    Liver-Type Glutaminase GLS2 Is a Druggable Metabolic Node in Luminal-Subtype Breast Cancer

    Efforts to target glutamine metabolism for cancer therapy have focused on the glutaminase isozyme GLS. The importance of the other isozyme, GLS2, in cancer has remained unclear, and it has been described as a tumor suppressor in some contexts. Here, we report that GLS2 is upregulated and essential in luminal-subtype breast tumors, which account for >70% of breast cancer incidence. We show that GLS2 expression is elevated by GATA3 in luminal-subtype cells but suppressed by promoter methylation in basal-subtype cells. Although luminal breast cancers resist GLS-selective inhibitors, we find that they can be targeted with a dual-GLS/GLS2 inhibitor. These results establish a critical role for GLS2 in mammary tumorigenesis and advance our understanding of how to target glutamine metabolism in cancer

    Large-scale detector testing for the GAPS Si(Li) Tracker

    Lithium-drifted silicon [Si(Li)] has been used for decades as an ionizing radiation detector in nuclear, particle, and astrophysical experiments, though such detectors have frequently been limited to small sizes (few cm2^2) and cryogenic operating temperatures. The 10-cm-diameter Si(Li) detectors developed for the General Antiparticle Spectrometer (GAPS) balloon-borne dark matter experiment are novel particularly for their requirements of low cost, large sensitive area (~10 m2^2 for the full 1440-detector array), high temperatures (near -40\,^\circC), and energy resolution below 4 keV FWHM for 20--100-keV x-rays. Previous works have discussed the manufacturing, passivation, and small-scale testing of prototype GAPS Si(Li) detectors. Here we show for the first time the results from detailed characterization of over 1100 flight detectors, illustrating the consistent intrinsic low-noise performance of a large sample of GAPS detectors. This work demonstrates the feasibility of large-area and low-cost Si(Li) detector arrays for next-generation astrophysics and nuclear physics applications.Comment: Updated to version accepted in IEEE Trans Nucl Sci.     Engineered Models of Metastasis with Application to Study Cancer Biomechanics

    Three-dimensional complex biomechanical interactions occur from the initial steps of tumor formation to the later phases of cancer metastasis. Conventional monolayer cultures cannot recapitulate the complex microenvironment and chemical and mechanical cues that tumor cells experience during their metastatic journey, nor the complexity of their interactions with other, noncancerous cells. As alternative approaches, various engineered models have been developed to recapitulate specific features of each step of metastasis with tunable microenvironments to test a variety of mechanistic hypotheses. Here the main recent advances in the technologies that provide deeper insight into the process of cancer dissemination are discussed, with an emphasis on three-dimensional and mechanical factors as well as interactions between multiple cell types

    A Preliminary Study Of Classroom Motivators And De-Motivators From A Motivation-Hygiene Perspective

    This study seeks to begin answering two simple questions: What motivates our students? and its corollary, What prevents our students from being motivated? The motivation-hygiene theory (F. Herzberg, Work and the nature of man, World Publishing, Cleveland, OH, 1966), a well-tested theory from organizational psychology, holds that people\u27s motivation stems from two sources: the desire to grow psychologically and the desire to avoid pain or unpleasantness. Previous research shows psychological growth factors serve as motivators, while pain avoidance factors serve as hygiene factors, neutral when present, but de-motivating when absent. Using this theory as a lens, the current study examines student motivation and finds that similar patterns are present in the classroom.© 2009 National Communication Association

    The Power Of Provisional/Immediate Language Revisited: Adding Student Personality Traits To The Mix

    Previous research found that relatively minor changes in the wording of written assessments can influence students\u27 motivation and affect toward the teacher. Not considered previously, however, is the role that student personality traits might play. Are the observed effects consistent across various personality types, or are different personality types affected differently by the same teacher behaviors? This study takes a first step toward answering that question, replicating the previous written feedback style research while adding the dimension of student personality traits to the investigation. The results suggest that the treatment may not be effective for those with high extroversion, low conscientiousness, and low neuroticism. When those cases were excluded from the analysis, the variance accounted for increased substantially, supporting the notion that efficacy of the teacher behaviors is, in part, dependent on student personality traits. © 2013 Copyright Eastern Communication Association
