    Identifying Potential Problems and Risks in GQM+Strategies Models Using Metamodel and Design Principles

    Although GQM+Strategies®1 assures that business goals and strategies are aligned throughout an organization and at each organizational unit based on the rationales to achieve the overall business goals, whether the GQM+Strategies grid is created correctly cannot be determined because the current definition of GQM+Strategies allows multiple perspectives when aligning goals with strategies. Here we define modeling rules for GQM+Strategies with a metamodel specified with a UML class diagram. Additionally, we create design principles that consist of relationship constraints between GQM+Strategies elements, which configure GQM+Strategies grids. We demonstrate that the GQM+Strategies grids can be automatically determined with the help of design principles described in OCL. In fact, an experiment is implemented using these approaches in order to show that this method helps identify and improve potential problems and risks. The results confirm that our approaches help create a consistent GQM+Strategies grid

    The AAA-ATPase VPS4 Regulates Extracellular Secretion and Lysosomal Targeting of α-Synuclein

    Many neurodegenerative diseases share a common pathological feature: the deposition of amyloid-like fibrils composed of misfolded proteins. Emerging evidence suggests that these proteins may spread from cell-to-cell and encourage the propagation of neurodegeneration in a prion-like manner. Here, we demonstrated that α-synuclein (αSYN), a principal culprit for Lewy pathology in Parkinson's disease (PD), was present in endosomal compartments and detectably secreted into the extracellular milieu. Unlike prion protein, extracellular αSYN was mainly recovered in the supernatant fraction rather than in exosome-containing pellets from the neuronal culture medium and cerebrospinal fluid. Surprisingly, impaired biogenesis of multivesicular body (MVB), an organelle from which exosomes are derived, by dominant-negative mutant vacuolar protein sorting 4 (VPS4) not only interfered with lysosomal targeting of αSYN but facilitated αSYN secretion. The hypersecretion of αSYN in VPS4-defective cells was efficiently restored by the functional disruption of recycling endosome regulator Rab11a. Furthermore, both brainstem and cortical Lewy bodies in PD were found to be immunoreactive for VPS4. Thus, VPS4, a master regulator of MVB sorting, may serve as a determinant of lysosomal targeting or extracellular secretion of αSYN and thereby contribute to the intercellular propagation of Lewy pathology in PD

    Fabrication of superoleophobic hierarchical surfaces for low-surface-tension liquids

    This study demonstrates the fabrication of hierarchical surfaces with super-repellency even for low-surface-tension liquids, including octane (surface tension of 21.7 mN m(-1)). Dual-pore surfaces were prepared by a combination of practical wet processes on an aluminium substrate: chemical etching, anodizing, and organic monolayer coating. The size of the larger pores formed by the chemical etching of aluminium is controlled by the concentration of HCl in the CuCl2/HCl etching solution. The etched aluminium is then anodized to introduce nanopores, followed by a pore-widening treatment that controls the nanopore size and porosity. The repellency for low-surface-tension liquids is enhanced by increasing the size of the larger pores as well as the porosity of the walls of the larger pores in this dual-pore morphology. Under optimized morphology with a fluoroalkyl-phosphate monolayer coating, an advancing contact angle close to 160 degrees, a contact angle hysteresis of less than 5 degrees and a sliding angle of 10 degrees is achieved even for octane

    Facile preparation of self-healing superhydrophobic CeO2 surface by electrochemical processes

    Herein we report simple electrochemical processes to fabricate a self-healing superhydrophobic CeO2 coating on Type 304 stainless steel. The CeO2 surface anodically deposited on flat stainless steel surface is hydrophilic, although high temperature-sintered and sputter-deposited CeO2 surface was reported to be hydrophobic. The anodically deposited hydrophilic CeO2 surface is transformed to hydrophobic during air exposure. Specific accumulation of contaminant hydrocarbon on the CeO2 surface is responsible for the transformation to hydrophobic state. The deposition of CeO2 on hierarchically rough stainless steel surface produces superhydrophobic CeO2 surface, which also shows self-healing ability; the surface changes to superhydrophilic after oxygen plasma treatment but superhydrophobic state is recovered repeatedly by air exposure.     Hybrid origin of gynogenetic clones and the introgression of their mitochondrial genome into sexual diploids through meiotic hybridogenesis in the loach,Misgurnus anguillicuadatus

    In a few Japanese populations of the loach Misgurnus anguillicaudatus (Teleostei: Cobitidae), clonal diploid lineages produce unreduced diploid eggs that normally undergo gynogenetic reproduction; however the origin of these clones remains elusive. Here, we show the presence of two diverse clades, A and B, within this loach species from sequence analyses of two nuclear genes RAG1 (recombination activating gene 1) and IRBP2 (interphotoreceptor retinoid-binding protein, 2) and then demonstrate heterozygous genotypes fixed at the two loci as the evidence of the hybrid nature of clonal lineages. All the clonal individuals were identified by clone-specific mitochondrial DNA haplotypes, microsatellite genotypes, and random amplified polymorphic DNA fingerprints; they commonly showed two alleles, one from clade A and another from clade B, whereas other wild-type diploids possessed alleles from either clade A or B. However, we also found wild-type diploids with clone-specific mitochondrial DNA and nuclear genes from clade B. One possible explanation is an introgression of a clone-specific mitochondrial genome from clonal to these wild-type loaches. These individuals likely arose by a cross between haploid sperm from bisexual B clade males and haploid eggs with clone-specific mtDNA and clade B nuclear genome, produced by meiotic hybridogenesis (elimination of unmatched A genome followed by meiosis after preferential pairing between two matched B genomes) in clone-origin triploid individual (ABB)
