    Prolonged Activation of ERK Contributes to the Photorejuvenation Effect in Photodynamic Therapy in Human Dermal Fibroblasts

    Photodynamic therapy (PDT) is known to be effective in the photorejuvenation of photoaged skin. However, the molecular mechanisms of rejuvenation by PDT remain elusive. In this study, we aimed to understand the molecular events occurring during the photorejuvenation after PDT in dermal fibroblasts in vitro. First, we found that PDT conditions resulted in an increased fibroblast proliferation and motility in vitro. Under this condition, cells had increased intracellular reactive oxygen species (ROS) production. Importantly, PDT induced a prolonged activation of extracellular signal–regulated kinase (ERK) with a corresponding increase in matrix metalloproteinase (MMP)-3 and collagen type Iα messenger RNA and protein. Moreover, inhibition of PDT-induced ERK activation significantly suppressed fibroblast proliferation and expression of MMP-3 and collagen type Iα following PDT. In addition, NAC (an antioxidant) inhibited PDT-induced fibroblast proliferation and ERK activation indicating that prolonged ERK activation and intracellular ROS contribute to the proliferation of fibroblasts and the dermal remodeling process for skin rejuvenation. We also identified increased collagen volume and decreased elastotic materials that are used as markers of photoaging in human skin samples using histochemical studies. Results from this study suggest that intracellular ROS stimulated by PDT in dermal fibroblasts lead to prolonged activation of ERK and, eventually, fibroblast proliferation and activation. Our data thus reveal a molecular mechanism underlying the skin rejuvenation effect of PDT

    SummaryStarvation activates MAPK in the pharyngeal muscles of C. elegans through a muscarinic acetylcholine receptor, Gqα, and nPKC as shown by the following results: (1) Starvation causes phosphorylation of MAPK in pharyngeal muscle. (2) In a sensitized genetic background in which Gqα signaling cannot be downregulated, activation of the pathway by a muscarinic agonist causes lethal changes in pharyngeal muscle function. Starvation has identical effects. (3) A muscarinic antagonist blocks the effects of starvation on sensitized muscle. (4) Mutations and drugs that block any step of signaling from the muscarinic receptor to MAPK also block the effects of starvation on sensitized muscle. (5) Overexpression of MAPK in wild-type pharyngeal muscle mimics the effects of muscarinic agonist and of starvation on sensitized muscle. We suggest that, during starvation, the muscarinic pathway to MAPK is activated to change the pharyngeal muscle physiology to enhance ingestion of food when food becomes available

    MicroRNAs: Synthesis, mechanism, function, and recent clinical trials

    AbstractMicroRNAs (miRNAs) are a class of small, endogenous RNAs of 21–25 nucleotides (nts) in length. They play an important regulatory role in animals and plants by targeting specific mRNAs for degradation or translation repression. Recent scientific advances have revealed the synthesis pathways and the regulatory mechanisms of miRNAs in animals and plants. miRNA-based regulation is implicated in disease etiology and has been studied for treatment. Furthermore, several preclinical and clinical trials have been initiated for miRNA-based therapeutics. In this review, the existing knowledge about miRNAs synthesis, mechanisms for regulation of the genome, and their widespread functions in animals and plants is summarized. The current status of preclinical and clinical trials regarding miRNA therapeutics is also reviewed. The recent findings in miRNA studies, summarized in this review, may add new dimensions to small RNA biology and miRNA therapeutics

    Analysis of Change in the Wind Speed Ratio according to Apartment Layout and Solutions

    Apartment complexes in various forms are built in downtown areas. The arrangement of an apartment complex has great influence on the wind flow inside it. There are issues of residents’ walking due to gust occurrence within apartment complexes, problems with pollutant emission due to airflow congestion, and heat island and cool island phenomena in apartment complexes. Currently, the forms of internal arrangements of apartment complexes are divided into the flat type and the tower type. In the present study, a wind tunnel experiment and computational fluid dynamics (CFD) simulation were performed with respect to internal wind flows in different apartment arrangement forms. Findings of the wind tunnel experiment showed that the internal form and arrangement of an apartment complex had significant influence on its internal airflow. The wind velocity of the buildings increased by 80% at maximum due to the proximity effects between the buildings. The CFD simulation for relaxing such wind flows indicated that the wind velocity reduced by 40% or more at maximum when the paths between the lateral sides of the buildings were extended

    Clinical Characteristics of Radiation Oncology in Korea during Past 10 Years

    To understand trends in the clinical characteristics of radiation oncology over the last 10 yr in Korea, annual survey questionnaires were sent to all of Korean radiation oncology facilities since 1990. Questionnaires addressed basic radiation therapy facilities and the clinical information. Responses were obtained from all facilities, and data collected from 1997 to 2006 was analyzed. The numbers of new patients that have undergone radiation therapy and the numbers of hospitals with a department of radiation oncology have steadily increased over the past 10 yr, and totaled 37,215 patients and 60 hospitals, respectively, in 2006. However, the proportion of patients irradiated among total cancer patients has remained below 30% over the last 10 yr. The numbers of prostate cancer, breast cancer, and hepatoma have increased by more than 3 fold over the past 10 yr. Moreover, the percentage of irradiated patients treated by brachytherapy was 10.3% in 1997, but this gradually fell to only 4.2% in 2006. The information collected described the role played by radiation oncology in Korea. Continuous surveys are required to enable trends to be detected

    The fusion rules and braiding statistics of anyons in (2+1)(2+1)D fermionic topological orders are characterized by the modular data of a super-modular category. On the other hand, the modular data of a super-modular category form a congruence representation of the Γθ\Gamma_\theta subgroup of the modular group SL2(Z)\mathrm{SL}_2(\mathbb{Z}). We provide a method to classify the modular data of super-modular categories by first obtaining the congruence representations of Γθ\Gamma_\theta and then building candidate modular data out of those representations. We carry out this classification up to rank 1010. We obtain both unitary and non-unitary modular data, including all previously known unitary modular data, and also discover new classes of modular data of rank 1010. We also determine the central charges of all these modular data, without explicitly computing their modular extensions.Comment: 32 pages, 2 figures, 6 table
