    Can translocated native fishes retain their trophic niche when confronted with a resident invasive?

    Diet interactions between native and non-native fishes may influence the establishment of native species within their historical range (i.e., reintroduction). Therefore, we illustrated the food web structure of and followed the transition of the federally endangered humpback chub Gila cypha into a novel food web following translocation and determined the potential for a non-native species, rainbow trout Oncorhynchus mykiss, to influence translocation success. Humpback chub and rainbow trout used resources high in the food web and assimilated similar proportions of native fishes, suggesting non-native rainbow trout may occupy an ecological role similar to humpback chub. Subsequently, humpback chub may be well suited to colonise tributaries because of their ability to consume resources high in the food web. Additionally, diet partitioning may occur between all members of the fish community as indicated by separation in trophic niche space and little trophic overlap; although all species, particularly bluehead sucker Catostomus discobolus, used a broad range of food resources. Rainbow trout stomach content analysis corroborated stable isotope analysis and suggested rainbow trout diet consisted of aquatic and terrestrial macroinvertebrates, while larger rainbow trout (\u3e120 mm total length) consumed a greater proportion of fish (incidence of piscivory = 5.3%). Trophic interactions may reveal an underutilized niche space or biotic resistance to the establishment of translocated native fishes. Continued translocation of humpback chub into tributaries appears to be one option for conservation. However, successful establishment of humpback chub may depend on continued removal of non-native trout, increasing availability of diet sources at higher trophic levels

    Textural characterization, major and volatile element quantification and Ar-Ar systematics of spherulites in the Rocche Rosse obsidian flow, Lipari, Aeolian Islands:A temperature continuum growth model.

    Spherulitic textures in the Rocche Rosse obsidian flow (Lipari, Aeolian Islands, Italy) have been characterized through petrographic, crystal size distribution (CSD) and in situ major and volatile elemental analyses to assess the mode, temperature and timescales of spherulite formation. Bulk glass chemistry and spherulite chemistry analyzed along transects across the spherulite growth front/glass boundary reveal major-oxide and volatile (H2O, CO2, F, Cl and S) chemical variations and heterogeneities at a ≤5 μm scale. Numerous bulk volatile data in non-vesicular glass (spatially removed from spherulitic textures) reveal homogenous distributions of volatile concentrations: H2O (0.089 ± 0.012 wt%), F (950 ± 40 ppm) and Cl (4,100 ± 330 ppm), with CO2 and S consistently below detection limits suggesting either complete degassing of these volatiles or an originally volatile-poor melt. Volatile concentrations across the spherulite boundary and within the spherulitic textures are highly variable. These observations are consistent with diffusive expulsion of volatiles into melt, leaving a volatile-poor rim advancing ahead of anhydrous crystallite growth, which is envisaged to have had a pronounced effect on spherulite crystallization dynamics. Argon concentrations dissolved in the glass and spherulites differ by a factor of ~20, with Ar sequestered preferentially in the glass phase. Petrographic observation, CSD analysis, volatile and Ar data as well as diffusion modeling support continuous spherulite nucleation and growth starting at magmatic (emplacement) temperatures of ~790–825 °C and progressing through the glass transition temperature range (T g ~ 750–620 °C), being further modified in the solid state. We propose that nucleation and growth rate are isothermally constant, but vary between differing stages of spherulite growth with continued cooling from magmatic temperatures, such that there is an evolution from a high to a low rate of crystallization and low to high crystal nucleation. Based on the diffusion of H2O across these temperature ranges (~800–300 °C), timescales of spherulite crystallization occur on a timescale of ~4 days with further modification up to ~400 years (growth is prohibitively slow <400 °C and would become diffusion reliant). Selective deformation of spherulites supports a down-temperature continuum of spherulite formation in the Rocche Rosse obsidian; indeed, petrographic evidence suggests that high-strain zones may have catalyzed progressive nucleation and growth of further generations of spherulites during syn- and post-emplacement cooling

    Elevated MCP-1, TNF-α, Monocyte Concentration, and Dyslipidemia in Obese Mexican-American Children

