    DMSA-coated cubic iron oxide nanoparticles as potential therapeutic agents

    AIM: Superparamagnetic cubic iron oxide nanoparticles (IONPs) were synthesized and functionalized with meso-2,3-dimercaptosuccinic acid (DMSA) as a potential agent for cancer treatment. METHODS: Monodisperse cubic IONPs with a high value of saturation magnetization were synthesized by thermal decomposition method and functionalized with DMSA via ligand exchange reaction, and their cytotoxic effects on HeLa cells were investigated. RESULTS: DMSA functionalized cubic IONPs with an edge length of 24.5 ± 1.9 nm had a specific absorption rate value of 197.4 W/gFe (15.95 kA/m and 488 kHz) and showed slight cytotoxicity on HeLa cells when incubated with 3.3 × 10^{10}, 6.6 × 10^{10} and 9.9 × 10^{10} NP/mL for 24, 48 and 72 h. CONCLUSION: To the best of our knowledge, this is the first study to investigate both the cytotoxic effects of DMSA-coated cubic IONPs on HeLa cells and hyperthermia performance of these nanoparticles

    High magnetisation, monodisperse and water-dispersible CoFe@Pt core/shell nanoparticles

    High magnetisation and monodisperse CoFe alloy nanoparticles are desired for a wide range of biomedical applications. However, these CoFe nanoparticles are prone to oxidation, resulting in the deterioration of their magnetic properties. In the current work, CoFe alloy nanoparticles were prepared by thermal decomposition of cobalt and iron carbonyls in organic solvents at high temperatures. Using a seeded growth method, we successfully synthesised chemically stable CoFe@Pt core/shell nanostructures. The obtained core/shell nanoparticles have high saturation magnetisation up to 135 emu g−1. The magnetisation value of the core/shell nanoparticles remains 93 emu g−1 after being exposed to air for 12 weeks. Hydrophobic CoFe@Pt nanoparticles were rendered water-dispersible by encapsulating with poly(maleic anhydride-alt-1-octadecene) (PMAO). These nanoparticles were stable in water for at least 3 months and in a wide range of pH from 2 to 11

    Dimpled SiO₂@γ-Fe₂O₃ nanocomposites – fabrication and use for arsenic adsorption in aqueous medium

    We report the synthesis of nanocomposites made of silica nanoparticles whose six surface dimples are decorated with magnetic maghemite nanoparticles and their use for detection and recovery of arsenic in aqueous media. Precursor silica nanoparticles have aminated polystyrene chains at the bottom of their dimples and the maghemite nanoparticles are surface functionalized with carboxylic acid groups in two steps: amination with 3-aminopropyltrimethoxysilane, then derivatization with succinic anhydride in the presence of triethylamine. In the end, the colloidal assembly consists of the regioselective grafting of the carboxylic acid-modified iron oxide nanoparticles onto the 6-dimple silica nanoparticles. Several characterization techniques such as transmission electron microscopy (TEM), Fourier-transform infrared spectroscopy (FTIR), dynamic light scattering (DLS) are employed to assess the grafting process and study the influence of the maghemite functional groups on the quality of the composites formed. The resulting magnetic nanocomposites are used for the environmentally benign detection and removal of arsenic from aqueous medium, being readily extracted through means of magnetic separation

    A titanium dioxide/nitrogen-doped graphene quantum dot nanocomposite to mitigate cytotoxicity: synthesis, characterisation, and cell viability evaluation

    Titanium dioxide nanoparticles (TiO2 NPs) have attracted tremendous interest owing to their unique physicochemical properties. However, the cytotoxic effect of TiO2 NPs remains an obstacle for their wide-scale applications, particularly in drug delivery systems and cancer therapies. In this study, the more biocompatible nitrogen-doped graphene quantum dots (N-GQDs) were successfully incorporated onto the surface of the TiO2 NPs resulting in a N-GQDs/TiO2 nanocomposites (NCs). The effects of the nanocomposite on the viability of the breast cancer cell line (MDA-MB-231) was evaluated. The N-GQDs and N-GQDs/TiO2 NCs were synthesised using a one- and two-pot hydrothermal method, respectively while the TiO2 NPs were fabricated using microwave-assisted synthesis in the aqueous phase. The synthesised compounds were characterised using Fourier transform infrared (FTIR) spectroscopy, high-resolution transmission electron microscopy (HRTEM), field emission scanning electron microscopy (FESEM) and UV-visible spectrophotometry. The cell viability of the MDA-MB-231 cell line was determined using a CellTiter 96® AQueous One Solution Cell Proliferation (MTS) assay. The obtained results indicated that a monodispersed solution of N-GQDs with particle size 4.40 ± 1.5 nm emitted intense blue luminescence in aqueous media. The HRTEM images clearly showed that the TiO2 particles (11.46 ± 2.8 nm) are square shaped. Meanwhile, TiO2 particles were located on the 2D graphene nanosheet surface in N-GQDs/TiO2 NCs (9.16 ± 2.4 nm). N-GQDs and N-GQDs/TiO2 NCs were not toxic to the breast cancer cells at 0.1 mg mL−1 and below. At higher concentrations (0.5 and 1 mg mL−1), the nanocomposite was significantly less cytotoxic compared to the pristine TiO2. In conclusion, this nanocomposite with reduced cytotoxicity warrants further exploration as a new TiO2-based nanomaterial for biomedical applications, especially as an anti-cancer strategy

