    Two dimensional electrophysiological characterization of human pluripotent stem cell-derived cardiomyocyte system.

    Stem cell-derived cardiomyocytes provide a promising tool for human developmental biology, regenerative therapies, disease modeling, and drug discovery. As human pluripotent stem cell-derived cardiomyocytes remain functionally fetal-type, close monitoring of electrophysiological maturation is critical for their further application to biology and translation. However, to date, electrophysiological analyses of stem cell-derived cardiomyocytes has largely been limited by biologically undefined factors including 3D nature of embryoid body, sera from animals, and the feeder cells isolated from mouse. Large variability in the aforementioned systems leads to uncontrollable and irreproducible results, making conclusive studies difficult. In this report, a chemically-defined differentiation regimen and a monolayer cell culture technique was combined with multielectrode arrays for accurate, real-time, and flexible measurement of electrophysiological parameters in translation-ready human cardiomyocytes. Consistent with their natural counterpart, amplitude and dV/dtmax of field potential progressively increased during the course of maturation. Monolayer culture allowed for the identification of pacemaking cells using the multielectrode array platform and thereby the estimation of conduction velocity, which gradually increased during the differentiation of cardiomyocytes. Thus, the electrophysiological maturation of the human pluripotent stem cell-derived cardiomyocytes in our system recapitulates in vivo development. This system provides a versatile biological tool to analyze human heart development, disease mechanisms, and the efficacy/toxicity of chemicals

    Extreme Peripheral Blood Plasmacytosis Mimicking Plasma Cell Leukemia as a Presenting Feature of Angioimmunoblastic T-Cell Lymphoma (AITL).

    Angioimmunoblastic T-cell lymphoma (AITL) is one of four major subtypes of nodal peripheral T cell lymphoma, characterized by its cell of origin, the follicular helper T-cell (TFH). Patients typically present with prominent constitutional (B) symptoms, generalized lymphadenopathy, hepatosplenomegaly, cytopenias, and rash. Here we present a case of a 62-year-old male with progressive cervical adenopathy, fevers and weight loss presenting with extreme polyclonal plasmacytosis and high plasma EBV viral load. While the initial presentation appeared to mimic plasma cell leukemia or severe infection, lymph node biopsy and bone marrow biopsy confirmed a diagnosis of AITL. This case highlights the heterogeneity of the clinical presentation of AITL to enable physicians to more promptly recognize, diagnose and initiate treatment

    Direct measurement of B(D_s^+ → φX^+)

    The absolute inclusive branching fraction of D_s^+→φX^+ has been measured from data collected by the BES detector at a center-of-mass energy of 4.03 GeV, corresponding to an integrated luminosity of 22.3 pb^(-1). At this energy, direct pair production e^+e^-→D_s^+D_s^- has been observed. We have selected D_s candidate events by reconstructing five hadronic decay modes D_s^+→φπ^+, K^(0*)K^+, K^0K^+, f0^(π+) and K^0K^-π^+π^+ and have searched for inclusive φ’s in the recoiling D_s^-. We observed three recoiling φ’s in the 166.4 ± 31.8 D_s candidate events, which leads to the absolute branching fraction B(D_s^+→φX^+)=(17.8(-7.2 -6.3)^(+15.1+0.6)) % and B(D_s-6.3+→φπ-6.3+)=(3.6_(-1.6 -1.3)(^_3.1+0.4) %.     Measurement of the mass of the Ï„ lepton

    The mass of the τ lepton has been measured at the Beijing Electron-Positron Collider using the Beijing Spectrometer. A search near threshold for e^+e^-→τ^+τ^- was performed. Candidate events were identified by requiring that one τ decay via τ→eνν¯, and the other via τ→μνν¯. The mass value, obtained from a fit to the energy dependence of the τ^+τ^- cross section, is m_τ=1776.9_(-0.5)^(+0.4)±0.2 MeV

    Energy Expenditure Overestimation Bias in Elliptical Trainer Machine

    Elliptical trainers are a common mode of aerobic exercise in recreationally active populations. Those with a weight loss goal might rely upon the energy expenditure (EE) estimation that many elliptical brands provide to keep track of calories (kcals) burned and make nutritional decisions. For this reason, it is important to evaluate the accuracy of the algorithms used by elliptical trainers to estimate EE. The purpose of this study was to compare EE estimates by a common brand of elliptical trainer to that measured using open circuit spirometry, at different combinations of resistance and pedal speed. Twenty subjects (10 male, 10 female; 34 ± 12 yr; 175.3 ± 10.7 cm; 77.1 ± 14.1 kg) consented to participate. Each completed three 15-min bouts of elliptical exercise on the same elliptical trainer, with at least 24 hr between exercise bouts. Pedal rates were held constant throughout each bout at 50, 60, or 70 RPM, and resistance was increased incrementally every 5 min from level 5 to 10 to 15. The different cadences were completed in a randomized order between participants. Expired gases were collected continuously throughout the 15 min. Heart rate, distance (mi), and EE from the elliptical readout were recorded every 1 min. RPE was collected twice per resistance level. A two-tailed paired samples t-test was used to compare elliptical EE to measured EE. A linear regression model was used to evaluate the ability of the elliptical EE to predict measured EE. Significance for all statistical measures was held at an alpha level of 0.05. The difference between EE estimates from the elliptical and measured VO2 was significant (p Measured EE = 0.95*(Elliptical EE) – 3.161 In conclusion, the elliptical trainer used for this study demonstrated a bias to overestimate EE. This should be taken into account by health/fitness professionals using these estimations to program for clients. There may be some variation in the EE correction regression depending on elliptical model, and proper machine calibration should be ensured