    Background: Obesity is an independent risk factor for chronic disease. The prevalence of obesity is especially high among Mexican-American children. Peripheral blood monocytes contribute to systemic inflammation and may mediate the relationship between obesity and chronic disease. Obesity affects monocytes and the circulating levels of cytokines/chemokines that influence monocyte behavior. Purpose: investigate alterations in blood monocytes and plasma cytokines/chemokine levels among healthy weight (zBMI ≤ 85th percentile; N=66), overweight (zBMI=85th-95th percentile; N=23), and obese (zBMI ≥ 95th percentile; N=39) Mexican-American children. Methods: Blood samples were analyzed for total monocyte concentration, pro-inflammatory monocyte concentration, and classic monocyte concentration via flow cytometry. Serum MCP-1, Fractalkine, IL-8, and TNF-α were measured using a Milliplex MagPix assay. Serum cholesterol, HDL, triglycerides, and glucose were measured using an enzymatic reagent kit. Results: Total monocyte concentration (P=0.012), classic monocyte concentration (P=0.045), MCP-1 (P=0.015), and TNF- α (P=0.002) were significantly greater in obese children compared to heathly weight children. Also, overweight and obese children had elevated triglycerides (P=0.001) and reduced HDL (P=0.033) compared to healthy weight children. Conclusion: Elevations in circulating monocytes, MCP-1, and TNF-α have been implicated in the development of obesity-related chronic disease in adults. Childhood obesity alters monocytes and circulating chemokines, putting children at a greater risk of developing obesity-related chronic diseases in adulthood. Further characterization of early immune alterations in childhood obesity may provide additional clinical insight into the assessment of obesity-related disease risk

    Trps1 Regulates Development of Craniofacial Skeleton and Is Required for the Initiation of Palatal Shelves Fusion

    Trichorhinophalangeal syndrome (TRPS) is an autosomal dominant disorder resulting from heterozygous mutations of the TRPS1 gene. Common craniofacial abnormalities in TRPS patients include micrognathia, hypoplastic zygomatic arch, high-arched palate, and, occasionally, cleft palate. Studies have demonstrated that mice with a heterozygous Trps1 mutation (Trps1+/− mice) have similar features to patients with TRPS, including high-arched palates. However, mice with a homozygous Trps1 mutation (Trps1−/− mice) exhibit similar but more severe abnormalities, including cleft palate. Our study aimed to characterize the craniofacial phenotype to understand the role of Trps1 in craniofacial development and gain insight on the cleft palate pathogenesis in Trps1 deficiency. Whole-mount skeletal staining revealed hypoplastic skeletal and cartilaginous elements, steep nasal slope, and missing presphenoid in Trps1−/− mice. Although several craniofacial skeleton elements were abnormal in Trps1−/− mice, the Trps1 deficiency did not appear to disrupt cranial vault development. All Trps1−/− mice presented with cleft palate. Analyses of Trps1 expression during palatogenesis detected Trps1 mRNA and protein in palatal mesenchyme and in specific regions of palatal epithelium, which suggested that Trps1 is involved in palatal fusion. Ex vivo culture experiments demonstrated that Trps1−/− palatal shelves were unable to initiate the fusion process. On the molecular level, Trps1 deficiency resulted in decreased epithelial expression of proteins involved in palatal fusion, including chondroitin sulfate proteoglycan, transforming growth factor-beta 3, Twist1, and beta-catenin. Mesenchymal expression of chondroitin sulfate proteoglycan expression was unaffected, indicating a cell type-specific mechanism of Trps1 regulation on chondroitin sulfate proteoglycan. In conclusion, we demonstrated that Trps1 is involved in the development of craniofacial skeletal elements and in the initiation of the palatal shelves fusion. Furthermore, our studies uncovered that Trps1 is required for epithelial expression of several proteins involved in the palatal shelves fusion

    Childhood Overweight is Associated with Increased Monocyte Concentration and Altered Subset Distribution

    Childhood obesity rates have nearly tripled in the last 30 years. Obesity increases risk for chronic disease. While monocytes, cells of the innate immune system that are altered with obesity, are purported to play an integral role in the development of these chronic diseases, no research has focused on early phenotypic changes in monocytes of overweight children. Two monocytes subsets exist, classic and pro-inflammatory; alterations in number and distribution may be implicated in disease development in obesity. The purpose of this study was to examine the concentration and relative distribution of monocytes among “normal weight” (N=66) and “at risk for being overweight/overweight” (N=56) Mexican American children. Blood samples were collected and analyzed for total monocyte concentration and monocyte subset concentration via flow cytometry. Total monocyte concentration, as well as the concentration of both classic and pro-inflammatory monocyte subsets was significantly greater in the “at risk for overweight/overweight” children (P\u3e0.05). Understanding early alterations in monocyte populations will be the first step in the development of early diagnosis and treatment techniques

    Early (and Later) LHC Search Strategies for Broad Dimuon Resonances

    Resonance searches generally focus on narrow states that would produce a sharp peak rising over background. Early LHC running will, however, be sensitive primarily to broad resonances. In this paper we demonstrate that statistical methods should suffice to find broad resonances and distinguish them from both background and contact interactions over a large range of previously unexplored parameter space. We furthermore introduce an angular measure we call ellipticity, which measures how forward (or backward) the muon is in eta, and allows for discrimination between models with different parity violation early in the LHC running. We contrast this with existing angular observables and demonstrate that ellipticity is superior for discrimination based on parity violation, while others are better at spin determination.Comment: 31 pages, 19 figures.
