    Development of a high-throughput field phenotyping rover optimized for size-limited breeding fields as open-source hardware

    Phenotyping is a critical process in plant breeding, especially when there is an increasing demand for streamlining a selection process in a breeding program. Since manual phenotyping has limited efficiency, highthroughput phenotyping methods are recently popularized owing to progress in sensor and image processing technologies. However, in a size-limited breeding field, which is common in Japan and other Asian countries, it is challenging to introduce large machinery in the field or fly unmanned aerial vehicles over the field. In this study, we developed a ground-based high-throughput field phenotyping rover that could be easily introduced to a field regardless of the scale and location of the field even without special facilities. We also made the field rover open-source hardware, making its system available to public for easy modification, so that anyone can build one for their own use at a low cost. The trial run of the field rover revealed that it allowed the collection of detailed remote-sensing images of plants and quantitative analyses based on the images. The results suggest that the field rover developed in this study could allow efficient phenotyping of plants especially in a small breeding field

    Evolution of two-step structural phase transition in Fe1+dTe detected by low-temperature x-ray diffraction

    The low-temperature crystal structure of Fe1.13Te, which exhibits an anomalous two-step magnetic transition, was clarified by the systematic x-ray diffraction measurements. It was found that two-step structural phase transition, tetragonal-orthorhombic-monoclinic, occurred correspondingly to the two-step magnetic transition. The detailed analysis of the profile at 5 K indicated the coexistence of the minor orthorhombic area inside the major monoclinic lattice, which could explain the lower-shift (suppression) of the antiferromagnetic transition temperature in Fe1.13Te and suggest a possibility of superconductivity at the domain boundary.Comment: 12 pages, 3 figure

    Effects of Dietary Supplementation With Enterococcus faecium and Clostridium butyricum, Either Alone or in Combination, on Growth and Fecal Microbiota Composition of Post-weaning Pigs at a Commercial Farm

    Lactic acid bacteria (LAB) and butyric acid bacteria (BAB) are commonly used as probiotics in swine production. However, their combined effect on post-weaning pigs has not been assessed. Therefore, here we investigated the individual and combined efficacy of dietary Enterococcus faecium and Clostridium butyricum on the growth and gut microbiota of post-weaning pigs at a commercial farm. Four independent trials were conducted, in each of which five pens containing 10 pigs were assigned to one of five treatments: C, basal diet; L, basal diet + live E. faecium; D, basal diet + heat-killed E. faecium; M, basal diet + C. butyricum; or L+M, basal diet + live E. faecium + C. butyricum. Each trial was conducted over a 90-day period that was divided into two phases (Phase 1, days 0–40 post-weaning; and Phase 2, days 40–90 post-weaning), with the probiotics being supplemented only during Phase 1. Ten pigs in each pen were used for body weight (BW) analysis and fecal samples were collected from five or six of these pigs. In addition, the fecal samples from one randomly selected trial were used for gut microbiota analysis. We found that pigs in the L, D, and L+M treatment groups had a significantly higher BW than those in C (p < 0.05) but pigs in the L+M treatment group had a similar BW to those in the L and M groups. Furthermore, there were no significant differences in alpha diversity among the treatments but the beta diversity (weighted UniFrac distances) showed distinct clustering patterns, with pigs in C having discrete microbiota from those in all of the probiotics treatment groups except D (C vs. L, q = 0.04; C vs. M, q = 0.06; C vs. L+M, q = 0.06). These findings indicate that dietary supplementation with live or heat-killed E. faecium enhances growth performance in pigs but there is no synergistic effect when E. faecium is used in combination with C. butyricum. Furthermore, the addition of live E. faecium and C. butyricum to the diet of pigs may change the structure of the gut microbiota

    Finishing the euchromatic sequence of the human genome

    The sequence of the human genome encodes the genetic instructions for human physiology, as well as rich information about human evolution. In 2001, the International Human Genome Sequencing Consortium reported a draft sequence of the euchromatic portion of the human genome. Since then, the international collaboration has worked to convert this draft into a genome sequence with high accuracy and nearly complete coverage. Here, we report the result of this finishing process. The current genome sequence (Build 35) contains 2.85 billion nucleotides interrupted by only 341 gaps. It covers ∼99% of the euchromatic genome and is accurate to an error rate of ∼1 event per 100,000 bases. Many of the remaining euchromatic gaps are associated with segmental duplications and will require focused work with new methods. The near-complete sequence, the first for a vertebrate, greatly improves the precision of biological analyses of the human genome including studies of gene number, birth and death. Notably, the human enome seems to encode only 20,000-25,000 protein-coding genes. The genome sequence reported here should serve as a firm foundation for biomedical research in the decades ahead

