    Using the Xenopus Developmental Eye Regrowth Stystem to Distinguish the Role of Developmental Versus Regenerative Mechanisms

    A longstanding challenge in regeneration biology is to understand the role of developmental mechanisms in restoring lost or damaged tissues and organs. As these body structures were built during embryogenesis, it is not surprising that a number of developmental mechanisms are also active during regeneration. However, it remains unclear whether developmental mechanisms act similarly or differently during regeneration as compared to development. Since regeneration is studied in the context of mature, differentiated tissues, it is difficult to evaluate comparative studies with developmental processes due to the latter’s highly proliferative environment. We have taken a more direct approach to study regeneration in a developmental context (regrowth). Xenopus laevis, the African clawed frog, is a well-established model for both embryology and regeneration studies, especially for the eye. Xenopus eye development is well-defined. Xenopus is also an established model for retinal and lens regeneration studies. Previously, we demonstrated that Xenopus tailbud embryo can successfully regrow a functional eye that is morphologically indistinguishable from an age-matched control eye. In this study, we assessed the temporal regulation of retinal differentiation and patterning restoration during eye regrowth. Our findings showed that during regrowth, cellular patterning and retinal layer formation was delayed by approximately 1 day but was restored by 3 days when compared to eye development. An assessment of the differentiation of ganglion cells, photoreceptor cells, and Müller glia indicated that the retinal birth order generated during regrowth was consistent with that observed for eye development. Thus, retina differentiation and patterning during regrowth is similar to endogenous eye development. We used this eye regrowth model to assess the role of known mechanisms in development versus regrowth. Loss-of-function studies showed that Pax6 was required for both eye development and regrowth whereas apoptosis was only required for regrowth. Together, these results revealed that the mechanisms required for both development and regrowth can be distinguished from regrowth-specific ones. Our study highlights this developmental model of eye regrowth as a robust platform to systematically and efficiently define the molecular mechanisms that are required for regeneration versus development

    Crisis, Shell-Shock, and the Temporality of Trauma: Cultural Memory and the Great War Combatant Experience in Owen, Graves, and Barker

    The year 2014 will mark the centennial of the outbreak of World War I in August 1914. This historic anniversary will likely provoke several discussions from all fields in the humanities concerning the Great War\u27s significance on contemporary culture through history, visual art, and in the case of this essay: literature. In light of this event, any serious discussion among scholars should undeniably begin with how the war continues to be represented today through a thorough, contemporary analysis of its many key literary texts. This essay will examine, in this regard, how past and contemporary discourses in literary theory-primarily concerned with how an individual combatant subject attempts to construct and understand their own traumatic experiences through poetic and literary discourse-can continue to incite discussion on why literature of the Great War and its influential role in defining how it has come to be understood in our cultural memory remains relevant even today. Under the guiding influence of Paul Fussell\u27s classic The Great War and Modern Memory, I will discuss how three important works-a poetry collection, a memoir, and a modern work of historical fiction-all contribute to how the war has become represented as a tragic rupture in history that reversed the idea of human progress and left an entire generation disillusioned in its aftermath, regardless of the historical veracity of this legacy. The texts I will be examining include: select poems of Wilfred Owen, Goodbye to All That by Robert Graves, and Regeneration by Pat Barker. In addition to this, I will conclude with an analysis of how a contemporary reading of these texts can contribute to a larger discussion of the crisis of historicity in our current post-modern cultural landscape

    Blood Glucose Levels and Performance in a Sports Camp for Adolescents with Type 1 Diabetes Mellitus: A Field Study