    Physical growth during the first year of life. A longitudinal study in rural and urban areas of Hanoi, Vietnam

    <p>Abstract</p> <p>Background</p> <p>Good infant growth is important for future health. Assessing growth is common in pediatric care all over the world, both at the population and individual level. There are few studies of birth weight and growth studies comparing urban and rural communities in Vietnam. The first aim is to describe and compare the birth weight distributions and physical growth (weight and length) of children during their first year in one rural and one urban area of Hanoi Vietnam. The second aim is to study associations between the anthropometric outcomes and indicators of the economic and educational situations.</p> <p>Methods</p> <p>Totally 1,466 children, born from 1<sup>st </sup>March, 2009 to June 2010, were followed monthly from birth to 12 months of age in two Health and Demographic Surveillance Sites; one rural and one urban. In all, 14,199 measurements each of weight and length were made. Birth weight was recorded separately. Information about demographic conditions, education, occupation and economic conditions of persons and households was obtained from household surveys. Fractional Polynomial models and standard statistical methods were used for description and analysis.</p> <p>Results</p> <p>Urban infants have higher birth weight and gain weight faster than rural infants. The mean birth weight for urban boys and girls were 3,298 grams and 3,203 grams as compared to 3,105 grams and 3,057 grams for rural children. At 90 days, the urban boys were estimated to be 4.1% heavier than rural boys. This difference increased to 7.2% at 360 days. The corresponding difference for girls was 3.4% and 10.5%. The differences for length were comparatively smaller. Both birth weight and growth were statistically significantly and positively associated with economic conditions and mother education.</p> <p>Conclusion</p> <p>Birth weight was lower and the growth, weight and length, considerably slower in the rural area, for boys as well as for girls. The results support the hypothesis that the rather drastic differences in maternal education and economic conditions lead to poor nutrition for mothers and children in turn causing inferior birth weight and growth.</p

    The Sudden Dominance of blaCTX–M Harbouring Plasmids in Shigella spp. Circulating in Southern Vietnam

    Shigellosis is a disease caused by bacteria belonging to Shigella spp. and is a leading cause of bacterial gastrointestinal infections in infants in unindustrialized countries. The Shigellae are dynamic and capable of rapid change when placed under selective pressure in a human population. Extended spectrum beta lactamases (ESBLs) are enzymes capable of degrading cephalosporins (a group of antimicrobial agents) and the genes that encode them are common in pathogenic E. coli and other related organisms in industrialized countries. In southern Vietnam, we have isolated multiple cephalosporin-resistant Shigella that express ESBLs. Furthermore, over two years these strains have replaced strains isolated from patients with shigellosis that cannot express ESBLs. Our work describes the genes responsible for this characteristic and we investigate one of the elements carrying one of these genes. These finding have implications for treatment of shigellosis and support the growing necessity for vaccine development. Our findings also may be pertinent for other countries undergoing a similar economic transition to Vietnam's and the corresponding effect on bacterial populations

    Transcriptome analysis of Bupleurum chinense focusing on genes involved in the biosynthesis of saikosaponins

