    Ground-Based 1U CubeSat Robotic Assembly Demonstration

    Key gaps limiting in-space assembly of small satellites are (1) the lack of standardization of electromechanical CubeSat components for compatibility with commercial robotic assembly hardware, and (2) testing and modifying commercial robotic assembly hardware suitable for small satellite assembly for space operation. Working toward gap (1), the lack of standardization of CubeSat components for compatibility with commercial robotic assembly hardware, we have developed a ground-based robotic assembly of a 1U CubeSat using modular components and Commercial-Off-The-Shelf (COTS) robot arms without humans-in-the-loop. Two 16 in x 7 in x 7 in dexterous robot arms, weighing 2 kg each, are shown to work together to grasp and assemble CubeSat components into a 1U CubeSat. Addressing gap (2) in this work, solutions for adapting power-efficient COTS robot arms to assemble highly-capable CubeSats are examined. Lessons learned on thermal and power considerations for overheated motors and positioning errors were also encountered and resolved. We find that COTS robot arms with sustained throughput and processing efficiency have the potential to be cost-effective for future space missions.     Distinct mechanoreceptor pezo-1 isoforms modulate food intake in the nematode Caenorhabditis elegans

    Two PIEZO mechanosensitive cation channels, PIEZO1 and PIEZO2, have been identified in mammals, where they are involved in numerous sensory processes. While structurally similar, PIEZO channels are expressed in distinct tissues and exhibit unique properties. How different PIEZOs transduce force, how their transduction mechanism varies, and how their unique properties match the functional needs of the tissues they are expressed in remain all-important unanswered questions. The nematode Caenorhabditis elegans has a single PIEZO ortholog (pezo-1) predicted to have 12 isoforms. These isoforms share many transmembrane domains but differ in those that distinguish PIEZO1 and PIEZO2 in mammals. We used transcriptional and translational reporters to show that putative promoter sequences immediately upstream of the start codon of long pezo-1 isoforms predominantly drive green fluorescent protein (GFP) expression in mesodermally derived tissues (such as muscle and glands). In contrast, sequences upstream of shorter pezo-1 isoforms resulted in GFP expression primarily in neurons. Putative promoters upstream of different isoforms drove GFP expression in different cells of the same organs of the digestive system. The observed unique pattern of complementary expression suggests that different isoforms could possess distinct functions within these organs. We used mutant analysis to show that pharyngeal muscles and glands require long pezo-1 isoforms to respond appropriately to the presence of food. The number of pezo-1 isoforms in C. elegans, their putative differential pattern of expression, and roles in experimentally tractable processes make this an attractive system to investigate the molecular basis for functional differences between members of the PIEZO family of mechanoreceptors.Fil: Hughes, Kiley.     Designer Lipid-Like Peptides: A Class of Detergents for Studying Functional Olfactory Receptors Using Commercial Cell-Free Systems

    A crucial bottleneck in membrane protein studies, particularly G-protein coupled receptors, is the notorious difficulty of finding an optimal detergent that can solubilize them and maintain their stability and function. Here we report rapid production of 12 unique mammalian olfactory receptors using short designer lipid-like peptides as detergents. The peptides were able to solubilize and stabilize each receptor. Circular dichroism showed that the purified olfactory receptors had alpha-helical secondary structures. Microscale thermophoresis suggested that the receptors were functional and bound their odorants. Blot intensity measurements indicated that milligram quantities of each olfactory receptor could be produced with at least one peptide detergent. The peptide detergents' capability was comparable to that of the detergent Brij-35. The ability of 10 peptide detergents to functionally solubilize 12 olfactory receptors demonstrates their usefulness as a new class of detergents for olfactory receptors, and possibly other G-protein coupled receptors and membrane proteins.United States.     An open toolkit for tracking open science partnership implementation and impact.

    Serious concerns about the way research is organized collectively are increasingly being raised. They include the escalating costs of research and lower research productivity, low public trust in researchers to report the truth, lack of diversity, poor community engagement, ethical concerns over research practices, and irreproducibility. Open science (OS) collaborations comprise of a set of practices including open access publication, open data sharing and the absence of restrictive intellectual property rights with which institutions, firms, governments and communities are experimenting in order to overcome these concerns. We gathered two groups of international representatives from a large variety of stakeholders to construct a toolkit to guide and facilitate data collection about OS and non-OS collaborations. Ultimately, the toolkit will be used to assess and study the impact of OS collaborations on research and innovation. The toolkit contains the following four elements: 1) an annual report form of quantitative data to be completed by OS partnership administrators; 2) a series of semi-structured interview guides of stakeholders; 3) a survey form of participants in OS collaborations; and 4) a set of other quantitative measures best collected by other organizations, such as research foundations and governmental or intergovernmental agencies. We opened our toolkit to community comment and input. We present the resulting toolkit for use by government and philanthropic grantors, institutions, researchers and community organizations with the aim of measuring the implementation and impact of OS partnership across these organizations. We invite these and other stakeholders to not only measure, but to share the resulting data so that social scientists and policy makers can analyse the data across projects

    Understanding Spray Attributes of Commercial UAAS as Impacted by Operational and Design Parameters

    Unmanned aerial application systems (UAAS) have recently gained momentum for the application of crop protection (CP) products. Due to their high operational efficiency, mobility, and low cost, UAAS may be perceived as a more viable option for spray application when compared to conventional application techniques, especially backpack/knapsack sprayers. However, being a relatively new technology and not a common practice in the USA, there are no best management practices or guidelines for testing UAAS spray performance. Therefore, this study was undertaken to assess the impact of different attributes pertinent to UAAS flight (e.g., altitude, speed, etc.), application (e.g., droplet size, tank additive, etc.), and UAAS design (e.g., nozzle placement relative to a the rotor) on spray performance. The spray performance was evaluated in terms of swath and spray drift (ground and airborne) using water-sensitive papers (WSP) and mylar card/string samplers, respectively. The samplers were respectively analyzed using image processing and fluorometry techniques. The different treatments under study were UAAS type (MG-1P and AG V6A+), flight altitude (1.5, 2.5, and 4.0 m) and speed (2 and 3 ms−1), and nozzle type (XR11001 and XR8002 flat fan nozzles) with different droplet volume median diameter (VMD) at ~207 kPa (very fine [140 µm] and fine [196 µm], respectively). The results indicated a highly variable swath for both platform types. While nozzles under each rotor may be a common design in commercial UAAS, the results indicated that placement of nozzles out on a boom might have a less variable swath and a lower drift potential. In addition, flying slower, using a relatively larger droplet VMD (i.e., 196 µm at ~207 kPa) may reduce both ground and airborne drift. This study may serve as a guideline for growers/operators to evaluate their UAAS platforms and optimize key attributes pertinent to UAAS operation for effective spraying