    Background. Acute hypo- and hyperglycemia causes cognitive and psychomotor impairment in individuals with type 1 diabetes mellitus (T1DM) that may affect sports performance. Objective. To quantify the effect of concurrent and antecedent blood glucose concentrations on sports skills and cognitive performance in youth with T1DM attending a sports camp. Design/Methods. 28 youth (ages 6–17 years) attending a sports camp carried out multiple skill-based tests (tennis, basketball, or soccer skills) with glucose monitoring over 4 days. Glucose levels at the time of testing were categorized as (a) hypoglycemic (<3.6 mM); (b) within an acceptable glycemic range (3.6–13.9 mM); or (c) hyperglycemic (>13.9 mM). Results. Overall, sports performance skill was ∼20% lower when glucose concentrations were hypoglycemic compared to either acceptable or hyperglycemic at the time of skill testing (P < .05). During Stroop testing, “reading” and “color recognition” also degraded during hypoglycemia, while “interference” scores improved (P < .05). Nocturnal hypoglycemia was present in 66% of subjects, lasting an average of 84 minutes, but this did not affect sports skill performance the following day. Conclusions. Mild hypoglycemia markedly reduces sports skill performance and cognition in young athletes with T1DM

    Do Biogeographical Patterns in Morphological Traits of Insect Host and Parasitoid Wasp Communities Contribute to Ecological Release of a Range-Expanding Host via Trait Mismatching?

    Climate change is causing species to expand their ranges poleward into new locations. As species move, they will lose interactions with species that fail to follow and gain new interactions with species with which they are not coevolved or coadapted. As a result, one common outcome for range expansions is that species outbreak in their expanded ranges in response to altered antagonistic interactions (“ecological release”). Insect hosts have a suite of morphological adaptations to evade parasitoid enemies, and enemies have adaptations to effectively attack hosts. Here, we study a recent range expansion of an insect host. Neuroterus saltatorius (Hymenoptera: Cynipidae) is a gall-forming wasp specializing on oaks that has undergone a recent range expansion from Washington state to Vancouver Island, British Columbia. In its expanded range, it experiences ecological release, causing foliar damage and impacting oak ecosystems. It interacts with an array of insect host competitors on its host plant Quercus garryana (other cynipids wasps) that are attacked by a suite of enemies (parasitoid wasps). The aim of this study is to measure morphological traits of cynipid hosts and parasitoids to examine if community traits of interacting hosts and enemies vary along a latitudinal gradient, and in the native and expanded range of N. saltatorius. Specifically, we ask if trait diversity of insect host-parasitoid communities provides biological resistance or facilitates ecological release of N. saltatorius in the native and expanded ranges. We predict that if trait mismatching contributes to ecological release, the degree of trait matching between the range-expanding host and other cynipid hosts and parasitoids will be lower in the expanded range, where it interacts with non-coadapted community members.     Does a Web-first Tailored Design Method Work with Rural Populations?

    Considering rural populations are historically difficult to reach, an important component of this research revolves around improving survey techniques in rural areas. To address this issue, a web-first Tailored Design Method (TDM), utilizing a mixed-mode of internet and postal mail surveys, was adapted to research the quality of life experienced by rural families. Aided by the Iowa State University Center for Survey Statistics and Methodology – Survey Research Services, data were collected from 62 rural counties in Arkansas. Socioeconomic-demographic factors were examined regarding survey response mode (i.e. mail vs. web) with some differences found. Logistic regression results demonstrated males were less likely to use web only responses compared to females. Similarly, higher education and income levels were associated with an increased likelihood of utilizing web response methods opposed to the mail-only response mode. Overall, the web-first TDM approach seems effective for garnering responses from harder to reach populations and should be considered when surveying rural populations

    From Cell Death to Regeneration: Rebuilding After Injury

    The ability to regrow lost or damaged tissues is widespread, but highly variable among animals. Understanding this variation remains a challenge in regeneration biology. Numerous studies from Hydra to mouse have shown that apoptosis acts as a potent and necessary mechanism in regeneration. Much is known about the involvement of apoptosis during normal development in regulating the number and type of cells in the body. In the context of regeneration, apoptosis also regulates cell number and proliferation in tissue remodeling. Apoptosis acts both early in the process to stimulate regeneration and later to regulate regenerative patterning. Multiple studies indicate that apoptosis acts as a signal to stimulate proliferation within the regenerative tissues, producing the cells needed for full regeneration. The conservation of apoptosis as a regenerative mechanism demonstrated across species highlights its importance and motivates the continued investigation of this important facet of programmed cell death. This review summarizes what is known about the roles of apoptosis during regeneration, and compares regenerative apoptosis with the mechanisms and function of apoptosis in development. Defining the complexity of regenerative apoptosis will contribute to new knowledge and perspectives for understanding mechanisms of apoptosis induction and regulation

