    Treatment assessment of road runoff water in zones filled with ZVI, activated carbon and mineral materials

    Reducing the discharge of contaminants present in runoff water is important for a clean environment. This paper analyses field test results of three pilot-scale horizontal runoff water treatment zones filled with mixtures of zero valent iron (ZVI), activated carbon (AC), silica spongolite (SS), zeolite (Z), and limestone (LS). The investigated systems were (S1) ZVI/AC/SS, (S2) ZVI/AC/Z and (S3) ZVI/AC/LS. The efficiency of the three systems in the removal of Cd, Cu, Ni, Pb, Zn, COD and ammonium ions from runoff water was compared and the factors (temperature, pH, redox potential, hydraulic conductivity) and relationships affecting treatment effectiveness were determined. A statistical analysis of effluent contaminant concentrations and physicochemical parameters of effluent solutions included descriptive statistics, analysis of variance (ANOVA), a multidimensional analysis using a Principal Component Analysis (PCA), a factor analysis (FA) and a cluster analysis (CA). The ANOVA and cluster analyses indicated similarities between systems containing SS and LS. As a consequence, using cheaper SS can reduce investment costs. In addition, there were no significant differences between the three systems regarding Cd and Ni removal, while Cu and Pb were removed to almost 100%. The results indicate that all the tested materials supported ZVI and AC in the removal of heavy metals in a similar way. However, runoff water was enriched with nitrogen oxides and sulfates while flowing through treatment zones with SS and LS. The enrichment increased with increasing temperature and redox potential. The conducted analyses indicate that the most suitable mixture is ZVI/AC/Z. It should be emphasized that the ongoing processes (precipitation and ZVI corrosion) reduced the hydraulic conductivity of the filters up to two orders of magnitude. Expansive iron corrosion was the most limiting factor in ZVI filtration systems.     Salinomycin – a breakthrough in the treatment of ovarian cancer?

    It is believed that cancer stem cells are the primary cause of cancer chemotherapy resistance, metastasis and relapse. The cancer stem cells form a small population of cells present in the tumor (accounting for less than 2% of the tumor mass) and have properties which enable them to survive chemo- and radiotherapy. These cells have the ability to self-renew, do not undergo apoptosis, display overexpression of the ALDH1A1 enzyme and ABC genes which encode transport proteins, and furthermore make use of various signaling pathways (Wnt, Notch, Hedgehog). Cancer stem cells may be identified and isolated from the tumor based on the characteristic biomarkers (CD44+, CD133+, CD117+, BMi1, Oct-4, nestin). It has been demonstrated that salinomycin, an antibiotic obtained from Streptomyces albus, eliminates cancer stem cells, which are resistant to treatment with cytostatics. Salinomycin causes apoptosis of these cells through a number of mechanisms, including the disruption of the Na+/K+ ion balance in biological membranes, inhibition of the Wnt pathway and resistance to transporters, increase in the activity of caspases, activation of the MAPKp38 pathway and inhibition of the nuclear transcription factor NF-κB. Salinomycin has an effect on many types of cancer. It may turn out to be a breakthrough in the therapy of chemotherapy-resistant cancers

    The role of nesfatin and selected molecular factors in various types of endometrial cancer

    Objectives: Endometrial cancers (ECs) are the most common gynaecological cancers in well developed countries. Diabetes and metabolic syndrome are among the biggest risk factors. Nesfatin-1, the adipokine derivative of NUCB2 (nucleobindin derivative 2) is linked to the clinical course of EC. Molecular factors, including mutations in MLH1 and MHS2 genes, c-MET and ARID1A are also related to prognosis in endometrial cancer. Material and methods: Using sections of paraffin-embedded preparations and immunohistochemistry, the expression of NESF1, MLH1, MSH2,c-MET and ARID1A were examined. Results: In this study on protein expression, EC tissues manifested (although insignificantly) an elevated expression of NESF-1 in type II EC. In type I EC, NESF-1 expression was significantly higher in G1 in comparison to G2 and G3 together. A significantly lower expression of MLH1 was demonstrated in type I EC. Conclusions: The most pronounced expression involved c-MET in all EC I and EC II tissues (in over 80% of cases). A tendency was detected for a high expression of NESF-1 in patients with type II EC, who also exhibited a high expression of MSH2

    The Effect of Silver Nanoparticles on Listeria monocytogenes PCM2191 Peptidoglycan Metabolism and Cell Permeability

