    Evaluation of Myelotoxicity in Dietary Restricted Rats

    The purpose of this study was to clarify the effect of decreased food consumption on evaluation of myelotoxicity in routine general toxicity studies. Male rats were divided into the following 7 groups: 12, 15, and 18 mg/kg 5-fluorouracil (5-FU) treatment groups (FU12, FU15 and FU18); dietary restriction groups (R12, R15 and R18 receiving the same amount of food as the rats in the FU12, FU15 and FU18 groups, respectively); and a nontreated control group (NT). We compared the changes in body weight, hematology and the results of cytological analyses of bone marrow and histopathology among the groups after administration and recovery periods of 14 and 7 days, respectively. At the end of the administration period, the FU15 and FU18 groups showed decreases in many hematologic and bone marrow parameters that were all similar to those in the corresponding dietary restriction groups (R15 and R18). A granulocyte abnormality (polyploidy: frequency of 1% or less) was also observed in all 5-FU treated groups. At the end of the recovery period, increases in the reticulocyte and platelet counts and extramedullary hematopoiesis of the spleen were observed in the 5-FU treated groups. These results indicate that the results of general toxicity studies in rats should be evaluated in consideration of dietary restriction effects when food consumption is decreased at about 30-40% or more. Careful morphological observation of hemocytes would be helpful in distinguishing the effect of a drug from that of dietary restriction in relation to hematological and bone marrow parameters. Performance of a recovery test to determine the reactive response of hematopoiesis is also recommended

    Importance of Starting Age for Myelotoxicity Study in Dietary Restricted Rats

    The aim of this study was to prove our hypothesis that adult rats with lowering of body weight gain, rats at 12 weeks of age as an example, are suitable for evaluation of myelotoxicity. Age-related differences between young rats (6-week-old study) and adult rats (12-week-old study) were analyzed in hematological examination values. The data of the young rats were reprinted from our previous report (Miyata et al., 2009) since our hypothesis was verified by comparison with that previous report. Several experimental groups were defined for the 12-week-old study as well as for the 6-week-old study; these included 5-fluorouracil (5-FU) treated groups receiving 12, 15 and 18 mg/kg/day (FU12, FU15 and FU18), pair-feeding groups (R12, R15 and R18 receiving the same amount of food as in the FU12, FU15 and FU18 groups, respectively) and a nontreated control group. Numerous hematologic and bone marrow parameters in the 5-FU treated groups were comparable to those in the corresponding pair-feeding groups in both age studies. Generally, the influences of undernutrition were more apparent in the young rats than in the adult rats. Histopathological examinations showed a decrease in hematopoiesis in the bone marrow in the 5-FU treated and pair-feeding groups. No apparent differences were observed in the decreased hematopoiesis between the 5-FU treated and pair-feeding groups in the 6-week-old study, but a difference between these groups was noted in the 12-week-old study; decreased hematopoiesis was more frequently noted in the 5-FU treated groups. These facts suggest that adult rats are more suitable than young rats for evaluation of 5-FU-induced myelotoxicity

    The Distribution and Migration of 137Cs in Oak (Quercus serrata) and Cedar (Cryptomeria japonica) Forest Organic Fractions

    To analyse the 137Cs distribution and migration under various fractions of organic matter layers, this study investigated easily recognizable, originally shaped organic L-fractions, and not easily recognizable, early fermented and fragmented organic F-fractions, of both oak (Quercus serrata) and cedar (Cryptomeria japonica) sampled from Osawa watershed sites at Nihonmatsu City, Fukushima Prefecture, Japan. The organic materials were put on top of soil columns from Field Museum (FM) Tamakyuryo in Hachioji City, Tokyo. The 137Cs vertical distribution in forest soil profiles was analyzed using the relaxation mass depth, ho (kg m−2). Soil columns with both L and F- organic layer fractions of both oak and cedar, labelled as Oak-L, Oak-F, Cedar-L and Cedar-F with four replications (n = 16), were set up by the laboratory column-based method and kept under five months’ incubation period. Soil columns after incubation were sampled at depths of 0–1 cm, 1–2 cm, 2–5 cm and 5–10 cm. Results of 137Cs inventory in the organic fractions showed that 86% (oak and cedar) of the total organic layer fractions 137Cs inventory accumulated within the F-layer, indicating that the transformation of litter is a huge source for potentially mobile 137Cs, especially the oak F-layer (67% 137Cs inventory) and further continuous transfer into the forest soil mineral layers. A higher ho in L treated soils (Oak-L and Cedar-L) compared to the F treatments implied that the low 137Cs amounts penetrated faster and deeper due to their water-soluble nature. Furthermore, Cedar-F showed a higher ho of 24.3 kg m−2 than Oak-F of ho, 14.0 kg m−2, and a significant positive relationship between 137Cs retention and total carbon (TC) (p < 0.05) suggested the influence of soil organic matter on 137Cs penetration and retention. The C/N (carbon nitrogen ratio) results revealed that organic matter fractions of high C/N including 137Cs, as observed in Cedar-F, in which decomposition does not advance, penetrates soil depths while the organic matter fraction of low C/N, observed in Oak-F, showed that decomposition advanced to release 137Cs which was held by adsorption unto the RIP (radiocesium interception potential) of soil surface.     Evaluation of Short-term Myelotoxicity Study in Dietary Reduced Rats

