. Here, we probe stability of function for variant Envs of a range of isolates from chronic and acute infection, and from clades A, B and C, all on a constant virus backbone. Stability is elucidated in terms of the sensitivity of isolate infectivity to destabilizing conditions. A heat-gradient assay was used to determine T90 values, the temperature at which HIV-1 infectivity is decreased by 90% in 1 h, which ranged between βΌ40 to 49Β°C (nβ=β34). For select Envs (nβ=β10), the half-lives of infectivity decay at 37Β°C were also determined and these correlated significantly with the T90 (pβ=β0.029), though two βoutliersβ were identified. Specificity in functional Env stability was also evident. For example, Env variant HIV-1ADA was found to be labile to heat, 37Β°C decay, and guanidinium hydrochloride but not to urea or extremes of pH, when compared to its thermostable counterpart, HIV-1JR-CSF. Blue native PAGE analyses revealed that Env-dependent viral inactivation preceded complete dissociation of Env trimers. The viral membrane and membrane-proximal external region (MPER) of gp41 were also shown to be important for maintaining trimer stability at physiological temperature. Overall, our results indicate that primary HIV-1 Envs can have diverse sensitivities to functional inactivation in vitro, including at physiological temperature, and suggest that parameters of functional Env stability may be helpful in the study and optimization of native Env mimetics and vaccines
