    The purpose of this study was to present metadata elements for managing biomechanical data of winter sports experiments and to establish an Integrated Data Management System with the presented metadata elements. To sort and select metadata elements, metadata crosswalk was done with four internationally used metadata schema; Dublin Core, OECD, DataCite, MODS. And a new metadata schema was designed with common elements from the crosswalk and the metadata elements drawn from general experimental designs and R&D portal. The schema was applied to a research data platform Datanest and the Integrated Management System for experimental data of winter sports was established

    Effect of extraction solvents on antioxidant and skin-whitening potentials of defatted Camellia seed cakes

    Defatted Camellia japonica L. seed cake is an important byproduct during the manufacture of Camellia seed oil. The present study evaluated the influence of two extraction solvents on the total contents of phenol and flavonoid, antioxidant activity and skin-whitening effect capable of inhibiting the biosynthesis of melanin of defatted Camellia seed cakes, a byproduct from Camellia oil production. The antioxidant capacities of 100% methanol and 70% ethanol extracts were analysed using radical scavenging (1,1-diphenyl-2-picrylhydrazyl, O2-, H2O2 and NO), SOD-like, ferrous ion chelating and reducing power assays. The total phenolic and flavonoid contents were further determined by the Folin-Ciocalteu method. Moreover, intracellular antityrosinase activity and melanin contents were evaluated in human malignant melanoma cells (SK mel-100). Ethanol extracts of defatted Camellia seed cake extracts exhibited higher phenolic (4097 mg gallic acid equivalents/100 g) and flavonoid (2899 mg rutin equivalents/100 g) contents with higher superoxide (IC50 = 1.9 mg/mL), nitric oxide (IC50 =1.6 mg/mL) radical scavenging, ferrous ion chelating (IC50 = 2.9 mg/mL) and reducing power (IC50 = 1.8 mg/mL) activities than those of methanol. These ethanol extracts also evidenced more effective inhibitory activities of tyrosinase and melanin synthesis than methanol extracts. Therefore, the present results demonstrated that defatted Camellia seed cakes could be a valuable source of antioxidative and whitening ingredients, and ethanol was more efficient in extracting antioxidants and bioactive compounds than methanol

    NKT cells promote antibody-induced joint inflammation by suppressing transforming growth factor β1 production

    Although NKT cells has been known to exert protective roles in the development of autoimmune diseases, the functional roles of NKT cells in the downstream events of antibody-induced joint inflammation remain unknown. Thus, we explored the functional roles of NKT cells in antibody-induced arthritis using the K/BxN serum transfer model. NKT cell–deficient mice were resistant to the development of arthritis, and wild-type mice administrated with α-galactosyl ceramide, a potent NKT cell activator, aggravated arthritis. In CD1d−/− mice, transforming growth factor (TGF)-β1 was found to be elevated in joint tissues, and the blockade of TGF-β1 using neutralizing monoclonal antibodies restored arthritis. The administration of recombinant TGF-β1 into C57BL/6 mice reduced joint inflammation. Moreover, the adoptive transfer of NKT cells into CD1d−/− mice restored arthritis and reduced TGF-β1 production. In vitro assay demonstrated that interleukin (IL)-4 and interferon (IFN)-γ were involved in suppressing TGF-β1 production in joint cells. The adoptive transfer of NKT cells from IL-4−/− or IFN-γ−/− mice did not reverse arthritis and TGF-β1 production in CD1d−/− mice. In conclusion, NKT cells producing IL-4 and IFN-γ play a role in immune complex–induced joint inflammation by regulating TGF-β1

    Postmenopausal osteoporosis is characterized by low bone density which leads to increased bone fragility and greater susceptibility to fracture. Current treatments for osteoporosis are dominated by drugs that inhibit bone resorption although they also suppress bone formation that may contribute to pathogenesis of osteonecrosis. To restore the extensive bone loss, there is a great need for anabolic treatments that induce osteoblasts to build new bone. Pre-osteoblastic cells produce proteins of the extra-cellular matrix, including type I collagen at first, and then to successively produce alkaline phosphatase (ALP) and osteocalcin during differentiation to osteoblasts. Finally, osteoblasts deposit calcium. Present study investigated the effects of egg yolk peptide (EYP) on osteogenic activities and bone matrix gene expressions in human osteoblastic MG-63 cells. The effects of EYP on cell proliferation, alkaline phosphatase (ALP) activity, collagen synthesis, and mineralization were measured. The expression of osteogenic genes including COL1A1 (collagen, type I, alpha 1), ALP, BGLAP (osteocalcin), and SPP1 (secreted phosphoprotein 1, osteopontin) were measured by quantitative realtime PCR. EYP dose-dependently increased MG-63 cell proliferation, ALP activity, collagen synthesis, and calcium deposition. Furthermore, COL1A1, ALP, and SPP1 gene expressions were increased by EYP treatment. Present study suggested that EYP treatment enhanced osteogenic activities and increased bone matrix osteogenicgenes. These results could provide a mechanistic explanation for the bone-strengthening effects of EYP

We selected data fields to design database schema considering which fields were contributing to the victory. We also sorted all fields into six tables to reflect the elements of ice hockey games; Game Information, Team Information, Offensice Stats, Defence Stats, Face-off Stats, Time On Ice Stats. This study was a prior research designing database schema in which the analysis of ice hockey official record is used to advance from statistical models to data science techniques.     IRT5 Probiotics Changes Immune Modulatory Protein Expression in the Extraorbital Lacrimal Glands of an Autoimmune Dry Eye Mouse Model

    PURPOSE. While the association between the gut microbiome and the immune system has been studied in autoimmune disorders, little is known about ocular disease. Previously we reported that IRT5, a mixture of five probiotic strains, could suppress autoimmune dry eye. In this study, we investigated the mechanism by which IRT5 performs its immunomodulatory function in a mouse model of autoimmune dry eye. METHODS. NOD.B10.H2b mice were used as an autoimmune dry eye model. Either IRT5 or PBS was gavaged orally for 3 weeks, with or without 5 days of antibiotic pretreatment. The effects on clinical features, extraorbital lacrimal gland and spleen proteins, and fecal microbiota were analyzed. RESULTS. The ocular staining score was lower, and tear secretion was higher, in the IRT5-treated groups than in the PBS-treated groups. After IRT5 treatment, the downregulated lacrimal gland proteins were enriched in the biological processes of defense response and immune system process. The relative abundances of 33 operational taxonomic units were higher, and 53 were lower, in the feces of the IRT5-treated groups than in those of the PBS-treated groups. IRT5 administration without antibiotic pretreatment also showed immunomodulatory functions with increases in the Lactobacillus helveticus group and Lactobacillus hamsteri. Additional proteomic assays revealed a decrease of proteins related to antigen-presenting processes in the CD11b(+) and CD11c(+) cells of spleen in the IRT5-treated groups. CONCLUSIONS. Changes in the gut microbiome after IRT5 treatment improved clinical manifestations in the autoimmune dry eye model via the downregulation of antigen-presenting processes in immune networks.11Ysciescopu
