Computational investigation of particle penetration and deposition pattern in a realistic respiratory tract model from different types of dry powder inhalers
The aim of this study was to evaluate the device performance of a new design by comparing with a typical commercial DPI. Computational fluid dynamics (CFD) coupled with the discrete element method (DEM) collision has been utilized in this study to characterize and examine the flow field and particle transportation, respectively. A typical commercial DPI and an in-house designed novel DPI with distinct design features were compared to explore their dispersion capabilities and suitability for delivery to the respiratory tract. For this exploration, realistic oral to larynx and tracheobronchial airway models consisting of bio-relevant features were adopted to enhance practical feasibility. Distinct aerosol performances were observed between the two DPIs in the respiratory tract, where the in-house DPI, in comparison with the commercial DPI, has shown approximately 30% lower deposition fraction in the mouth-throat region with approximately 7% higher escape rate in the tracheobronchial region under the identical inhalation condition. This observation demonstrates that a novel in-house designed DPI provides higher device efficiency over the selected typical commercial DPI
Tumour-derived exosomes and their role in cancer-associated T-cell signalling defects
Dendritic and lymphoid ‘exosomes' regulate immune activation. Tumours release membranous material mimicking these ‘exosomes,' resulting in deletion of reactive lymphocytes. Tumour-derived ‘exosomes' have recently been explored as vaccines, without analysis of their immunologic consequences. This investigation examines the composition of tumour-derived ‘exosomes' and their effects on T lymphocytes. Membranous materials were isolated from ascites of ovarian cancer patients (n=6) and Western immunoblotting was performed for markers associated with ‘exosomes.' Using cultured T cells, ‘exosomes' were evaluated for suppression of CD3-ζ and JAK 3 expressions and induction of apoptosis, measured by DNA fragmentation. ‘Exosome' components mediating suppression of CD3-ζ were isolated by continuous eluting electrophoresis and examined by Western immunoblotting. ‘Exosomes' were shown to be identical with previously characterised shed membrane vesicles by protein staining and TSG101 expression. ‘Exosomes' expressed class I MHC, placental alkaline phosphatase, B23/nucleophosmin, and FasL. ‘Exosomes' suppressed expression of T-cell activation signalling components, CD3-ζ and JAK 3 and induced apoptosis. CD3-ζ suppression was mediated by two components: 26 and 42 kDa. Only the 42 kDa component reacted with anti-FasL antibody. These results indicate that, while ‘exosomes' express tumour antigens, leading to their proposed utility as tumour vaccines, they also can suppress T-cell signalling molecules and induce apoptosis
Optimal client recommendation for market makers in illiquid financial products
The process of liquidity provision in financial markets can result in
prolonged exposure to illiquid instruments for market makers. In this case,
where a proprietary position is not desired, pro-actively targeting the right
client who is likely to be interested can be an effective means to offset this
position, rather than relying on commensurate interest arising through natural
demand. In this paper, we consider the inference of a client profile for the
purpose of corporate bond recommendation, based on typical recorded information
available to the market maker. Given a historical record of corporate bond
transactions and bond meta-data, we use a topic-modelling analogy to develop a
probabilistic technique for compiling a curated list of client recommendations
for a particular bond that needs to be traded, ranked by probability of
interest. We show that a model based on Latent Dirichlet Allocation offers

Influences of operational parameters on phosphorus removal in batch and continuous electrocoagulation process performance

Performance of an electrocoagulation (EC) process in batch and continuous operating modes was thoroughly investigated and evaluated for enhancing wastewater phosphorus removal under various operating conditions, individually or combined with initial phosphorus concentration, wastewater conductivity, current density, and electrolysis times. The results revealed excellent phosphorus removal (72.7–100%) for both processes within 3–6 min of electrolysis, with relatively low energy requirements, i.e., less than 0.5 kWh/m3 for treated wastewater. However, the removal efficiency of phosphorus in the continuous EC operation mode was better than that in batch mode within the scope of the study. Additionally, the rate and efficiency of phosphorus removal strongly depended on operational parameters, including wastewater conductivity, initial phosphorus concentration, current density, and electrolysis time. Based on experimental data, statistical model verification of the response surface methodology (RSM) (multiple factor optimization) was also established to provide further insights and accurately describe the interactive relationship between the process variables, thus optimizing the EC process performance. The EC process using iron electrodes is promising for improving wastewater phosphorus removal efficiency, and RSM can be a sustainable tool for predicting the performance of the EC process and explaining the influence of the process variables

Using the \prospector\ spectral energy distribution (SED) fitting code, we
analyze the properties of 19 Extreme Emission Line Galaxies (EELGs) identified
in the bluest composite SED in the \zfourge\ survey at .
\prospector\ includes a physical model for nebular emission and returns
probability distributions for stellar mass, stellar metallicity, dust
attenuation, and nonparametric star formation history (SFH). The EELGs show
evidence for a starburst in the most recent 50 Myr, with the median EELG having
a specific star formation rate (sSFR) of 4.6 Gyr and forming 15\% of its
mass in this short time. For a sample of more typical star-forming galaxies
(SFGs) at the same redshifts, the median SFG has a sSFR of 1.1 Gyr and
forms only of its mass in the last 50 Myr. We find that virtually all of
our EELGs have rising SFHs, while most of our SFGs do not. From our analysis,
we hypothesize that many, if not most, star-forming galaxies at
undergo an extreme H+[\hbox{{\rm O}\kern 0.1em{\sc iii}}] emission
line phase early in their lifetimes. In a companion paper, we obtain
spectroscopic confirmation of the EELGs as part of our {\sc MOSEL} survey. In
the future, explorations of uncertainties in modeling the UV slope for galaxies
at are needed to better constrain their properties, e.g. stellar

Tumor-derived exosomes confer antigen-specific immunosuppression in a murine delayed-type hypersensitivity model
Exosomes are endosome-derived small membrane vesicles that are secreted by most cell types including tumor cells. Tumor-derived exosomes usually contain tumor antigens and have been used as a source of tumor antigens to stimulate anti-tumor immune responses. However, many reports also suggest that tumor-derived exosomes can facilitate tumor immune evasion through different mechanisms, most of which are antigen-independent. In the present study we used a mouse model of delayed-type hypersensitivity (DTH) and demonstrated that local administration of tumor-derived exosomes carrying the model antigen chicken ovalbumin (OVA) resulted in the suppression of DTH response in an antigen-specific manner. Analysis of exosome trafficking demonstrated that following local injection, tumor-derived exosomes were internalized by CD11c+ cells and transported to the draining LN. Exosome-mediated DTH suppression is associated with increased mRNA levels of TGF-β1 and IL-4 in the draining LN. The tumor-derived exosomes examined were also found to inhibit DC maturation. Taken together, our results suggest a role for tumor-derived exosomes in inducing tumor antigen-specific immunosuppression, possibly by modulating the function of APCs. © 2011 Yang et al