    Harnessing Modern Web Application Technology to Create Intuitive and Efficient Data Visualization and Sharing Tools

    Neuroscientists increasingly need to work with big data in order to derive meaningful results in their field. Collecting, organizing and analyzing this data can be a major hurdle on the road to scientific discovery. This hurdle can be lowered using the same technologies that are currently revolutionizing the way that cultural and social media sites represent and share information with their users. Web application technologies and standards such as RESTful webservices, HTML5 and high-performance in-browser JavaScript engines are being utilized to vastly improve the way that the world accesses and shares information. The neuroscience community can also benefit tremendously from these technologies. We present here a web application that allows users to explore and request the complex datasets that need to be shared among the neuroimaging community. The COINS (Collaborative Informatics and Neuroimaging Suite) Data Exchange uses web application technologies to facilitate data sharing in three phases: Exploration, Request/Communication, and Download. This paper will focus on the first phase, and how intuitive exploration of large and complex datasets is achieved using a framework that centers around asynchronous client-server communication (AJAX) and also exposes a powerful API that can be utilized by other applications to explore available data. First opened to the neuroscience community in August 2012, the Data Exchange has already provided researchers with over 2500 GB of data

    Diseñar una propuesta didáctica de Educación Artística como estrategia de enseñanza y aprendizaje transdisciplinar para fortalecer la comprensión en las ciencias sociales de los estudiantes del grado séptimo del Asociación Alianza Educativa Colegio Argelia, utilizando como herramienta artística la pintura, el dibujo artístico y el teatro pedagógico.La investigación APRENDIZART se desarrolla mediante un marco teórico que se fragmenta en tres secciones las cuales son: propuesta pedagógica, Educación Artística y Aprendizaje transdisciplinar, secciones que dieron lugar a la completa construcción de las estrategias didácticas requeridas al realizar la construcción de las clases, se decidió investigar desde la asignatura de las ciencias sociales debido a que los temas curriculares coincidían con el momento de la aplicación, dicha aplicación se realizó en el Colegio Argelia ubicado en Bosa la paz. En base al trabajo en la asignatura de las ciencias sociales se desarrollaron temas específicos de las regiones naturales de Colombia, en donde se realizó énfasis en la hidrografía, pisos térmicos, comidas y bailes típicos correspondientes a cada región, temas en los cuales la transdisciplinariedad de la educación artística fue fundamental en el aprendizaje significativo de los estudiantes. La propuesta de estrategias didácticas en la transdisciplinariedad de la Educación Artística tuvo un enfoque importante en el dibujo artístico y la pintura como factores que permite que los estudiantes obtengan un aprendizaje significativo dado que al expresar gráficamente los conocimientos obtenidos permitirán una mayor retención de información adquirida, a esto también se le atribuye el manejo de colores que según un estudio que realizó Johann Göethe (1749- 1832),científico Alemán, comprobó las modificaciones psicológicas en el ser humano ante el manejo de colores y la reacción positiva que se tenía frente a estos, por consiguiente el dibujo acompañado P á g i n a 14 | del uso de colores y pintura fortalece el conocimiento que el estudiante recibe. El dibujo y la pintura se utilizó como fundamento de acompañamiento en la aplicación de la hidrografía de Colombia, a partir de un rompecabezas didáctico con la forma de Colombia en material de foamy realizado por los investigadores, por lo tanto los estudiantes al utilizar como material principal la pintura dibujaron y pintaron en los rompecabezas los respectivos ríos que conforman cada región, la estrategia didáctica se evidencio en el aprendizaje de los estudiantes y como estos por medio de la pintura aprendieron a ubicar los ríos, a conocer los respectivos nombres, así mismo conocer por medio del dibujo la magnitud y el tamaño respectivo en cada región, por consiguiente se establece que las didácticas innovadoras en la educación deben atraer al estudiante a disfrutar del conocimiento y asimismo facilitar el propósito de la enseñanza
