Sıçanlarda Hareket Kısıtlama Stresinin Torasik Aortanın Yapı ve İşlevlerinde Oluşturduğu Glikasyon/Lipoksidasyon-Temelli Değişiklikler Üzerine Doksisiklinin Dozla İlişkili Etkisi
Doksisiklin matriks metalloproteinaz (MMP) enzimlerini inhibe eder. MMP‘ler ekstraselüler matrikste çeşitli proteinlerin yıkımına yol açan, çeşitli hastalık ve durumlarda etkili rolleri olan enzimlerdir. Doksisiklin aynı zamanda sistemik ve vasküler oksidatif stresi de azaltır.
Doksisiklinin yüksek kan glikozunun protein, lipoprotein ve/veya nükleik asitlerle non-enzimatik glikasyonunu engelleyerek ileri glikasyon son ürünleri (AGE; Advanced Glycation Endproducts)‘nin oluşumunu azaltır. Karboksimetillizin (CML) AGE ve ileri lipoperoksidasyon son ürünleri (ALE) yolağının ortak son ürünlerindendir. CML oluşumunun, diyabet (DM), yaşlanma, hiperlipidemi, sigara içme, hipertansiyon, inflamasyon, Alzheimer gibi hastalıklarla da ilişkili olduğu bilinmektedir.
Doksisiklin hücreyi stres ve apoptozise karşı korur.
Stres hormonları hiperglisemik hormonlardır. Hiperglisemi oksidatif stresi tetikler. Biyokimyasal kökenli olan tüm stresler gibi davranışsal stres de oksidan hasara sebep olur. Hareket kısıtlama stresi dislipidemi, karbonhidrat metabolizmasında bozukluk, nitrik oksit (NO) üretiminde azalma, ateroskleroz ve antioksidan durumda dengesizlik sonucu oksidan hasara yol açar.
Kronik strese maruziyetin de oksidatif stresi artırarak lipit peroksidasyonunu artırması, yol açacağı yüksek kan şekeri düzeyleriyle protein glikasyonunu artırması ve sonuçta AGE ve ALE düzeylerini yükseltmesi beklenir. Bu durumda, doksisiklin, hareket kısıtlaması stresine maruz kalmış sıçanlarda oksidatif hasara ve protein glikasyonuna karşı koruyucu olacaktır.
Bu çalışma, davranışsal stresin, kardiyovasküler sistemde ileri glikasyon ve lipoperoksidasyon son ürünlerinin birikimi ile sonuçlanıp sonuçlanmadığını yapısal ve işlevsel olarak ortaya koymak ve bu süreçte doksisiklinin doza bağımlı koruyucu etkisini belirlemek amacı ile 48 adet erkek Sprague Dawley sıçan üzerinde gerçekleştirildi. Sıçanların vücut ağırlıkları (VA), kan şekeri ve HbA1c düzeyleri değerlendirildi. İzole torasik aorta preperatları endotel ve düz kas işlevleri açısından izole organ banyosu sisteminde değerlendirildi. MMP aktiviteleri jelatin zimografi yöntemi ile ölçüldü. Oksidan/anti-oksidan durum belirteci olarak glutatyon ve malondialdehit derişimleri

saptandı. İmmunohistokimyasal boyama ile AGE/ALE varlığı, apoptozis, anjiyogenez değerlendirildi. Uygulamaların davranışlar üzerine etkisini belirlemek için Açık Alan ve
Yükseltilmiş Artı Labirent testleri uygulandı.
Kronik stres uygulaması VA kaybına yol açtı. Akut stres kan şekeri değerlerini artırdı. Yapılan uygulamalar, glikasyonu artırmamaları nedeniyle HbA1c değerlerini değiştirmedi. Kronik stres, sıçan izole torasik aortalarında endotel hasarı yanında düz kasta gevşemeyi artırıcı bir etki yarattı. Bu etki üzerinde doksisiklin 15 mg/kg/gün dozunda koruyucu olurken, 30 mg/kg/gün dozunda ise stres ile benzer etkiler oluşturdu. Doksisiklin 30 mg/kg/gün uygulaması genel olarak stres ile aynı yönde etki gösterdi. Düşük doz doksisiklinin anti-oksidan, yüksek doz doksisiklinin ise oksidan etki eğiliminde olduğu saptandı. Kronik stres proMMP-2 ve MMP-9 aktivitelerini artırdı. Doksisiklinin her iki dozu MMP-2 aktivitesini inhibe etti. Yapılan davranış testleri ile stres uygulamalarının anksiyete eğilimini artırdığı 15 mg/kg/gün dozunda doksisiklin uygulamasının ise anksiyeteye karşı koruyucu eğilim oluşturduğu saptandı

Investigation of the Possible Protective Effects of Ketamine and Dantrolene on the Hippocampal Apoptosis and Spatial Learning in Rats Exposed to Repeated Electroconvulsive Seizures as a Model of Status Epilepticus
AIM: To evaluate the possible neuroprotective effects of ketamine and
dantrolene on the hippocampal apoptosis and spatial learning in rats
exposed to repeated electroconvulsive seizures (ECS) as a model of
status epilepticus (SE).
MATERIAL and METHODS: Twenty-four rats were assigned to 4 groups. 1st
Group was Sham. 2nd Group was ECS: ECS was induced by ear electrodes via
electrical stimulation. The same ECS protocol was applied to the 3th and
4th Groups which received ketamine (40 mg/kg s.c.) or dantrolene (5
mg/kg i.p.) 1 h before each ECS, respectively. Following 30 days of
recovery, the cognitive status of the animals was evaluated via Morris
Water Maze (MWM). The same experimental protocol was repeated 14 days
afterward to evaluate the retention of the memory. Hippocampal apoptosis
was examined in corresponding experimental groups.
RESULTS: All the animals in four groups learned the task with no
significant difference between groups in MWM. The ECS+ketamine group
showed memory impairment 14 days afterward. ECS+dantrolene group was not
different from controls. ECS caused long term apoptotic processes in
dentate gyrus (DG) and non-apoptotic neuronal injury in CA1 and CA2.
CONCLUSION: Dantrolene and ketamine inhibited apoptosis and showed
neuroprotective effects. Although ketamine and dantrolene inhibited
ECS-induced apoptosis and non-apoptotic injury, they did not produce
similar effects on memory retention. It will be warranted to evaluate
cognitive dysfunction by taking into consideration the other factors in
addition to apoptosis and neurodegenerative changes