    <p>Abstract</p> <p>Background</p> <p><it>Bupleurum chinense </it>DC. is a widely used traditional Chinese medicinal plant. Saikosaponins are the major bioactive constituents of <it>B. chinense</it>, but relatively little is known about saikosaponin biosynthesis. The 454 pyrosequencing technology provides a promising opportunity for finding novel genes that participate in plant metabolism. Consequently, this technology may help to identify the candidate genes involved in the saikosaponin biosynthetic pathway.</p> <p>Results</p> <p>One-quarter of the 454 pyrosequencing runs produced a total of 195, 088 high-quality reads, with an average read length of 356 bases (NCBI SRA accession SRA039388). A <it>de novo </it>assembly generated 24, 037 unique sequences (22, 748 contigs and 1, 289 singletons), 12, 649 (52.6%) of which were annotated against three public protein databases using a basic local alignment search tool (E-value ≤1e-10). All unique sequences were compared with NCBI expressed sequence tags (ESTs) (237) and encoding sequences (44) from the <it>Bupleurum </it>genus, and with a Sanger-sequenced EST dataset (3, 111). The 23, 173 (96.4%) unique sequences obtained in the present study represent novel <it>Bupleurum </it>genes. The ESTs of genes related to saikosaponin biosynthesis were found to encode known enzymes that catalyze the formation of the saikosaponin backbone; 246 cytochrome P450 (<it>P450</it>s) and 102 glycosyltransferases (<it>GT</it>s) unique sequences were also found in the 454 dataset. Full length cDNAs of 7 <it>P450</it>s and 7 uridine diphosphate <it>GT</it>s (<it>UGT</it>s) were verified by reverse transcriptase polymerase chain reaction or by cloning using 5' and/or 3' rapid amplification of cDNA ends. Two <it>P450</it>s and three <it>UGT</it>s were identified as the most likely candidates involved in saikosaponin biosynthesis. This finding was based on the coordinate up-regulation of their expression with <it>β-AS </it>in methyl jasmonate-treated adventitious roots and on their similar expression patterns with <it>β-AS </it>in various <it>B. chinense </it>tissues.</p> <p>Conclusions</p> <p>A collection of high-quality ESTs for <it>B. chinense </it>obtained by 454 pyrosequencing is provided here for the first time. These data should aid further research on the functional genomics of <it>B. chinense </it>and other <it>Bupleurum </it>species. The candidate genes for enzymes involved in saikosaponin biosynthesis, especially the <it>P450</it>s and <it>UGT</it>s, that were revealed provide a substantial foundation for follow-up research on the metabolism and regulation of the saikosaponins.</p

    Measurement of isomeric yield ratios for the Ho-nat(gamma,xn)Ho-164m,Ho-g;162m,Ho-g reactions in the bremsstrahlung energy region from 45 to 65 MeV

    We have measured the isomeric yield ratios for the Ho-164m,Ho-g and the Ho-162m,Ho-g isomeric pairs formed via photonuclear reactions Ho-165(gamma,n)Ho-164m,Ho-g and Ho-165(gamma,3n)Ho-162m,Ho-g in the bremsstrahlung energy region from 45 to 65 MeV in steps of a dagger E = 5 MeV by the activation method. The induced gamma-activities of the irradiated holmium foils were measured by a coaxial high-purity germanium detector coupled to a PC-based multichannel analyzer. In order to improve the accuracy of the experimental results the necessary corrections were made. The experimental results at bremsstrahlung energies of 45, 50, 55, 60, and 65 MeV are 0.408 +/- A 0.027, 0.413 +/- A 0.027, 0.427 +/- A 0.029, 0.452 +/- A 0.032, and 0.448 +/- A 0.031 for the Ho-164m,Ho-g isomeric pair, and 0.587 +/- A 0.041, 0.624 +/- A 0.044, 0.652 +/- A 0.045, 0.637 +/- A 0.045, and 0.668 +/- A 0.046 for the Ho-162m,Ho-g isomeric pair, respectively. The present results are the first measurements at bremsstrahlung energies just above the giant dipole resonance region. The obtained results confirm the dependence of isomeric ratios on the incident photon energy as observed in some earlier experiments.X110sciescopu

    Thermal neutron capture and resonance integral cross sections of Sc-45

    The thermal neutron cross section (sigma(0)) and resonance integral (I-0) of the Sc-45(n,gamma)Sc-46 reaction have been measured relative to that of the Au-197(n,gamma)Au-198 reaction by means of the activation method. High-purity natural scandium and gold foils without and with a cadmium cover of 0.5 mm thickness were irradiated with moderated pulsed neutrons produced from the Pohang Neutron Facility (PNF). The induced activities in the activated foils were measured with a high purity germanium (HPGe) detector. In order to improve the accuracy of the experimental results the counting losses caused by the thermal (G(th)) and resonance (G(epi)) neutron self-shielding, the gamma-ray attenuation (F-g) and the true gamma-ray coincidence summing effects were made. In addition, the effect of non-ideal epithermal spectrum was also taken into account by determining the neutron spectrum shape factor (alpha). The thermal neutron cross-section and resonance integral of the Sc-45(n,gamma)Sc-46 reaction have been determined relative to the reference values of the Au-197(n,gamma)Au-198 reaction, with sigma(o,Au) = 98.65 +/- 0.09 barn and = 1550 +/- 28 barn. The present thermal neutron cross section has been determined to be sigma(o,Sc) = 27.5 +/- 0.8 barn. According to the definition of cadmium cut-off energy at 0.55 eV, the present resonance integral cross section has been determined to be I-o,I-Sc = 12.4 +/- 0.7 barn.
