Role of Rho GTPases During Primordial Germ Cell Migration in Zebrafish

Cell migration is an essential biological process during development, adult homeostasis, and disease. Actin cytoskeleton plays crucial role in generating forces required for cell migration. Rho GTPases are key regulators of actin cytoskeleton remodeling during cell migration. To understand the function of Rho GTPases it is important to have the information regarding their spatial-temporal activation pattern in migrating cell. Accessibility of cells migrating in vivo has proven to be a major challenge in studying molecular mechanisms underlying cell migration in their natural milieu. Primordial germ cells (PGCs) in zebrafish arise at distinct positions of the developing embryo and migrate towards their target guided by the chemokine SDF-1a. PGC migration in zebrafish is an excellent in vivo model system to study cell migration processes in vertebrates. It was shown that PGC migrate by forming blebs at their leading edge. The present work focuses at elucidating the role of Rho GTPases in actin cytoskeleton remodeling during PGC migration in zebrafish. To this end I studied actin cytoskeleton dynamics in the presence of mutant forms for three important Rho GTPases: Rac, Cdc42 and RhoA. To study the spatial-temporal activation pattern for Rho GTPases in migrating PGCs, I optimized ratio imaging with FRET biosensors in zebrafish embryo. Utilizing FRET biosensors I showed that Rac and RhoA activities are elevated at the front of the migrating PGC, while Cdc42 is active in the nucleus. Imaging actin cytoskeleton in PGCs revealed that depending on Rac activity, actin accumulates at the front of the cell forming a dense network, which then flows backwards in the process that requires RhoA activity. I propose that in migrating PGCs, Rac function promotes actin accumulation at the front of the cell, while RhoA mediates the actin backward flow thereby generating forces essential for cell locomotion

Онтологічне дослідження сутності продовольчої безпеки (ONTOLOGICAL RESEARCH OF FOOD security ESSENCE)

У статті проведено дослідження еволюції сутності продовольчої безпеки, здійснено історичну періодизацію та виділено основні її формотворчі етапи. З’ясовано особливості продовольчої безпеки на кожному етапі, а також виокремлено шляхи її досягнення. Проаналізовано світові концепції забезпечення продовольчої безпеки. Узагальнено трактування продовольчої безпеки у міжнародному та вітчизняному законодавстві та наукових працях. Обґрунтовано соціально-економічну сутність продовольчої безпеки. (Research of the food security’s essence evolution is carried out, historical periodization is made and main formative stages are identified. The features of food security are found out on every stage and also the ways of her achievement are distinguished. World conceptions of food security providing are analysed. Interpretations of food security in the international and domestic legislation and scientific works are generalized. Socio-economic essence of food security is proved.

Наскільки він чисельний та впливовий? Яку методологію застосовувати для його дослідження та ідентифікації? (This study aims to try to answer some of the common and discussed in the Ukrainian political science problems. Does Ukraine have a middle class? How he numerical and influential? What is the methodology used for its study and identification?

Відображення професійної стратифікації зайнятого населення в соціальній структурі українського суспільства після Революції Гідності (Reflection of the employed population’ professional stratification in the social structure of Ukrainian society after the Revolution of Dignity)

Соціальна структура завжди займала важливе місце у дослідженнях суспільствознавців. Не тільки для соціологів, але й для політологів, актуальними для дослідження виглядають питання: які соціальні групи і класи формують соціальну структуру, як вони між собою взаємодіють як первинні суб’єкти політики, як держава намагається вливати на соціальну структуру суспільства. В даному дослідженні ми не беремося дати відповіді на всі ці питання. Основна наша увага буде зосереджена на тому, куди рухається соціальна структура українського суспільства після Революції Гідності, анексії Криму та війни на Сході України. У цій роботі буде представлено один із можливих дослідницьких підходів до вивчення згаданих мною питань. Ми спробуємо подивитися на соціальну структуру крізь призму професійної структури зайнятого населення в офіційному секторі. (Sociologists have always paid special attention to the social structure in their studies. What social groups and classes form the social structure, how do they interact with each other as the primary subjects of politics, how the state tries to influence on the social structure of society. All these questions are relevant to study not only for sociologists but also for political scientists. In this research we are not trying to give an answer to all these questions. Our major attention will be paid to what changes took place with the social structure of Ukrainian society after the Revolution of Dignity, annexation of Crimea and war on the East of Ukraine. One of the possible research approaches to the study of the mentioned above questions will be represented in this paper. We will try to take a look at the social structure through the prism of the employed population’ professional structure in an official sector.