    Listeria monocytogenes is Gram-positive bacterial pathogen, a causative agent of food poisoning and systemic disease – listeriosis. This species is still susceptible to several conventionally used antibiotics but an increase in its resistance has been reported. For this reason the search for new, alternative therapies is an urgent task. Silver nanoparticles seem to be the promising antibacterial agent. Minimal inhibitory concentration of silver nanoparticles was determined. Sublethal concentrations were used in study of nanosilver effect on cells lysis by estimation of the number of cells surviving the treatment with 0.25 or 0.5 of minimal inhibitory concentrations of silver nanoparticles. Autolysis of isolated peptidoglycan was studied by measuring the absorbance of preparation subjected to nanosilver treatment. Silver nanoparticles effect on L. monocytogenes envelopes permeability was determined by measuring the efflux of cF, DNA and proteins. It was demonstrated that nanosilver enhanced the lysis of L. monocytogenes cells and, to the lesser extent, autolysis of isolated peptidoglycan. The increase in the efflux of carboxyfluoresceine, DNA and proteins was also noted. The obtained results allow to postulate that L. monocytogenes peptidoglycan, constituting the main component of cell wall, is the target of silver nanoparticles activity against this pathogen

    Serological Survey of \u3ci\u3eLeptospira\u3c/i\u3e Infection in Arabian Horses in Poland

    Leptospirosis is one of the most common zoonotic infections worldwide, including in most livestock, some companion animals, horses, wildlife, and humans. Epidemiological estimation of its prevalence in all species is difficult due to the variety of clinical presentations and challenges regarding laboratory diagnosis. The purpose of this study was to measure the seroprevalence of leptospiral infection in Arabian horses kept in the largest breeding farms in Poland, representing over 15% of the Polish Arabian horse population. Leptospira antibodies were detected by MAT (cut-off 1:100) in 33.2% of serum samples (204 of 615 animals) (CI 95%: 29.6–37.0%), most frequently reacting with the serovar Grippotyphosa, similar to previous reports in populations of randomly selected horses. These results indicated high Leptospira seropositivity, thus, although any form of clinical leptospirosis is rare, it may be postulated that the leptospiral exposure is widespread

    Can selected physiotherapeutic techniques really help in treating back pain and improving the quality of life of advanced cancer patients: a randomised controlled study

    Background: The available literature lacks studies using the KinesioTaping (KT) method and the hold-relax (HR) technique in working with advanced cancer patients. Patients and methods: The study involved 72 patients (38 women and 34 men), diagnosed with advanced cancer. Patients were randomly assigned either to KT group (exercise programme and KT), HR group (exercise programme and the HR technique of the proprioceptive neuromuscular facilitation — PNF method) or C group (a control group, exercise programme). To assess selected parameters the Numerical Rating Scale (NRS) scale and Edmonton Symptom Assessment System — revised (ESAS-r) were used. The physiotherapeutic programme lasted three weeks and took place 5 days a week for 30 minutes within each group. Additionally, in the KT group, kinesio tapes were applied on the paraspinal muscles of the lumbar spine. In the HR group the therapy with the Hold–Relax technique was applied. Results: There was a statistically significant decrease in pain in all groups and improvement in the quality of life in patients from both experimental groups. Those changes were significantly greater in the KT group than in both the C group and the HR group. Conclusions: Both KT and HR techniques of the PNF method are effective in reducing pain and improving the quality of life in the examined advanced cancer patients, however KT has a stronger impact

    IL-33 Gene Polymorphisms as Potential Biomarkers of Disease Susceptibility and Response to TNF Inhibitors in Rheumatoid Arthritis, Ankylosing Spondylitis, and Psoriatic Arthritis Patients

    ObjectiveRheumatoid arthritis (RA), ankylosing spondylitis (AS), and psoriatic arthritis (PsA) belong to inflammatory rheumatic diseases, the group of conditions of unknown etiology. However, a strong genetic component in their pathogenesis has been well established. A dysregulation of cytokine networks plays an important role in the development of inflammatory arthritis. Interleukin 33 (IL-33) is a recently identified member of the IL-1 family. To date, the significance of IL-33 in inflammatory arthritis has been poorly studied. This research aimed to investigate the potential of IL-33 gene polymorphisms to serve as biomarkers for disease susceptibility and TNF inhibitor response in RA, AS, and PsA patients.Materials and MethodsIn total, 735 patients diagnosed with RA, AS, and PsA and 229 healthy individuals were enrolled in the study. Genotyping for three single nucleotide polymorphisms (SNPs) within the IL-33 gene, namely, rs16924159 (A/G), rs10975519 (T/C), and rs7044343 (C/T), was performed using polymerase chain reaction amplification employing LightSNiP assays.ResultsIn the present study, the IL-33 rs10975519 CC genotype was associated with a decreased risk of developing RA in females, while the IL-33 rs16924159 polymorphism was associated with the efficacy of anti-TNF therapy and clinical parameters for RA and AS patients. The IL-33 rs16924159 AA genotype correlated with higher disease activity and worse clinical outcomes in RA patients treated with TNF inhibitors, and AS patients carrying the IL-33 rs16924159 AA genotype had higher disease activity and a worse response to anti-TNF therapy. That indicates a deleterious role of the IL-33 rs16924159 AA genotype in the context of RA, as well as AS.ConclusionsThe obtained results suggest that IL-33 gene polymorphisms might be potential candidate biomarkers of disease susceptibility and anti-TNF treatment response in patients with inflammatory rheumatic diseases