    New Traffic Conflict Measure Based on a Potential Outcome Model

    A key issue in the analysis of traffic accidents is to quantify the effectiveness of a given evasive action taken by a driver to avoid crashing. Since 1977, the widely accepted definition for this effectiveness measure, which is called traffic conflict, has been “the risk of a collision if the driver movement remains unchanged.” Although the definition is expressed counterfactually, the full power of counterfactual analysis was not utilized. In this paper, we propose a counterfactual measure of traffic conflict called Counterfactual Based Conflict (CBC). The CBC is interpreted as the probability that a driver avoided a crash actually by taking an evasive action in the counterfactual situation in which the crash would have occurred if he/she had not taken an evasive action and the crash would not have occurred if he/she had taken an evasive action. The CBC captures realistic aspects of the traffic situation, and lends itself to modern causal analysis. In addition, we provide some of identification conditions for the CBC. Furthermore, we formulate bounds on the CBC when the proposed identification conditions are violated. Finally, through an application of the CBC to the 100-Car Naturalistic Driving Study, we discuss the usefulness and limitations of the proposed measure

    Influence of environmental parameters on habitat use by sympatric freshwater eels, Anguilla marmorata and A. japonica on Yakushima Island, Japan

    It is known that environmental parameters, such as water depth, size of substrate material, and current velocity influence the organization of stream fish communities. However, few studies have investigated the effects of these parameters on sympatric freshwater eels. Here, the habitat use of two anguillid eels, Anguilla marmorata Quoy and Gaimard, 1824 and A. japonica Temminck and Schlegel, 1846, coexisting in the same river systems in Japan was investigated. It was found that the tropical species A. marmorata exclusively utilized habitats with larger substrate materials, fewer fallen leaves and less leaf detritus, higher current velocity, and lower turbidity than the temperate species A. japonica, and the distributions of the two anguillid eels were clearly segregated. Thus, when both species co-occur in the same river systems, clear flowing mainstems and tributaries are preferred habitats for A. marmorata, while stagnant muddy estuaries, backwater areas, irrigation channels, and reservoirs are preferred habitats for A. japonica. These habitat segregations were consistent through body-size increases and life-stage developments from elver to yellow eel and did not show ontogenetic shifts. The findings indicated that both species had strong habitat preferences, and these environmental parameters must be considered in the conservation of anguillid eels in sympatric zones.The accepted manuscript in pdf format is listed with the files at the bottom of this page.     Evaluation of Rate of Adhesion of Lactobacillus namurensis Strain GYP-74 to Porous Fine Ceramics

    This study aimed to evaluate the ratio of adherence of lactic acid bacteria (LAB) to porous fine ceramics in order to develop a novel LAB-rich pickle container for the production of functional fermented vegetables. Some LAB were isolated from the salted rice bran used for pickling (Nukadoko in Japanese). These isolates were classified in Lactobacillus namurensis by phylogenetic analysis. Some pottery-shard (PS) samples were prepared by varying the mixing rate of polyacetal (POM) resin to clay (0–30% (v/v)) and the burning temperature (1000 °C or 1100 °C). A test of the adherence of strain GYP-74 to the PSs was performed. The results showed that the adherence rate was significantly higher in the PSs burned at 1100 °C as compared with those burned at 1000 °C. A pore distribution analysis showed that pore sizes of less than a few µm and pore sizes in the range of a few µm to a few hundred µm were mainly distributed in the PSs without and with POM, respectively. X-ray diffraction analysis showed that both PSs with and without POM contained quartz and hematite. The PSs burned at 1000 °C and 1100 °C specifically contained microcline and mullite, respectively.     Pars plana vitrectomy combined with penetrating keratoplasty and transscleral-sutured intraocular lens implantation in complex eyes: a case series

    Background The aim of this study was to evaluate the clinical outcomes of pars plana vitrectomy (PPV) combined with penetrating keratoplasty (PKP) and transscleral-sutured intraocular lens (IOL) implantation (IOL-suture) in complex eyes. Methods In this prospective, consecutive interventional case series, patients who underwent PKP combined with PPV and IOL implantation from July 2014 to March 2018 at Yokohama Minami Kyosai Hospital were enrolled. The postoperative best corrected visual acuity (BCVA) (converted to logarithm of the minimal angle of resolution [logMAR] units), intraocular pressure (IOP, mmHg), endothelial cell density (ECD, cells/mm(2)), graft survival, complications, astigmatism, and spherical equivalent (dioptres [D]) were evaluated. Results This study included 11 eyes of 11 patients (three females and eight males; mean age, 61.8 +/- 13.9 years) with an injury (n = 6) or bullous keratopathy (n = 5). The BCVA significantly improved from 1.50 +/- 0.66 logMAR preoperatively to 0.78 +/- 0.59 logMAR (p < 0.001) postoperatively. The baseline ECD significantly decreased from 2396 +/- 238 cells/mm(2)preoperatively to 1132 +/- 323 cells/mm(2)(p < 0.001) postoperatively. Despite two rejection episodes, graft survival rates were 100%. The mean follow-up period was 38.0 +/- 20.5 months. Two patients required combined glaucoma surgery, and three patients underwent subsequent glaucoma surgery. Postoperative astigmatism and spherical equivalent were 3.9 +/- 3.2 D and 0.29 +/- 2.18 D, respectively. Conclusion The combination of PKP, PPV, and IOL-suture implantation could be a safe and effective approach for eyes requiring anterior segment surgery; however, these eyes are associated with a higher incidence of glaucoma surgery