Політична модернізація та соціальна структура суспільства в постреволюційній Україні: у пошуках залежності (Political modernization and social structure of society in post-revolutional Ukraine: in respect of dependence)

В статті звертається увага на взаємозалежність двох складних феноменів: модернізації та соціально-професійної структури населення. Використовуючи модель модернізації Р. Інглхарта та К. Вельцеля автор обґрунтовує вплив соціально-економічних факторів (особливості соціальної структури суспільства) на процеси демократизації в Україні. Соціальна структура в цій роботі досліджується крізь призму професійної структури суспільства, під якою розуміється загальний розподіл професій (кваліфікацій). Обґрунтовуючи необхідність подібних досліджень, автор шукає відповідь на деякі важливі питання: чи можлива в Україні успішна модернізація, виходячи із наявних якісних та кількісних характеристик професійної структури? Які політичні та соціально-економічні наслідки може мати трансформація соціальних груп, що формують професійну структуру? (This article draws attention to the interdependence of two complex phenomena: modernization and socio-occupational structure of the population. Using modernization theory by R. Inglehart and K. Welzel, the author substantiates the influence of socio-economic factors (peculiarities of the social structure of society) on the processes of democratization in Ukraine. In this research the social structure is investigated through the prism of the occupational structure of society, which refers to the general occupational (qualificational) differentiation. In substantiating the need for such studies, the author seeks to answer some of the important questions: is successful modernization possible in Ukraine, based on the existing qualitative and quantitative characteristics of the occupational structure? What kind of political and socio-economic consequences can the transformation of social groups that form an occupational structure have?

Формування «нового» середнього класу в Україні: проблеми та перспективи (Forming of "new" middle class in Ukraine: problems and prospects)

Це пошукове дослідження спрямоване на з’ясування деяких важливих та проблемних питань становлення та формування «нового» середнього класу в українському суспільстві. Зокрема, нас буде цікавити питання важливості формування середнього класу та пошук тих, хто повинен стати його майбутньою основою. (This search study is aimed at finding out some important and problematic questions of development and forming of "new" middle class in Ukrainian society. In particular, we will be interested in the question of importance of middle class forming, and finding those, who shall become its future basis.

Специфіка монологічного мовлення: психологічний та лінгвістичний аспекти

У статті досліджено монолог як форму мови, утворену в результаті активної мовленнєвої діяльності людини, розраховану на пасивне, опосередковане сприйняття і практично не пов’язану з відповідною промовою співрозмовника ні в змістовному, ні в структурному відношенні; в окремих випадках монолог розглянуто як інтраперсональний мовленнєвий акт, який не пристосований до безпосереднього спілкування, він припускає, що хтось тільки слухає, але не відповідає. У пропонованій науковій розвідці автор характеризує комунікативні, психологічні й лінгвістичні особливості монологу, висвітлює основні комунікативні ситуації його вживання (сфера театрального мистецтва, ораторські виступи, взаємодія з аудиторією на телебаченні і радіо, педагогічна діяльність). Результати дослідження засвідчили, що комунікативні особливості монологу розкривають його як безперервний зв’язний виклад думок мовця до однієї чи кількох осіб із певною метою; у побутовому спілкуванні монологічна мова трапляється вкрай рідко; монолог часто вживають як літературний прийом, що слугує для передачі думок і почуттів персонажа. (An important part in the people’s communication plays the monologue speech, which is represented in oral and written combinations. It is more difficult form of communication in comparison with the dialogue as monologue texts demand previous thinking and planning. Being rather extensive, the monologue speech with its content and grammar form is in many aspects close to the written form. The article researches the monologue as a form of language, formed as a result of active communicative human activity, expecting a passive, satisfactory perception and practically not connected with corresponding speech of the communicative partner neither in plot nor in structural connection; the monologue is seen as an interpersonal speech act in some cases, that is not appropriate to the natural communication and it foresees, that someone not only hers but also answers. The author gives a characteristic of communicative, psychological and linguistic peculiarities of the monologue, highlights the main communicative situations and their usage (the sphere of theater art speakers reports, cooperation with the audience on TV and radio, pedagogical activity) in the proposed scientific research. The results of the research witnesses that the communicative peculiarities of the monologue represent it as a non-stop sequence of speaker’s thoughts to the one or other persons with peculiar purpose, in everyday the monologue speech occurs rather rarely, the monologue is used as a literary mean which helps in transmission of the personage’s thoughts and feelings.