    This study attempted to prove our hypothesis that a short-term toxicity study, using a 4-day dosing regimen as an example, is suitable for evaluating myelotoxicity in rats. We compared the hematological, bone marrow cytological and histopathological results of 5-fluorouracil (5-FU) treated and pair-feeding groups after a 4-day administration period. Several experimental groups were defined for this 4-day study as well as for our previously reported 14-day study (Miyata et al., 2009); these included 5-FU treated groups receiving 12, 15 and 18 mg/kg/day (FU12, FU15 and FU18), pair-feeding groups (R12, R15 and R18 receiving the same amount of food as the FU12, FU15 and FU18 groups, respectively) and a nontreated control group. Although severe reductions in body weight gain and food consumption were reported in the 14-day study, only slight reductions were observed in the 4-day study. In the 4-day study, a decrease in blood reticulocytes and a decreasing trend of marrow erythroid cells were only observed in the FU18 group, and no effects were observed in the pair-feeding groups. The erythroblastic changes observed in this 4-day study were thought to reflect the direct influence of 5-FU administration. Since concerns regarding the influence of secondary changes related to undernutrition were minimized in the 4-day study, it was thought to clarify the direct influence of 5-FU administration on erythroblastic cells. Thus, a 4-day study protocol might be helpful for distinguishing secondary changes related to undernutrition

    Circumferential Spinal Cord Decompression through a Single Posterior Approach with Microendoscopy for Thoracic and Thoracolumbar Ossification of the Posterior Longitudinal Ligament

    Thoracic and thoracolumbar ossification of the posterior longitudinal ligament (OPLL) can be difficult to treat due to the anatomical position. The purpose of this study was to report the significance of a novel surgical technique that represented two cases of thoracic or thoracolumbar OPLL. The first patient was a 72-year-old woman who had a beak-type OPLL at the T11/12. The second was a 45-year-old woman who had a beak-type OPLL at the T12/L1. We performed circumferential spinal cord decompression through a single posterior approach with microendoscopy in both cases. The postoperative computed tomography revealed the complete removal of the OPLL, and the magnetic resonance imaging confirmed adequate decompression of the spinal cord. Preoperative symptoms were substantially improved in both patients. To date, we have used this novel technique to treat five patients with thoracic or thoracolumbar OPLL. This new surgical technique is likely to be useful in patients with a beak-type OPLL of the thoracic or thoracolumbar spine

    Experimental anemia induced by excess iron excretion.

    The effect of desferrioxamine on several hematological parameters was studied in growing rats. Animals given desferrioxamine intramuscularly (150 mg/kg daily) had increased urinary iron after one week, and decreased hemoglobin after 2 weeks. The difference in hemoglobin concentration between the treated and control groups was significant after 4 weeks of treatment with desferrioxamine, but the difference in the numbers of erythrocytes was not significant. The anemia was of hypochromic type. Desferrioxamine at this dosage did not retard growth. These findings support our previously reported concept of &#34;iron-losing anemia&#34;

    Non-destructive high-throughput measurement of elastic-viscous properties of maize using a novel ultra-micro sensor array and numerical validation

We developed an ultra-compact sensor array inspired by earthquake engineering and proposed a method for the high-throughput evaluation of the elastic modulus of maize cultivars. A natural vibration analysis based on the obtained Young’s modulus using finite element analysis (FEA) was performed and compared with the experimental results, which showed that the estimated Young’s modulus is representative of the individual Young’s modulus. FEA also showed the hotspot where the stalk was most deformed when the corn was vibrated by wind. The six tested cultivars were divided into two phenotypic groups based on the position and number of hotspots. In this study, we proposed a non-destructive high-throughput phenotyping technique for estimating the modulus of elasticity of maize stalks and successfully visualized which parts of the stalks should be improved for specific cultivars to prevent lodging

    Overlap lattice fermion in a gravitational field

    We construct a lattice Dirac operator of overlap type that describes the propagation of a Dirac fermion in an external gravitational field. The local Lorentz symmetry is manifestly realized as a lattice gauge symmetry, while it is believed that the general coordinate invariance is restored only in the continuum limit. Our doubler-free Dirac operator satisfies the conventional Ginsparg-Wilson relation and possesses gamma_5 hermiticity with respect to the inner product, which is suggested by the general coordinate invariance. The lattice index theorem in the presence of a gravitational field holds, and the classical continuum limit of the index density reproduces the Dirac genus. Reduction to a single Majorana fermion is possible for 8k+2 and 8k+4 dimensions, but not for 8k dimensions, which is consistent with the existence of the global gravitational/gauge anomalies in 8k dimensions. Other Lorentz representations, such as the spinor-vector and the bi-spinor representations, can also be treated.
