Scorpion peptides: Potential use for new drug development
Several peptides contained in scorpion fluids showed diverse array of biological activities with high specificities to their targeted sites. Many investigations outlined their potent effects against microbes and showed their potential to modulate various biological mechanisms that are involved in immune, nervous, cardiovascular, and neoplastic diseases. Because of their important structural and functional diversity, it is projected that scorpion-derived peptides could be used to develop new specific drugs. This review summarizes relevant findings improving their use as valuable tools for new drugs development
Oxidative stress and histopathological changes induced by methylthiophanate, a systemic fungicide, in blood, liver and kidney of adult rats
Background: Methyl-thiophanate (MT), a fungicide largely used in agriculture throughout the world including Tunisia, protects many vegetables, fruits and field crops against a wide spectrum of fungal diseases. Oxidative stress has been proposed as a possible mechanism involved in MT toxicity on non-target organism.Methods: In the present study, the effect of MT injected intraperitoneally to adult rats at 300 or 500 mg/kg of body weight was studied on blood, liver and kidney.Results: Our results showed 3 days after MT injection, a significant decrease in hemoglobin and hematocrit values. A disruption in total white blood cells and platelets also occurred. Accordingly, an increased in malondialdehyde, H2O2 and advanced oxidation protein levels in liver and kidney were noted with the two doses. A significant change in plasma biomarkers and organ enzymatic and non-enzymatic activities were observed after MT treatment. The modifications in biochemical parameters were substantiated by histopathological data.Conclusion: These data confirmed the pro-oxidant effects of this fungicide. Étude de l’allergie croisée entre les différents inhibiteurs de la pompe à protons
Bien que des réactions allergiques aux inhibiteurs de la pompe à protons (IPP) aient été
décrites par certains auteurs, la présence de réactions croisées entre les différentes
molécules est un sujet de discussion. Le but de notre travail est d’analyser tous les cas
de réactions cutanées allergiques aux IPP notifiés au service régional de
pharmacovigilance de Sfax durant une période de 12 ans allant du mois d’août 2000 au mois
de juillet 2012 et d’évaluer la possibilité de réactions croisées entre les différentes
molécules de cette classe. Une enquête de pharmacovigilance a été réalisée pour chaque cas
selon la méthode française d'imputabilité. Nous avons convoqué ensuite, tous les patients
qui ont développé une réaction allergique à un IPP avec une imputabilité plausible ou
vraisemblable. Un patch-test à toute les molécules a été réalisé afin d’étudier la
possibilité d’allergie croisée entre les IPP. Trente-sept patients ont développé des
réactions cutanées aux IPP durant une période de 12 ans, avec un total de 1 172
toxidermies, soit 3 % du total des toxidermies rapportées au centre de pharmacovigilance
de Sfax. La toxidermie la plus fréquemment observée était l’éruption maculo-papuleuse (19
cas soit 51 %), suivie de l’urticaire dans 9 cas (24 %). L’oméprazole était l’IPP le plus
incriminé (dans 31 cas soit 83,78 %). La prise ultérieure du lansoprazole a été faite chez
5 patients ayant développé une allergie à l’oméprazole et s’est déroulée sans incidents.
Les patch-tests ont Ă©tĂ© rĂ©alisĂ©s chez 6 patients ayant dĂ©veloppĂ© une rĂ©action cutanĂ©e Ă
l’oméprazole. La lecture de ces tests à 72 h a été positive à l’oméprazole dans tout les
cas, et négatifs au lansoprazole dans 5 cas. Dans un tiers des cas, le lansoprazole a été
une bonne alternative en cas d’allergie à l’oméprazole, à l’ésoméprazole ou au
pantoprazole. Dans 1 cas, nous avons contre-indiqué tous les IPP. Dans les autres cas, une
surveillance s’imposera lors de l’utilisation ultérieure du lansoprazole
Ageusia as a side effect of clopidogrel treatment
Clopidogrel is prescribed in cardiac and extracardiac vascular diseases. It is generally well tolerated; however, few cases of taste disorders have been reported. We present a case of clopidogrel-induced ageusia notified in Sfax pharmacovigilance center on March 13, 2009. A 46-year-old patient developed ageusia with decreased appetite five weeks after starting clopidogrel. Other etiologies including ear nose throat (ENT) examination were ruled out. Five months after reduction of clopidogrel dose, ageusia partially decreased. Clopidogrel was strongly suspected as a causal drug. According to the French imputation method, score of imputability was considered as plausible (C2S2) I2. Physiopathology of this side effect is not yet understood. However, it seems to be a reversible and dose-related event. Although it is not life-threatening, loss of taste can have significant effect on the quality of life of patients
Effets indésirables de la sulfasalazine : discussion de leur mécanisme et du rôle de la composante sulfamidée
La sulfasalazine est largement utilisée dans le traitement des maladies inflammatoires
chroniques de l’intestin (MICI) et de certaines affections rhumatologiques. Cependant, son
utilisation est associée à un taux élevé d’effets indésirables (EI). Ces effets peuvent
être cutanés, hématologiques, rénaux, hépatiques, digestifs ou neurologiques. L’objectif
de notre travail était de colliger tous les cas d’EI suspectés d’être liés à l’utilisation
de la sulfasalazine, chez des patients hospitalisés au service de gastroentérologie de
l’hôpital Hédi Chaker de Sfax (Tunisie) durant une période de 5 ans et de préciser la
fraction responsable de la réaction (mésalazine ou sulfapyridine). Notre population
d’étude a comporté 69 patients qui ont reçu la sulfasalazine pour le traitement d’une
MICI. Nous avons colligé, chez 23 patients (33 %), 25 EI suspects d’être liés à la
sulfasalazine. Ces EI sont surtout de type cutané et hématologique. L’administration
ultérieure de la mésalazine a été effectuée chez 15 patients. Elle a été bien tolérée chez
14 patients. Ainsi, la responsabilité de la fraction sulfamidée a été fortement suspectée
dans ces cas. Le mécanisme des EI induits par la sulfasalazine peut être toxique ou
immuno-allergique avec possibilité d’allergie croisée avec les autres sulfamides et/ou
salicylés
